Leucine beta-naphthylamidase associated with the microvilli membranes of rabbit small intestine was solubilized with papain [EC 3.4.22.2] and purified by Sephadex G-200 gel filtration, DEAE-cellulose column chromatography, passage through a column of Sepharose 4B coupled with anti-
sucrase
antibodies and preparative disc electrophoresis in polyacrylamide gel. The purified enzyme was homogeneous on ultracentrifugation and disc electrophoresis, but a double immunodiffusion test showed the presence of a minor component which was probably denatured enzyme. The molecular weight of the purified enzyme was estimated to be 225,000 by Sephadex G-200 gel filtration and the sedimentation coefficient (S-0-20, w) was found to be 6.90S. Purified enzyme required bovine serum albumin for maximal activity, perhaps for its protection from autodigestion. It hydrolyzed, in addition to
L-leucine
beta-naphthylamide, various L-amino acid beta-naphthylamides and dipeptides with a free alpha-amino group, but did not hydrolyze benzoyl-L-arginine beta-naphthylamide. Therefore, the purified enzyme is an aminopeptidase. Hg-2+ and Cu-2+ ions strongly inhibited the enzyme activity, but other metal ions and EDTA showed no or only slight effect. N-Ethylmaleimide exhibited a weak inhibition. Purified enzyme had an optimal pH and Km value for leucine beta-naphthylamide similar to those of enzymes from other sources. Antibodies against the purified enzyme were raised in guinea pigs. The antibodies obtained were found by double immunodiffusion to be specific for the enzyme. They precipitated the enzyme quantitatively and partially inhibited the enzyme activity.

Nicarbazin is an anticoccidial drug, used mainly in birds, which has shown several other effects including inhibition of growth and feed efficiency in poultry, and stimulation of sugar and amino acid intestinal absorption in rabbit. The present work was designed to determine whether nicarbazin added to the feed, affects growth and feed intake in rabbit, and whether the continuous ingestion of nicarbazin can alter the mechanisms of intestinal nutrient absorption or digestion in this species. Nicarbazin, administered at the recommended dose (125 ppm) had no harmful effects either on growth or on feed intake of animals. After treatment for one month with nicarbazin at the dose of 125 ppm added to the feed, rabbits displayed a higher transport ability of both D-glucose and
L-leucine
through the enterocyte plasma membranes than did untreated rabbits. These animals also showed higher specific activities of two brush-border enzymes,
sucrase
and aminopeptidase N, than the control animals.

Metronidazole (Flagyl), an antibiotic commonly used in treating intestinal infections, when administered orally at a dose level of 100 mg/kg body weight daily for 7 days to rats brought about a significant elevation of the uptake of end-product nutrients like D-glucose, L-alanine, L-aspartic acid and
L-leucine
in the intestinal segments. Brush border membrane-bound hydrolytic enzymes, i.e.
sucrase
, lactase, maltase, alkaline phosphatase and leucine aminopeptidase levels, were also elevated. Substrate kinetic analysis of the uptake of nutrients as well as the enzymes indicated that the drug increased the maximum of apparent initial velocity, while the substrate affinity constants did not change. Studies of the temperature-dependent parameters of the nutrient uptake and the enzyme activity revealed that metronidazole did not induce any shift in the transition temperature (T(o)) for the uptake but the energy of activation (Ea) was reduced in all the cases except those of maltase and leucine aminopeptidase, which registered an increase in Ea and a marginal shift in T(o), respectively. A significant elevation was seen in the levels of membrane cholesterol, phospholipid, ganglioside and plasmalogen in metronidazole-treated animals, while triglycerides and the non-esterified fatty acids remained unaffected. The effects produced by metronidazole treatment persisted in the animals, which were allowed a recovery period of 7 days after the drug regimen.

Oral administration of embelin (75 mg/kg per day, daily for 15 and 30 days) to male rats caused significant elevation in the uptake of D-glucose, L-alanine,

and calcium in small intestinal segments. Embelin also produced significant increases in intestinal brush border membrane-associated enzymes (
sucrase
, lactase, maltase, alkaline phosphatase and leucine aminopeptidase) in both intestinal homogenates and partially purified brush border membrane preparations. Significant increases were also noted for microsomal glucose-6-phosphatase and cytosolic lactate dehydrogenase. Increase in brush border membrane-associated total lipids, phospholipids, cholesterol, triacylglycerol, unesterified fatty acids and ganglioside sialic acid were seen but not in the cholesterol/phospholipid molar ratio. All these changes returned to control or near control levels following withdrawal of the drug.

