The enzymological studies on the sediment of the accumulation lake that has the main purpose of supplying drinking water to the city of Cluj-Napoca and the nearby villages, were aimed at the comprehensive understanding of the complex processes that happen in these habitats of special significance. In the sediment samples the following enzymatic activities have been quantitatively determined:
phosphatase
, actual and potential dehydrogenase, catalase, urease and protease. Non-enzymatic catalytic activity was also measured. Based on the relative values for the enzymatic activities, the enzymatic indicator of the sediment quality (EISQ) was calculated (ranging from 0.1 to 0.7). The enzymatic activities have been qualitatively determined for maltase,
saccharase
, lactase, cellobiase, amylase, dextranase, levanase, cellulase and inulinase. The correlation between the enzymatic and bacteriologic potential was statistically calculated.

As measured 7, 14, and 21 days after the application of 10(-2) M vanadyl sulfate solution to the foliage of 4.5-month-old sugar beet plants, significantly less growth of the leaves and an increase in the sucrose content of the storage root resulted. Accompanying these alterations were a higher rate of carbon dioxide fixation, a lower rate of respiration, and a decreased rate of nitrate reductase, glutamic-pyruvic transaminase,
phosphatase
, and
invertase
activity. The enzymes of sucrose synthesis, sucrose synthetase, sucrose phosphate synthetase and uridine diphosphate glucose-pyrophosphorylase were stimulated. The content of reducing sugar, nitrite N, amino acids and protein was less, and that of nitrate N was greater in the vanadium-treated plants. In the majority of cases the greatest magnitude of change occurred during the first 7 days following treatment. The changes in growth and chemical composition are believed to be closely related to the stimulation or inhibition of the various enzymes by vanadyl sulfate.

The hydrolase content of vacuoles isolated from protoplasts of suspension-cultured tobacco cells, of tulip petals, and of pineapple leaves, and the sedimentation behavior of tobacco tonoplasts were studied. Three precautions were found to be important for the analysis of vacuolar hydrolases and of the tonoplast. (a) Purification of protoplasts in a Ficoll gradient was necessary to remove cell debris which contained contaminating hydrolases adsorbed from the fungal cell-wall-degrading enzyme preparation. (b) Hydrolase activities in the homogenates of the intact cells or the tissue used and of the purified protoplasts had to be compared to verify the absence of contaminating hydrolases in the protoplast preparation. (c) Vacuoles obtained from the protoplasts by an osmotic shock had to be purified from the lysate in a Ficoll gradient. Since the density of the central vacuole approximates that of the protoplasts, about a 10% contamination of the vacuolar preparation by surviving protoplasts could not be eliminated and had to be taken into account when the distribution of enzymes and of radioactivity was calculated.THE INTRACELLULAR ACTIVITIES OF THE FOLLOWING ACID HYDROLASES WERE PRIMARILY LOCALIZED IN THE VACUOLE OF TOBACCO CELLS: alpha-mannosidase, beta-N-acetylglucosaminidase,

Proteinase, a hydrolase with low activity in tobacco cells and tulip petals and therefore difficult to localize unequivocally, was found to be vacuolar in pineapple leaves, a tissue containing high levels of this enzyme. Our data support the hypothesis that the central vacuole of higher plant cells has an enzyme composition analogous to that of the animal lysosome.None of the vacuolar enzymes investigated was found to be bound to the tonoplast. When vacuoles were isolated from cells labeled with radioactive choline, the vacuolar membrane was found to contain radioactivity. On sucrose gradients, the label incorporated into tonoplasts banded around a density of 1.10 grams per cubic centimeter (24% sucrose, w/w).

A plasma membrane-enriched fraction was isolated from various tissues of developing lima bean seedlings, Phaseolus lunatus var Cangreen, to study beta-1,3-glucan synthase activity changes. All tissues contained an active beta-glucan synthase, including the cotyledons that will be senescent in mature lima bean plants. Young primary leaves exhibited a very active beta-glucan synthase; but this activity dropped markedly, about fivefold, as the leaves gained weight and became photosynthetic. Some tissues, such as the hypocotyl and young stem, exhibited an increase in beta-glucan synthase activity as the tissues were growing and a decrease as the growth rate slowed. Roots exhibited a high activity early in development that only decreased slightly, about 30%, as root growth increased. Surprisingly the senescent cotyledons contained an activity equivalent to some other tissues that was maintained over our measurement time of 21 days. Perhaps this callose synthesis activity is related to translocation processes as the cotyledons transfer their reserves to the growing seedling. We concluded that beta-glucan synthase was not a good indicator of sink strength in these lima bean tissues. The plasma membrane fractions also were tested for other enzymes that might be present because an electron microscope study revealed a low contamination by other types of membranes. The membrane fractions had low but detectable activities of sucrose synthase, UDPglucose pyrophosphorylase, UDPase,
alkaline invertase
, and a general
phosphatase
; but these enzymes exhibited no consistent pattern(s) of activity change with plant development.

