The activity of gold sodium thiomalate (GST) given i.m. to adjuvant-induced polyarthritic rats was studied alone or in combination with active doses of aspirin, indomethacin and hydrocortisone. In addition to paw volume and body weight changes, erythrocyte sedimentation rate, serum albumin/globulin and gold levels as well as plasma activities of
beta-glucuronidase
, acid phosphatase, lysozyme and
lactic acid dehydrogenase
were measured. In prophylactic studies the beneficial activity of GST was unaffected by aspirin, suggesting a positive drug interaction, but additive with indomethacin or hydrocortisone for the 1st but not 2nd lesion of the disease. These results were closely correlated with increased serum gold levels. Similar clinical findings were observed in therapeutic studies except that a positive drug interaction occurred between GST and hydrocortisone. Unlike in the prophylactic experiments, serum gold levels were unaffected by any of the agents tested in the therapeutic studies.

Hematological and serum biochemical changes in response to hemorrhagic stress were determined in both sexes of juvenile and adult Coturnix coturnix japonica over 3 day period following a mechanical hemorrhage of 30% of the calculated total blood volume. There was an initial shift posthemorrhage towards greater numbers of more mature erythrocytes and fewer circulating reticulocytes. Reticulocytosis was indicated 48-72 hr posthemorrhage. Glucose and
lactic acid dehydrogenase
levels increased after hemorrhage. Serum
beta-glucuronidase
was elevated only in adults. Japanese quail seemed to recover from hemorrhage more rapidly than had been reported for chickens, other birds and mammals.

Beta-glucuronidase-inhibitory and hepatoprotective effects of Reduohanxiao-tang (Yuldahanso-tang), which has been used for liver diseases and stroke, on carbon tetrachloride (CCl4)-induced hepatotoxicity of rats were investigated. Reduohanxiao-tang potently inhibited beta-glucuronidases. Serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and
lactic acid dehydrogenase
(
LDH
) levels of the CCl4 group orally treated with Reduohanxiao-tang (100 mg/kg) were lowered to 54%, 71.5% and 66.1% of the CCl4-treated control group, respectively. Among the ingredients of the Reduohanxiao-tang, the rhizomes of Pueraria thunbergiana and Scutellaria baicalensis potently inhibited beta-glucuronidases and protected against CCl4-induced liver injury. Orally administered puerarin, which is a main component of Pueraria thunbergiana, showed potent hepatoprotective activity, but did not inhibit
beta-glucuronidase
. However, daidzein, which is produced from puerarin by human intestinal bacteria, potently inhibited
beta-glucuronidase
. These results suggest that
beta-glucuronidase
inhibition by herbal medicines may protect against CCl4-induced liver injury.

Inflammatory reactions that result from microbial infections, both localized and systemic, are reported to cause transient or permanent male infertility. The cellular mechanisms underlying the inhibitory effect of microbial infection on spermatogenesis is not fully understood. However, there is evidence that spermatogenesis is affected by bacterial lipopolysaccharides (LPS) that induce acute inflammatory responses. The aim here was to use LPS treatments to investigate the potential oxidative stress and toxicity in primary cultures of adult rat Sertoli cells. The Sertoli cells were established and incubated with different concentrations of LPS (5, 10 or 20 microg/ml) for 6, 12 and 24h. Lipid peroxidation (LPO) and hydrogen peroxide (H(2)O(2)) production, along with superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione-S-transferase (GST), glutathione reductase (GR), reduced glutathione (GSH), lactate,
lactic acid dehydrogenase
(
LDH
), gamma-glutamyl transpeptidase (gamma-GT) and
beta-glucuronidase
were measured in these cells. LPO as well as H(2)O(2) production were significantly increased while antioxidant enzyme activities and GSH concentration were significantly depressed. Effects were dose and time-dependent at all incubation periods with 10 and 20 microg/ml LPS. Moreover, markers of Sertoli cell function such as lactate production,

beta-glucuronidase
activities were decreased in a time and dose-dependent manner. Incubation of Sertoli cells with 5 microg/ml LPS for 12 and 24h significantly increased oxidative status but significantly decreased the antioxidant enzyme activities, GSH concentration and Sertoli cell markers. In contrast, the oxidative and antioxidant status and markers of Sertoli cell function did not show any significant change in treated Sertoli cells with 5 microg/ml LPS for 6h. Therefore, it may be concluded that LPS induces oxidative stress in Sertoli cells and adversely affects Sertoli cell functions.
