Female wistar rats were inoculated intratracheally with 10 mg/ml suspensions of various dusts, viz: quartz, fly ash, mica and corundum in physiological saline. Biochemical markers of bronchoalveolar lavage fluid (BALF) were analysed 8 days after the instillation of the dusts. Elevated levels of proteins, sialic acid and phospholipid contents and the activity of lactate dehydrogenase correlated well with the degree of the known fibrogenic potential of different dusts in the lungs in the following order, quartz greater than fly ash greater than mica greater than corundum. beta-glucuronidase activity, was however, only elevated in the quartz inoculated group of rats. It is suggested that biochemical constituents of BALF analysed shortly after the exposure to different dusts can be useful to mirror alterations in the tissue response to mineral dusts.

Dry sedimented volcanic ash samples from each of three widely separated volcanoes of the "Circum Pacific" region have been subjected to mineralogic analysis and in vitro tests for cytotoxicity. The ash samples from the three different volcanoes varied in particle size, surface area, and concentration of silica. Total crystalline silica in the respirable fraction of ashes was 1.5% (Mount St. Helens, Moses Lake); 1.36% (Galunggung, Bandung-1); 1.95% (Gallunggung, Bandung-2); and 1.72% (El Chichon, Tuxtla). Hemolysis as an index of cytotoxicity was measured by in vitro tests on sheep blood erythrocytes and indicated wide differences in hemolytic activity among ash samples. Alveolar macrophage cytosolic (lactate dehydrogenase) and lysosomal (beta-glucuronidase and beta-N-acetyl glucosaminidase) enzymes were measured as an index of cellular integrity following dust exposure. Hemolysis and release of enzymes from alveolar macrophages were greater with volcanic ash from Galunggung (Bandung-1) and El Chichon (Tuxtla) than the other ashes. Although crystalline silica induced an effect similar to volcanic ash from Galunggung (Bandung-1) on the release of enzymes from alveolar macrophages, the hemolytic potency of silica was much greater. Light and electron microscopic observations of dust-exposed alveolar macrophages indicated that the ash particles were readily phagocytized. These results indicate that volcanic ash is moderately cytotoxic and that exposure may lead to overt reactions and the exacerbation of preexisting chronic inflammatory processes.

In a previous study we found increased SCE frequencies in peripheral blood lymphocytes (PBLs) of workers occupationally exposed in a coal fly ash processing industry, as compared to a non-exposed control population. Shortly after this study, measures were taken in this plant to reduce fly ash levels. The objective of the present study, conducted 2 years later in the same plants, was to evaluate the effect of these measures with respect to genotoxic risk. A group of 18 male workers of the coal fly ash processing industry agreed to participate in the study. The control population consisted of 18 male workers from a flour processing industry, who were matched for age and smoking behavior. In contrast to our previous study, no increased SCE frequencies were found in PBLs of workers potentially exposed to coal fly ash when compared to the control group (mean SCEs: 6.4 +/- 1.2 and 7.0 +/- 0.9, respectively). In addition, no differences were observed between the exposed and control groups for frequencies of gene mutations at the hypoxanthine guanine phosphoribosyltransferase (hprt) locus in PBLs, for micronucleus frequencies using the cytokinesis block method, or for urinary mutagen excretion measured with Salmonella typhimurium tester strains TA98 and TA97 with and without metabolic activation. In smokers, however, SCE frequencies in PBLs were significantly increased in comparison to non-smokers (7.1 +/- 1.1 vs. 6.1 +/- 0.5; P < 0.005), as was 24-h urinary mutagen excretion measured with strain TA98 with S9 mix (2373 +/- 1870 vs. 156 +/- 211; P < 0.001) and with TA98 with S9 mix and beta-glucuronidase/arylsulfatase (2361 +/- 1958 vs. 538 +/- 396; P < 0.005). In addition, hprt variant frequencies in PBLs were higher in smokers than in non-smokers (15.0 +/- 23.5 x 10(-6)6 vs. 2.6 +/- 2.8 x 10(-6); P < 0.05). No differences were observed for micronucleus induction between smokers and non-smokers. It is concluded that the protective measures taken in the coal fly ash processing plant appear to have been sufficient, since an effect of exposure to coal fly ash on parameters of genetic risk was not found any longer.

A genetic transformation protocol for green ash (Fraxinus pennsylvanica) hypocotyl explants was developed. Green ash hypocotyls were transformed using Agrobacterium tumefaciens strain EHA105 harboring binary vector pq35GR containing the neomycin phosphotransferase (nptII) and beta-glucuronidase (GUS) fusion gene, and an enhanced green fluorescent protein gene. Pre-cultured hypocotyl explants were transformed in the presence of 100 microM acetosyringone using 90 s sonication plus 10 min vacuum-infiltration. Kanamycin at 20 mg l(-1) was used for selecting transformed cells. Adventitious shoots regenerated on Murashige and Skoog medium supplemented with 13.3 microM 6-benzylaminopurine, 4.5 microM thidiazuron, 50 mg l(-1) adenine sulfate, and 10% coconut water. GUS- and polymerase chain reaction (PCR)-positive shoots from the cut ends of hypocotyls were produced via an intermediate callus stage. Presence of the GUS and nptII genes in GUS-positive shoots were confirmed by PCR and copy number of the nptII gene in PCR-positive shoots was determined by Southern blotting. Three transgenic plantlets were acclimatized to the greenhouse. This transformation and regeneration system using hypocotyls provides a foundation for Agrobacterium-mediated transformation of green ash. Studies are underway using a construct containing the Cry8Da protein of Bacillus thuringiensis for genetic transformation of green ash.

Fly-ash particles of a diameter smaller than 2 mu were collected from the electrostatic precipitator of a coal-fired power plant, and their cytotoxicity was studied in culture using alveolar macrophages from healthy and hypothyroid rats. The mortality rate and the secretion rates of cytoplasmic (lactate dehydrogenase) and lysosomal (acid phosphatase and beta-glucuronidase) enzymes into the culture medium were greater in the alveolar macrophages from hypothyroid rats than in the control macrophages. Electron microscopy showed a retraction of filapodia, loss of ruffled membrane and distortion of cell shape in the alveolar macrophages from hypothyroid rats. These results demonstrate that the cytotoxicity of fly-ash particles was enhanced in the macrophages from hypothyroid rats, and indicate that the normal functions of alveolar macrophages are impaired by changes in thyroid status.
