Conflicting results have been obtained in previous studies concerning the adaptation of intestinal blush border membrane enzymes to starvation. This study was designed to clarity the changes in these enzymes under starvation conditions, using a molecular biological approach. Sprague-Dawley rats were starved or given total parenteral nutrition (TPN) for 5 days. Rats allowed free access to food were used as controls. Changes in the activity and expression of jejunal brush border membrane enzymes were compared between three groups. In the starved group, aminopeptidase N and dipeptidyl peptidase IV activity was significantly elevated to 177% and 166%, respectively, of control values. In contrast, sucrase and maltase activity was significantly decreased. The activity of these peptidases also tended to be increased at the renal brush border membrane. Up-regulation of peptidase activity was not evident in the TPN group. Western and Northern blot analysis revealed that the changes in aminopeptidase N activity were attributable to increases in the protein and mRNA level. The activity and expression of brush border membrane peptidases in rat jejunum is up-regulated during starvation, and these changes are considered to be an effect of whole-body malnourishment, rather than an absence of luminal nutrition.

Our objective was to examine the distribution of enterocyte digestive enzyme activity along the crypt-villus and longitudinal axes of the small intestine in formula-fed neonatal pigs between the ages of 14 and 18 d. The distended intestinal sac method was used to isolate 12 sequential fractions (F1 through F12) of epithelial cells. Enterocyte migration rate was measured in the proximal and distal intestine using in vivo bromodeoxyuridine labeling. Specific activities of representative villus cell marker enzymes of alkaline phosphatase, aminopeptidase N, sucrase, and lactase increased 6- to 17-fold from F12 (crypt cells) to F1 (villus cells), whereas the crypt cell marker [3H]thymidine incorporation increased 8- to 18-fold from F1 (villus cells) to F12 (crypt cells). Enterocyte migration rate was similar (3.2 vs 3.0 microm/h), whereas the villus height (547.4 vs 908.5 microm) and enterocyte life span (4.7 vs 10.2 d) were markedly lower (P < 0.05) in the proximal than in the distal segments, respectively. In general, the specific activities of all enzymes were lowest in the crypt fractions (F9 through F12) but increased markedly (ranging from 8- to 17-fold) from F12 to F1. The activity of aminopeptidase N was higher and that of sucrase was lower in the distal than in the proximal segment. The activities of the remaining enzymes were similar in the proximal and the distal segments. Our results suggest that the enterocyte life span in the distal small intestine is approximately twice as long as in the proximal small intestine. However, despite the difference in life span, the patterns of enzyme activities along the crypt-villus axis were generally similar in the proximal and the distal regions.

The effects of 10-day bilateral adrenalectomy on morphometry, proliferation, and apoptosis were determined in the small intestine of 3-month-old Sprague-Dawley rats. The activities of sucrase, lactase, and its respective mRNA, aminopeptidase N, and intestinal alkaline phosphatase were also evaluated. Adrenalectomy lead to partial atrophy and disorganization of the epithelium, with an increased number of goblet and Paneth cells and a reduction of crypt cell proliferation paralleled by a marked increase in villus apoptosis. Biochemical assays revealed that aminopeptidase N and intestinal alkaline phosphatase activities were significantly decreased, whereas disaccharidases were increased by adrenalectomy. The corresponding induction of lactase mRNA suggests an active response of the epithelium. In conclusion, adrenalectomy modified maturation and the differentiation processes of the small intestinal mucosa, especially in the proximal part of the small intestine. This result points to an important role of adrenals and glucocorticoids in the trophic status of the adult small intestinal mucosa.

1. Investigations were conducted into the development of intestinal enzyme function in broiler chickens on a commercial starter diet. The differences between intestinal regions and localisation of enzymes on the villus were assessed. 2. The specific activity of maltase, sucrase, aminopeptidase N (APN) and alkaline phosphatase (AP) at all intestinal sites decreased with age. There were also variations between intestinal sites although this variation depended on age. The specific activity of maltase was higher than that of the other enzymes examined, regardless of age and intestinal site. The total activities of the enzymes also increased with age at all intestinal sites. 3. Results of the localisation of enzymes on the crypt: villus axis showed that activity was expressed over a large proportion of the villus. There was an increase in the total villus activity of alpha-glucosidase (AG), APN and AP with age. Activity per unit villus surface area was similar between ages, except for jejunal AP. At hatch enzyme activity was expressed over 44.1, 55.8 and 63.3% of villus height in the duodenum, jejunum and ileum, respectively. At 21 d of age, corresponding values were 68.7, 65.6 and 77.2%. The point of peak activity from the crypt: villus junction increased with age. In the jejunum, most enterocytes were capable of secreting active enzymes within 1 h of formation. Cells maintained their secretory capabilities until they were more than 60 h old in the case of AG. 4. Although the specific activities of the enzymes were maximal at hatch, the digestive capacity of older birds may be sustained by an increase in total enzyme activity brought about by increased surface area. The pattern of enzyme activity along the gastrointestional tract (GIT) and crypt: villus axis is similar to that reported for some mammalian species.

