Ammonium chloride
, a known inhibitor of lysosomal function, was found to be a rapid and potent inhibitor of 45Ca release from fetal rat bones in organ culture. The response to parathyroid hormone and prostaglandin E2 was inhibited in a dose-related, reversible fashion. The activity of the lysosomal enzyme
beta-glucuronidase
in the medium closely paralleled 45Ca release.
Ammonium chloride
may now be added to the list of antilysosomal agents that inhibit bone resorption in vitro.