In the present experiments, selective quenching by trinitrophenyl groups as well as steady-state fluorescence polarization and differential polarized phase fluorescence techniques, using three different lipid soluble fluorophores, were used to directly examine the fluidity of the exofacial and cytofacial leaflets of rat small intestinal brush-border membranes. These studies revealed that the fluidity of the exofacial hemileaflet was greater than the cytofacial hemileaflet. Differences in the distribution of phosphatidylcholine and phosphatidylethanolamine, as assessed by phospholipase A2 treatment and trinitrophenylation of aminophospholipids, were, at least partially, responsible for the asymmetrical fluidity of the hemileaflets. Moreover, in vitro addition of benzyl alcohol (final concn 25 mM) preferentially fluidized the exofacial leaflet and concomitantly decreased leucine aminopeptidase activity but did not affect the activities of maltase,
sucrase
, alkaline phosphatase, or gamma-glutamyltranspeptidase. In vivo addition of the membrane-mobility agent 2-(2-methoxyethoxy)ethyl 8-(cis-2-n-octylcyclopropyl)octanate] (A2C) (final concn 7.5 microM) preferentially fluidized the cytofacial leaflet and increased Na(+)-gradient-dependent D-glucose transport but not Na(+)-gradient-dependent

, and calcium in the small intestinal segments. An increase was also noted in the intestinal brush border membrane (BBM)-associated enzymes,
sucrase
, lactase, maltase, alkaline phosphatase, and leucine aminopeptidase in both the intestinal homogenates and partially purified BBM preparations, particularly after 30-day administration of the drug. Embelin treatment also caused a significant increase in the microsomal glucose-6-phosphatase and the cytosolic enzyme, lactate dehydrogenase. In the Embelin-treated animals BBM-associated total lipids, phospholipids, cholesterol, triacylglycerol, unesterified fatty acids, ganglioside-sialic acids as well as the cholesterol/phospholipids molar ratio showed a considerable increase. All these changes in the Embelin-treated animals were restored back to the normal or near normal biochemical makeup when the drug therapy was withdrawn and the animals were allowed to recover for another 15 and 30 days, respectively.

Full-value diets of similar composition were given to male rats weighing 207-230 g, by intravenous (group 1) or intragastric (group 2) routes. The proportion of amino acids, fats and carbohydrates was 9.9:15.7:74.4 (with regard to their calorific value). The diet calorific value comprised 60.6 kcal/rat/day. dipeptidase were assayed in the intestinal mucosa of the proximal portion of the small intestine in group 1 rats, while a decreased alpha-amylase activity in the distal portion of the small intestine was recorded in the animals of group 2. The mass of the pancreas in the rats of group 1 and 2 was authentically lower than in the control rats which received oral feeding with natural foods. The lowest mass of the pancreas was observed in the rats of group 1. Specific activity of trypsin, lipase and RNase in the pancreatic tissues of rats in groups 1 and 2 was similar. The results of the study have evidenced a lowered function of the digestive system under conditions of artificial feeding, especially in case of intravenous nutrition.

The influence of two new 1-desoxynojirimycin derivatives, BAY m 1099 and BAY o 1248, on rat small intestinal disaccharidases (
sucrase
, maltase, isomaltase, glucoamylase, lactase, trehalase) and alkaline phosphatase activity has been investigated in vitro. Both compounds are very potent alpha-glucosidase inhibitors. Tested in the range of 0.1-5.0 micrograms/ml, inhibition is strongest on
sucrase
(up to 97.1%) and glucoamylase (up to 96.7%). BAY m 1099 also reduced (up to 56.4%) beta-galactosidase (lactase) activity. For both inhibitors a competitive type of
sucrase
inhibition was demonstrated (Lineweaver-Burk plot). Affinity versus
sucrase
was unusually tight. The Ki of BAY m 1099 versus
sucrase
amounted to 1.14 x 10(-7) M and of BAY o 1248 to 6.92 X 10(-8) M (Dixon plot). Both inhibitors did not impair active transport of
L-leucine
or methyl-alpha-D-glucoside into everted rings of rat jejunum in vitro.

The development and distribution of peptidase activity in mucosal homogenates of rat small intestine has been investigated. Substrates used were
glycyl-L-leucine
(GL), L-seryl-L-methionine (SM), and L-leucyl-glycyl-glycine (LGG). During the first 2 weeks of life there was high peptidase activity toward GL and SM in the distal regions of the small intestine. In the third postnatal week, activity in the distal small intestine toward GL and SM decreased, while activity in the proximal small intestine increased. In contrast, there was no difference in activity toward LGG along the length of the small intestine, nor was there a developmental change. Activity toward all three substrates was not affected by cortisone acetate treatment. However, the classical effect of glucocorticoids on
sucrase
activity and body weight was observed. All peptidases studied showed maximal activities at neutral pH, indicating that they were not lysosomal in origin. Activity towards GL and SM was predominantly located in the cytosol. It is suggested that these dipeptidase activities play a role in the terminal steps of protein digestion following pinocytosis and lysosomal hydrolysis.
