Acid phosphatase and beta-glucuronidase were used as marker enzymes of lysosome, and their role in adjuvant-induced uveitis was studied. These enzyme activities in the iris-ciliary processes were increased in the inflamed tissues. Changes in these enzyme activities in the tissues paralleled the development of uveitis. While protein concentration in the aqueous humor as a parameter of vascular permeability was significantly correlated with these enzyme activities, there was no correlation in the leucocyte counts in the aqueous humor. Topically applied dexamethasone reduced the increase in the aqueous protein, the leucocyte migration and the swelling of the iris-ciliary processes, while topically applied indomethacin reduced only the leucocyte migration among these inflammatory parameters. Acid phosphatase activities in the inflamed tissues were reduced also by dexamethasone, but not by indomethacin.

Chemotactic factors can cause polymorphonuclear leukocytes to release the contents of azurophilic granules, including the enzymes beta-glucuronidase and myeloperoxidase. In the presence of aqueous humor from the anterior chamber of the rabbit eye, the supernatant from stimulated leukocytes contains beta-glucuronidase, but myeloperoxidase is not detectable. Studies with aqueous humor and partially purified human myeloperoxidase suggest that this phenomenon is not due to a failure of enzyme release. The factor responsible for the inability to detect MPO in the assay system is heat-labile, dialyzable, and reversed by ascorbate oxidase. Comparable assay inhibition is produced by ascorbic acid at a concentration present in either human or rabbit aqueous humor. The ability of aqueous humor to protect against myeloperoxidase-induced oxidation may contribute to several diverse phenomena, including the susceptibility of the eye to Candida infection and a prolonged half-life for several inflammatory mediators in the anterior chamber.

To specify mechanisms of pathology development, we studied the activity of lysosomal glycosides in rabbit ocular tissues in experimental ophthalmoherpes. Rabbits with herpetic kepatitis show activation of beta-glucuronidase, beta-glycosidase, alpha-mannosidase in cornial epithelium and stroma, iris, aqueous humor of herpes-infected and contralateral eye. The activity of the above three glycosidases in the tears of children with herpetic keratitis was enhanced. If their activity does not regress after the treatment, ophthalmoherpes recurrence may be expected in the near future. For realization of its genetic program herpes simplex virus in infected cells activates synthesis of acid glycosidases this leading to degradation of cornial cell membranes, virus spread in the tissues and release of lysosomal enzymes into tear fluid.
