The 230-kbp murine cytomegalovirus (MCMV) genome is predicted to encode 182 open reading frames (orfs). One gene whose functional role is not known is encoded by the 762-bp m136 orf. Sequence analysis of rat cytomegalovirus (RCMV) strains Maastricht and English revealed homologous orfs, pr136, and
ORF
HJ4, respectively. Conservation of these orfs suggested that m136 and the RCMV homologs might play a role during virus replication. Expression of an epitope tagged form of m136 (m136-V5) yielded a polypeptide of 34 kDa that localized to the perinuclear region of transfected mouse 3T3 fibroblasts. Three independently generated MCMV m136 mutants were isolated and characterized. Mutations were introduced into the m136 orf by inserting either a
beta-glucuronidase
(m136-beta-gluc) or a guanosine phosphoribosyl transferase (m136-gpt) expression cassette into a unique BglII site, or by inserting a gpt cassette into a deleted region (Deltam136) of m136. No differences were observed in viral yield, plaque size, and plaque morphology between the parental strain and any of the m136 mutant viruses. In vivo analysis using a SCID mouse virulence model showed a consistently measurable attenuated phenotype for all three m136 mutants. The results showed that although the m136 gene was not essential for replication in vitro or in vivo, an intact m136 gene was necessary to yield wild type virulence during infection of the host.

Polyamines are small polycations essential for cell growth. Ornithine decarboxylase (ODC), the first enzyme of the polyamine synthesis pathway, is one of the most regulated enzymes of eukaryotes. In the present report, the transcriptional regulation of the gene encoding ODC from the dimorphic fungus Yarrowia lipolytica (YlODC) was analyzed. To this end we made a transcriptional fusion of its promoter with the
ORF
of the
beta-glucuronidase
gene from Escherichia coli, and analyzed the effect of polyamines and the dimorphic transition on the rate of transcription. The results obtained show that there exists a weak but reproducible transcriptional regulation of YlODC under the analyzed conditions. Our data suggest that gene transcription is down-regulated by putrescine, and stimulated by spermidine. It is suggested that the simultaneous operation of transcriptional and post-translational regulatory mechanisms endows the cell with a more efficient mechanism to control the physiological processes that depend on polyamines.

gene cloned from bacterial species. The

Finder analysis of a sequenced DNA (EMBL, AJ890474) revealed a presence of 1,785 bp large
ORF
potentially coding for a 594 aa protein. Three protein families in (Pfam) domains were identified using the Conserved Domain Database (CDD) analysis: Pfam 02836, glycosyl hydrolases family 2, triose phosphate isomerase (TIM) barrel domain; Pfam 02837, glycosyl hydrolases family 2, sugar binding domain; and Pfam 00703, glycosyl hydrolases family 2, immunoglobulin-like beta-sandwich domain. To gain more insight into the enzymatic activity, the domains were used to generate a bootstrapped unrooted distance tree using ClustalX. The calculated distances for two domains, TIM barrel domain, and sugar-binding domain were comparable and exhibited similarity pattern based on function and thus being in accordance with recently published works confirming
beta-glucuronidase
activity of the enzyme. The calculated distances and the tree arrangement in the case of centrally positioned immonoglobulin-like beta-sandwich domain were somewhat higher when compared to other two domains but clustering with other beta-glucuronidases was rather clear. Nine proteins, including beta-glucuronidases, beta-galactosidase, and mannosidase were selected for multiple alignment and subsequent distance tree creation.