This paper studied the dynamics of microbial flora and enzyme activity in the rhizosphere of allelopathic rice PI312777 (PI) and non-allelopathic rice Lemont (LE) at the growth stage of 3 - 7 leaves. The results showed that in the rhizosphere of PI, the amounts of bacteria, actinomycetes and azotobacter were 11.2% - 28.3%, 40% - 78.6% and 111.5% - 173.9%, respectively, while that of fungi was lower, with the maximum being 25.5% of that in the rhizosphere of LE, suggesting that allelopathic rice PI promoted the growth of bacteria, actinomycetes and azotobacter, but inhibited that of fungi. Further analysis on the physiological groups of microbial flora showed that PI favored the growth of ammonifier, aerobic azotobacter, aerobic cellulose - decomposer, sulphate - reducer, nitrite - bacteria and nitrate - bacteria, among which, ammonifier and aerobic azotobacter increased by 53.7% and 57.6%, respectively, while inhibited the growth of desulphate bacteria and denitrifyier. were sensitive to heavy metals pollution, and their activities could be used as the indicators of heavy metals' joint pollution. The effects of rhizosphere environment on the soil enzyme activities were in the sequence of

> polyphenol-oxidase, and the affecting rate was 131.562%, 92.492%, 87.557%, 59.673% and 34.076%, respectively. The test enzyme activities were negatively correlated with soil heavy metals pollution, and the correlation coefficients were all higher than -0.898, suggesting the inhibitory effects of heavy metals' joint pollution on soil enzyme activities. P. ostii could effectively improve soil environment, and thus, enhance the activities of soil enzymes.

With simulation test, this paper studied the effects of Hg on the activities of urease,
invertase
and neutral phosphotase in four soils. The results showed that Hg inhibited soil urease and
invertase
activities markedly, but its inhibitory effect differed with test soils. There was a significant logarithmic correlation between the concentration of HgCl2 and the activities of these two enzymes (P < 0.05). In test soils, the ED50 of urease activity was 87.99, 5.47, 24.05 and 19.88 mg x kg(-1), and that of
invertase
activity was 76.68, 727.49, 236.52 and 316.59 mg x kg(-1), respectively. Urease was more sensitive than
invertase
to Hg contamination, while organic matter had a protective effect on soil enzymes. Soil neutral
phosphatase
was not sensitive to Hg contamination, except that it was significantly activated by Hg in the meadow brown soil applied with plenty of organic fertilizer.

We report the complete nucleotide sequence and characterization of a cryptic plasmid, pHLHK26, recovered from a strain of Laribacter hongkongensis isolated from a patient with community acquired gastroenteritis. pHLHK26 consists of 8700 bp, with G + C content 51.3%. The copy number (mean +/- SD) is 0.57 +/- 0.07 and it is stable after four passages (about 240 generations) in the absence of selection. There is a predicted origin of replication that consists of a DnaA box and five 22-bp direct repeats. pHLHK26 has four ORFs with two genes encoded in the sense direction and the other two in antisense direction. These four ORFs encode a putative plasmid partitioning protein of the ParA family, a putative protein that contains putative ADP-ribose 1"-
phosphatase
activity belonging to the Appr-1-p processing enzyme family, a putative recombinase (TniR) of the resolvase/
invertase
family, and a putative replication protein, respectively. We speculate that pHLHK26 is a theta, possibly Class A, replicative plasmid, as it contains an origin of replication with AT-rich region, a number of iterons and a DnaA box and a gene that encodes a replicative protein most homologous to those of other theta replicative plasmids and it shares eight of the nine positions of the consensus sequence TTAT(C/A)CA(C/A)A (TTTTCCACA in pHLHK26) in the DnaA boxes observed in other classical examples of Class A plasmids of this group.

In a long-term experimental plot at the Hailun Agricultural Ecology Station of Chinese Academy of Sciences in Heilongjiang Province, Northeast China, the effects of three land management practices including natural restoration, fallowing and cropping on black soil microbial biomass C and enzyme activities at the depths of 0-10, 20-30 and 40-50 cm were investigated. The results showed that at 0-10 cm, soil microbial biomass C and soil urease,
phosphatase
, catalase and

activities were in the sequence of natural restoration > cropping > fallowing, while at 20-30 cm and 40-50 cm, they had less significant differences as those at 0-10 cm. Under natural restoration and cropping, soil microbial biomass C and soil urease,
phosphatase
and catalase activities decreased with increasing soil depth, while under fallowing, they were higher at 20-30 cm. Correlation analysis showed that there existed significant positive correlations between soil total C and total N, soil total C and microbial biomass C, and soil microbial biomass C and test enzyme activities. The indices soil microbial biomass C and soil enzyme activities showed that black soil under long-term natural restoration had better soil quality.