Uptake of colostrum just after birth is essential to stimulate intestinal growth and function, and in many species, including pigs, colostrum also provides immunological protection via the absorption of immunoglobulin G (IgG). In this study, intestinal growth, IgG absorptive capacity and enzyme activities were investigated in newborn pigs in response to different diets. Newborn piglets were bottle-fed porcine colostrum (PC), bovine colostrum (BC), porcine plasma (PP), porcine milk (PM), bovine colostrum containing porcine plasma (BCP) or a milk replacer (MR) every 3 h (15 mL/kg) for up to 2 d. Bovine serum albumin (BSA) was added to the diets as a macromolecule marker. The percentage of absorbed BSA just after birth was highest for piglets fed the PC diet (30-50%), lower for those fed the BC and BCP diets (23-30%) and lowest for the PP, PM and MR diet-fed piglets (7-20%, P < 0.05 relative to those fed colostrum). Porcine IgG was absorbed more efficiently than bovine IgG. Intestinal closure occurred earlier in MR and BCP piglets (within 12 h after birth) than in PC pigs. At 2 d of age, intestinal mucosal weight (+120% increase from birth) and villus morphology were similar in the PC, BCP and MR groups. All 3 groups also had increased aminopeptidase A activity compared with values at birth (+100% increase). Compared with PC pigs, the BCP group had higher sucrase and maltase activities (+50% and +200%, respectively) and lower aminopeptidase N activity (-50%, P < 0.05). Similarly, MR pigs showed elevated sucrase activity (+40%) and lowered maltase, lactase and aminopeptidase N activities (-20% to -50%, P < 0.05) compared with PC pigs. We conclude that porcine and bovine colostrum contain factors that stimulate the intestinal endocytotic and enzymatic capacity in newborn pigs. A milk replacer can produce normal gut growth, but may be inefficient in mediating normal macromolecule transport and disaccharidase activity. Bovine colostrum mixed with porcine plasma proteins may be a useful substitute for porcine colostrum in artificial rearing of newborn pigs.

Our objectives were to determine postnatal changes in the maximal enzyme activity (V(max)) and enzyme affinity (K(m)) of jejunal mucosal membrane-bound alkaline phosphatase, aminopeptidase N and sucrase using a porcine model which may more closely resemble the human intestine. Jejunal brush border membrane was prepared by Mg(2+)-precipitation and differential centrifugation from pigs of suckling (8 days), weaning (28 days), post-weaning (35 days) and adult (70 days) stages. p-Nitrophenyl phosphate (0-8 mM), L-alanine-p-nitroanilide hydrochloride (0-28 mM) and sucrose (0-100 mM) were used in alkaline phosphatase, aminopeptidase N and sucrase kinetic measurements. V(max) of alkaline phosphatase was the lowest in the adult (4.27 micromol.mg(-1) protein.min(-1)), intermediate in the suckling (9.75 micromol.mg(-l) protein.min(-l)) and the highest in the weaning and post-weaning stage (12.83 and 10.40 micromol.mg(-l) protein.min(-l)). K(m) of alkaline phosphatase was high in the suckling and weaning stages (5.14 and 9.93 mM) and low in the adult (0.66 mM). V(max) of aminopeptidase N was low in the suckling (7.04 micromol.mg protein(-1).min(-1)) and high in the post-weaning stage (13.36 micromol.mg(-l) protein.min(-l)). K(m) of aminopeptidase N was the highest in the two weaning stages (2.96 and 3.39 mM), intermediate in the adult (2.33 mM) and the lowest in the suckling stage (1.66 mM). V(max) of sucrase increased from the suckling to the adult (0.48-1.30 micromol.mg(-l) protein.min(-l)). K(m) of sucrase ranged from 11.19 to 16.57 mM. There are dramatic postnatal developmental changes in both the maximal enzyme activity and enzyme affinity of jejunal brush border membrane-bound alkaline phosphatase, aminopeptidase N and sucrase in the pig.

Maturation of the fetal gastrointestinal tract (GIT) is influenced by both luminal stimuli (e.g. swallowed fluid) and hormonal factors (e.g. endogenous cortisol release). The aims of the present study were 1) to investigate GIT growth and maturation during the last 20% of gestation in pigs (term = 114 +/- 2 d), and 2) to investigate the effect of esophageal ligation, to prevent fetal swallowing, at 80% to 91% gestation. In normal fetuses, marked increases occurred during late gestation in body weight (+95%), relative intestinal weight (+79%, g kg(-1) body weight), activity of some digestive enzymes (1.5- to 10-fold), and absorption of glucose and intact proteins (3- to 6-fold). Fetuses with ligated esophagi had lowered body weight (-20%), reduced intestinal weight (-43%), aminopeptidase A activity (-24%), and glucose absorption (-27%), while lactase, sucrase, and dipeptidylpeptidase IV activities were increased (+40-50%), compared with sham-operated fetuses (all p < 0.05). Other parameters of GIT function remained unchanged by esophageal obstruction (absorption of amino acids and immunoglobulin, activity of chymosin, amylase, trypsin, chymotrypsin, maltase, aminopeptidase N -- all expressed per gram GIT tissue). Ligated fetuses had elevated cortisol levels, which is known to stimulate fetal GIT maturation. We conclude that the rapid development of GIT function in late gestation is diminished by esophageal obstruction, mainly due to slower GIT growth and not inhibition of normal functional development of enterocytes.

