The exocellular D-glucosyltransferase from Streptococcus mutans 6715 has been highly purified with minimal loss of enzymic activity. The organisms were cultured in trypticase soy-broth that had been treated with
invertase
and filtered through an ultrafilter fitted with a membrane having a cut-off molecular weight at 10,000. To the growth medium was added
Tween 80
, which prevented the enzyme from aggregating. The final step in the purification employed insoluble, streptococcal dextran as an affinity support. Two D-glucosyltransferase activities were detected, viz., one that did not adsorb to the insoluble dextran and one that did. The enzymic fraction that had adsorbed to the insoluble dextran in the affinity column was strongly inhibited by added insoluble dextran.

sucrase
, beta-1 --> 3 glucanase, xylanase, purine nucleosidase, and benzoyl esterase. The action appears to be an effect of the surfactant on cell permeability.

A simple, rapid and reproducible procedure for the identification and determination of extracellular
saccharase
from culture medium of watermelon cell suspension cultures is described. The culture medium (without cells) was used for the identification and determination of extracellular enzyme activity. Intracellular activity was estimated from the cell suspension. Watermelon cell suspension was permeabilized by
Tween 80
and immobilized by glutaraldehyde. The highest
saccharase
activity was at pH 4.6 at a temperature of 50 degrees C. The hydrolysis of substrate was linear 5h after reaching 60% conversion. The cells had high
saccharase
activity and good stability, and in long-term storage they showed convenient physico-mechanical properties.

Bioremediation of PAHs contaminated soil from Beijing Coking Plant was performed using a novel fungal strain Lasiodiplodia theobromae (L. theobromae). Moreover, enhanced bioremediation of PAHs contaminated soil was investigated in the presence of different concentrations of
Tween 80
and hydroxypropyl-beta-cyclodextrin (HPCD). The correlation of the dynamics of enzyme activities during remediation and the degradation of PAHs was analyzed. The results showed that the degradation rate of PAHs increased to 45.3% on the 70th day after addition of L. theobromae, which was 30 percentage points higher than that of the control group. At an optimum concentration of 2 g x kg(-1) for
Tween 80
and 1 g x kg(-1) for HPCD, the degradation rate of PAHs was enhanced to 65.8% and 63.9%, respectively, which was 50 percentage points higher than that of the control group. Hydrogen peroxidase and
invertase
activities in soil in the bioremediation group with only L. theobromae and the surfactant enhanced group were both enhanced twice more than that of the control group. These results showed that L. theobromae may produce hydrogen peroxidase and
invertase
or have synergic effect with indigenous microorganisms. Correlation analysis showed that the correlation coefficients of PAHs degradation rate and maximum enzyme activities of hydrogen peroxidase and
invertase
were 0.781 and 0.837, respectively. Therefore, the correlation between
