Spontaneous malignant fibrous histiocytoma (MFH) of the chest wall was found in a 10-year-old male Japanese white rabbit. Histologically, the MFH consisted mainly of areas of storiform, pleomorphic and myxoid patterns. Positive reactions for acid phosphatase (Ac-P), non-specific esterase (N-SE) and beta-glucuronidase (beta-GL) were demonstrated in fibroblast-like, histiocyte-like and giant cells. Moreover, a strongly positive fibronectin (FN) reaction was observed mainly in histiocyte-like and giant cells. In electron microscopy, tumor cells were composed morphologically of various types of cells such as fibroblast-like, histiocyte-like, undifferentiated and giant cells. This case was quite similar to those reported in man or other animals.

Malignant fibrous histiocytoma (MFH) was produced by injection of 9,10-dimethyl-1,2-benzanthracene (DMBA) into the rat knee joint. The tumor was observed in or around the knee in nearly all the animals 13 to 36 weeks after the initial DMBA administration. Histologically, these lesions were of the storiform-pleomorphic type (39/58, 67.2%), myxoid type (9/58, 15.5%), or giant cell type (8/58, 13.8%). Six cell types reported in human MFH were confirmed and phagocytosis of 0.81-micron latex particles by histiocyte-like cells was noted by electron microscopic examination. Acid phosphatase, beta-glucuronidase, and alpha-naphthyl acetate esterase were positive in enzyme histochemical examinations. Acid phosphatase activity was electron microscopically noted primarily in the lysosomes and the Golgi apparatus of the histiocyte-like cells. Cells from the storiform-pleomorphic (M1) and myxoid (M2) type tumors were serially transplanted subcutaneously in the back of the rats, and are now at the thirtieth and fortieth passage, respectively. They also were studied by enzyme histochemical and electron microscopic techniques. Our observations suggested an undifferentiated mesenchymal cell origin of MFH. Transplantable MFH can be produced in rats by intra-articular injection of DMBA, and lesions thus produced are a useful experimental model for the investigation of the histogenesis and the effect of chemotherapy of MFH.

A total of 19 cases with bone tumors, including six osteosarcomas. three giant cell tumors of bone, one malignant fibrous histiocytoma, four nonossifying fibromas, four chondromas and one chondrosarcoma, were examined as to enzyme histochemistry; the enzymes consisted of alkaline phosphatase (ALPase), acid phosphatase (ACPase), nonspecific esterase (NSE), adenosine triphosphatase (ATPase), 5'-nucleotidase (5'-Nucl) and beta-glucuronidase (beta-Gl). Osteosarcoma was strongly positive for ALPase followed by 5'-Nucl. Giant cell tumor, malignant fibrous histiocytoma and nonossifying fibroma showed enzyme histochemistry similar to each other: multinucleated giant cells and round cells in these tumors were strongly positive for ACPase, NSE, ATPase and 5'-Nucl simulating osteoclasts and histiocytes, whereas spindle cells were positive for ATPase and 5'-Nucl in their cytoplasm and weakly positive for ACPase. Chondroma and chondrosarcoma were focally positive for ACPase and NSE; the ACPase was sensitive to tartaric acid treatment. These observations showed that ALPase activity is very characteristic to osteosarcoma, and is useful for its diagnosis. From enzyme histochemistry, giant cell tumor, malignant fibrous histiocytoma and nonossifying fibroma can be regarded as a histiocyte-derived tumor of bone in contrast to osteosarcoma and cartilaginous tumors.

A tumor developed spontaneously in the subcutaneous tissue of the hind leg of a 7-month-old female ddY mouse. Light and electron microscopical examinations revealed that the original tumor was composed of an admixture of fibroblast-like and histiocyte-like cells arranged predominantly in a storiform or cartwheel pattern. The tumor cells gave positive reactions for acid phosphatase, N-acetyl-beta-glucosaminidase, non-specific esterase, beta-glucuronidase, alpha-1 antitrypsin and fibronectin. The original tumor was diagnosed as a malignant fibrous histiocytoma (MFH). The tumor was serially transplanted into syngeneic mice up to the 92nd generation. The tumor was also consistently transplanted into allogeneic mice of several inbred strains. The allogeneic mice used in the present study were strains having different H-2 haplotypes. During succeeding passages, transplanted tumors showed aberrant growth properties. The tumor transplanted into mice of inbred strains took well to back transplantation for mice of original strain and allotransplantation for other inbred strains. The pathological features of these transplantable tumors were basically similar to those of the original tumor. As mentioned above, a MFH developed spontaneously in the ddY mouse was consistently transplantable into both syngeneic and allogeneic mice.
