In 24 men with precancerous states of the larynx, i.e. leukoplakia, papillomas and pachydermia, the peripheral blood lymphocytes were cytochemically stained for N-acetyl-beta-glucosaminidase,
beta-glucuronidase
, acid phosphatase and glycogen (
PAS
reaction). The results were expressed in terms of the absolute counts of enzyme- (or compound-) positive cells. The serum immunoglobulin IgG, IgA and IgM levels were also determined by Mancini's method. The results obtained were compared with those in 20 healthy men aged 20 to 30 years. It was found that the patients exhibited elevated numbers of N-acetyl-beta-glucosaminidase- and
beta-glucuronidase
-positive lymphocytes. A characteristic feature was an increase in the absolute counts of lymphocytes with diffuse and granular-diffuse types of cytochemical reaction for all enzymes studied. The number of cells with the granular type of enzymatic reaction (intact enzyme-positive lysosomes) was significantly diminished. These cytochemical alterations were accompanied by a significant increase in the serum IgA level. These results are discussed with reference to the lymphoid system response to tissues with precancerous lesions of the larynx.

The histochemistry of the hepatic parenchymal cells was studied in four Callithrix jacchus. A large amount of glycogen was noted throughout the lobules while the UDPG-GT and the phosphorylases were found unevenly distributed by the hepatic strands with different degrees of reactivity. -positive nuclear corpuscles that were more conspicuous in the hepatic cells with a larger amount of cytoplasmic glycogen and weaker UDPG-GT and phosphorylase reactivities. G-6PA (in a larger amount) and LDH (in a moderate amount) were found evenly distributed in the hepatic strands. F-1-6PA was seen sometimes with a stronger reactivity at the peripheral part of the lobules. The enzymes of the pentose shunt (G-6PDH, 6-PGDH and NADPH-2-TR) reacted strongly and as a rule evenly distributed near the hepatic lobules. Occasionally they reacted more intensely in the row of hepatic cells disposed just around the central vein. Cytochrome oxidase showed a very faint reaction. Cis-aconitase and ICDH were weak or moderate. NADH-2-TR more than SDH more than MDH were seen frequently diffused near the hepatic strands. SDH and MDH in some instances showed a stronger reactivity in the row or group of hepatic cells around the central vein. ATPase at pH 6.3 was negative in the marmoset liver; ATPase at pH 7.4 was mainly found in the wall of the portal area vessels; ATPase at pH 8.5 showed a stronger reactivity in the cytoplasm of the hepatic cells and ATPase at pH 9.4 was more abundant in the bile capillaries. The reactivity of the lipid metabolism enzymes was moderate with regard to alpha-GPDH or negligible with regard to beta-OHBDH. Acid phosphatase showed a stronger reaction, but almost limited to the Kupffer cells. The hepatic cells showed only a moderate amount of RNA. Some enzymes of the protein metabolism, such as GDH and leucine aminopeptidase showed a stronger reactivity while some others, such as alanyl aminopeptidase and MAO, were seen diffused near the hepatic lobules in a small amount. Enzymes of the mucopolysaccharide metabolism were not found at all (
beta-glucuronidase
) or showed only a weak reactivity, such as xylitol dehydrogenase.

The value of the cytochemical
beta-glucuronidase
(beta-gluc.) reaction in the differential diagnosis of acute leukaemia was assessed in a series of 100 adult patients. A purely granular type of reaction was observed in 7 out of 8 cases of lymphoblastic leukaemia and in 2 of 11 cases of acute leukaemia of uncertain type. Such an exclusively granular reaction was never seen in other types of acute leukaemia. In most cases of myeloblastic, promyelocytic, myelomonocytic and monocytic leukaemia, a positive staining reaction was noted which was either diffuse or a combination of diffuse and finely granular. The cells of one patient with lymphoblastic leukaemia were negative for beta-gluc. A coarsely granular
PAS
reaction was noted in 5 cases of lymphoblastic leukaemia including the one with negative beta-glu, reaction. Our results show that the beta-gluc, reaction is of definite value in the diagnosis of lymphoblastic leukaemia, and that it is probably more sensitive than the
PAS
reaction. In monocytic or myelomonocytic leukaemia, the pattern and intensity of staining did not differ appreciably from that seen in myeloblastic leukemia.

A case of diffuse large cell lymphoma of B-cell type with unusual azurophilic granules is reported. Lymphoma occurred primary in the upper anterior mediastinum and was suggested to be of thymic origin. Histologically, lymphoma cells showed diffuse proliferation and were large in size, frequently with multilobulated nuclei. In imprint preparations stained by May-Giemsa, most lymphoma cells had basophilic cytoplasm with azurophilic granules. Cytochemical studies showed the granules to be negative for

. Electron-dense granules and electron-lucent granules were found on ultrastructural analysis. method. In the youngest embryos, a high activity of DPP IV was recorded in the epithelium of differentiating primitive glandular tubules. Activity of other peptidases was low. The activity of AcP was found in tubular epithelium and mesenchymal cells. After week 7, glycogen was present in the supranuclear zone of tubule epithelium. In older fetuses, especially after week 15 of the intrauterine life, the activity of all studied enzymes gradually intensifies. In acinic anlage, a high activity of DPP IV was observed, activity of APM and GGT increased, activity of APA was lower. A relatively high activity of peptidases was recorded even in the epithelium of ducts. The capillaries showed a high activity of AlP and ATP-ase.

In 84 leukemic non-Hodgkin-lymphomas the diagnostic evidence of peripheral blood smears in Pappenheim-staining and diverse cytochemical reactions (
PAS
, alpha-naphthyl acetate esterase, acid phosphatase, acid esterase,
beta-glucuronidase
) was analysed using a homogeneous cell grid (lymphatic differential blood picture). In all entities the small lymphocyte proved to be the most frequent cell-form. The other lymphomas of low malignity (intermediate malignity) can clearly be demarcated from the CLL by the more intense polymorphism of the blood smears and in the majority can be classified entity-related by the differential blood picture. For the diagnostic evidence of the differential blood picture proved significant that entity-typical cell-forms (e.g. lymphoplasmacytoid lymphocytes, centrocytes) following the small (mature) lymphocyte as cell-type of second frequency are flooded out into the peripheral blood in non-Hodgkin-lymphomas. Highly malignant lymphomas could clearly be demarcated from low-malignant ones by their higher proportion of blasts in the differential blood picture. Of the cytochemical reactions only the acid phosphatase, with definitely focal-perinuclear reaction, an importance as T-cell marker is ascribed.