To simulate the effects of nutritionally adequate and inadequate vegetarian diets, rats were fed, for 28 days, an isonitrogenous, isocaloric, amino acid unbalanced cereal diet (CD) deficient in lysine and tryptophan or a balanced cereal-legume diet (CLD). The impact of these diets on enzymes responsible for digestion of proteins and carbohydrates were measured. Neither experimental diet significantly affected the animal's final weight or feed consumption in comparison with controls fed a standard mixed diet from plant and animal sources. However, during the first three weeks, the weight gain of rats fed the CD was significantly lower (p < 0.01; p < 0.05) than that of the controls. CD fed rats also had a higher feed efficiency ratio (p < 0.05), demonstrating increased feed consumption per unit of body weight. They also had decreased pancreatic alpha-amylase activity (p < 0.05), serum phytolytic and zoolytic alpha-amylase activity (p < 0.05) and serum protein level (p < 0.05) than the controls. Activity of pancreatic trypsin and intestinal enzymes (sucrase, maltase, aminopeptidase N) were the same as in the controls. In rats fed CLD, growth, food consumption, and enzyme activities did not change, however serum protein and glucose levels were higher (p < 0.025; p < 0.005) than in the controls. It is hypothesized that decrease in alpha-amylase activity was mostly related to the tryptophan deficiency in the CD because this enzyme contains the highest amount of tryptophan units among all tested enzymes.

Research on cancer and other conditions has shown flavonoids and sphingolipids to be food components capable of exerting chemoprotective action. Nevertheless, little is known about their effects on healthy individuals and their potential usefulness as therapeutic agents. The present study examined the possible action of a dietary flavonoid, quercetin, and a sphingolipid, sphingomyelin, as functional foods in healthy animals. In particular, the effect on animal growth of supplementing a conventional diet with one or other of these substances (0.5 % quercetin and 0.05 % sphingomyelin) was considered. Possible action affecting intestinal physiology was also analysed by measuring the uptake of sugar and dipeptide, mediated by the Na(+)-dependent sugar transporter SGLT1 and the dipeptide Na(+)/H(+) exchanger PEPT1 respectively, and the activity of related intestinal enzymes such as sucrase, maltase and aminopeptidase N. Both substances seemed to modify small intestinal activity in healthy mice, altering intestinal enzymatic activity and nutrient uptake. These effects observed in the small intestine did not impair normal development of the animals, as no differences in serum biochemical parameters or in organ and body weights were found. The findings should help in elucidating the mechanisms of action of these food components with a view to their possible use in the prevention of certain pathological conditions.

The main objective of this study was to determine the effect of fiber source and concentration on morphological characteristics, mucin staining pattern, and mucosal enzyme activities in the gastrointestinal tract of pigs. The experiment included 50 pigs from 10 litters weaned at 4 wk of age (BW 8.6 +/- 1.4 kg) and divided into 5 treatment groups. Diets containing fiber of various physico-chemical properties and concentrations were formulated to contain 73, 104, or 145 g of dietary fiber/kg of DM. The diets were based on raw wheat and barley flours. Pectin and barley hulls, representing soluble and insoluble fiber sources, respectively, were used to increase the fiber concentration. The pigs were fed the experimental diets for 9 d, and then the pigs were euthanized and the entire gastrointestinal tract was removed. Tissue samples were taken from the mid and distal small intestine and from the mid colon. Inclusion of pectin in the diets significantly decreased (P < 0.001) ADFI and ADG compared with pigs fed no pectin. The villi and the crypts were shorter in pigs fed pectin-containing diets, but the villous height/crypt depth ratio was unaltered. Pectin significantly decreased the area of mucins in the crypts of the small intestine, indicating that the pigs fed the pectin-containing diet would probably be more susceptible to pathogenic bacteria, although this cannot be separated from the impact on ADFI. The lectin-binding pattern of the intestinal mucosa was unaffected by diet. The activity of lactase and maltase was increased in pigs fed diets with high fiber content, whereas sucrase activity was increased in pigs fed the pectin-containing diets. The activity of the peptidases, aminopeptidase N and dipeptidylpeptidase IV, was increased when feeding high fiber diets, whereas the activity of gamma-glutamyl transpeptidase remained unaffected by the experimental diets. In conclusion, the reduced feed intake observed with the pectin-containing diets could explain the lower villous height and crypt depth observed in this study. However, direct effects of pectin also are possible, and thus further study is warranted. Feeding pigs high insoluble fiber diets improved gut morphology by increasing villi length and increased mucosal enzyme activity when compared with pigs fed pectin-containing diets. The mucin content as determined by staining characteristics suggests that pigs fed high insoluble fiber diets might be better protected against pathogenic bacteria than pigs fed diets high in soluble fiber.
