1. A technique is described for the removal of subcellular contaminants from intact rat intestinal brush borders, and for the subsequent separation of a microvillus membrane fraction from a fibrillar residue. 2. Increments in invertase activity, microscopic homogeneity and low nucleic acid content indicate that the microvillus plasma membrane has been extensively purified. Multiple membrane preparations have been shown to be highly reproducible with respect to their invertase specific activity, cholesterol content and phospholipid content. Alkaline phosphatase, leucine aminopeptidase, Mg(2+)- and Ca(2+)-dependent adenosine triphosphatase and seven separate disaccharidases were shown to be predominantly confined to the membrane fraction. 3. The fibrillar fraction has been shown to contain approximately 30% of the total protein of purified brush borders, plus most of the residual nucleic acid contaminant. No evidence was found for the localization of any specific enzyme in this fraction.

The effect of unavailable carbohydrates on the intestinal absorption of calcium was studied in rats raised for 7 or 8 weeks on diets containing 10 or 20% of cellulose, glucomannan, or pullulan. The following results were obtained a) Body weight gain was diminished more severely in glucomannan groups than in cellulose groups. b) Serum calcium levels were slightly lower in all groups fed unavailable carbohydrates, whereas serum inorganic phosphorus levels were similar to that of the control group. c) There was a significant reduction of bone ash from rats fed glucommanan or cellulose with 620 glycoside bonds. d) Calcium transport measured in the everted duodenal sac remarkable decreased in the glucomannan 20% group, but slightly increased in the cellulose groups. e) Calcium binding activity was lowered significantly in all groups fed unavailable carbohydrates. f) Alkaline phosphatase and sucrase activities in the duodenum were also markedly decreased by prolonged intake of unavailable carbohydrates. These results indicate that inhibitory effect of unavailable carbohydrates on intestinal calcium absorption is partially due to the loss of calcium binding protein caused by gastrointestinal transit of large amounts of undigested substances.

We have studied changes in the activity of some lytic enzymes contained in mycelium of Aspergillus niger in cultures relative to the autolytic phase of growth. Acid phosphatase, polygalacturonidase and alpha-amylase activity reached its highest level (40.7, and 8 U/sample, respectively) at the initiation of the autolytic phase of growth. 1.3-beta-Glucanase and beta-N-acetylglucosaminidase reached its highest level (3.5 and 2 U/sample, respectively) during the first days of autolysis. Alkaline phosphatase, cellulase, invertase, esterase, chitinase and proteolytic activity is also present in autolysing mycelium of A. niger, though comparatively low. Their maximum activity coincided with the beginning of the autolytic phase of growth. In all enzymes studied here, as autolyis proceeded, enzyme activity decreased by about 90%. Only esterase activity remained nearly constant throughout the whole period of autolysis described here.

Explants of suckling mouse jejunum have been maintained in serum-free organ culture with or without insulin added to the medium in order to determine the possible direct effect of this hormone on the hydrolytic functions of the brush border and on the proliferation of the crypt cells. The addition of insulin induced the precocious appearance of sucrase activity and increased trehalase, glucoamylase and lactase activities. Alkaline phosphatase activity remained unaffected in the tissue as well as in the medium. An increased DNA content and 3H-thymidine incorporation into DNA were already recorded after 24 h of culture. The mitotic index was significantly increased after 24 h and remained elevated when the culture was extended to 48 h. These results show that insulin directly influences the enzymatic maturation and the proliferation of intestinal epithelial cells of suckling mouse.

Human small bowel early organogenesis was studied by scanning electron microscopy and found to be correlated to brush border enzymology. The appearance of the brush border enzymes sucrase, lactase, and aminopeptidase (measured in a purified apical membrane fraction) coincides with the first outgrowth of villi (eight weeks). Alkaline phosphatase was detected at seven weeks. The content of these enzymes furthermore increased up to the 14th week when both sucrase and aminopeptidase activities were comparable with adult values.

To determine which region of the intestinal villus was primarily responsible for calcium uptake and whether cells from the different regions of the villus differed in their response to 1,25-dihydroxyvitamin D3 [1,25-(OH)2D3], we studied cells eluted from the duodenal villus in a sequential fashion at various times after vitamin D-deficient chicks had received 1,25-(OH)2D3. The elution scheme employed removes cells from the villus tip first and cells from the villus base last, as was documented by the distribution of alkaline phosphatase activity, sucrase activity, and cytosolic calcium-binding protein (CaBP) in the eluted fractions. Brush border membrane vesicles (BBMV) were prepared from different fractions of the villus. Calcium uptake was greatest in BBMV from cells eluted from the villus tip and least in those from the villus base. The distribution of calcium uptake and alkaline phosphatase activity in the same BBMV were parallel. After 1,25-(OH)2D3 treatment, cytosolic CaBP was observed in the cells from the villus base by 4 h and in all fractions by 8 h; at all times (from 4-24 h), cells from the villus base contained more cytosolic CaBP than did cells from the villus tip. Alkaline phosphatase activity in BBMV was stimulated in all fractions by 4 h; at all times, alkaline phosphatase activity was greatest in BBMV from cells of the villus tip. In contrast, calcium uptake by BBMV was stimulated 2 h after 1,25-(OH)2D3 administration only in cells from the villus tip and was not stimulated even by 24 h in cells from the villus base. These results indicate that the cellular response to 1,25-(OH)2D3 depends on the location of the cell on the villus and that 1,25-(OH)2D3-stimulated calcium flux across the brush border can be dissociated from 1,25-(OH)2D3-stimulated alkaline phosphatase activity and CaBP production.

We studied the uptake of D-glucose and L-tryptophan by the small intestine and estimated the activities of the intestinal brush border enzymes (sucrase, lactase, NA+-K+-ATPase and alkaline phosphatase) and lysosomal enzymes in rats receiving T-2 toxin orally. considerable decrease occurred in glucose and tryptophan uptake and in brush border sucrase, lactase and (Na+-K+)-ATPase. Alkaline phosphatase activity and release of lysosomal enzymes (acid phosphatase and acid ribonuclease) was unchanged.

The effect of starvation and refeeding on the developmental pattern of intestinal sucrase-isomaltase (SI) was analyzed in preweaned rats. Starvation at postnatal day 12 caused a precocious expression of SI activity and mRNA. Alkaline phosphatase activity was slightly reduced, and no significant change was observed for aminopeptidase and lactase activities. Immunostaining showed that SI molecules appear in cells at the base of the villus. Sucrase expression was further increased by prolonged food deprivation, whereas enzyme activity as well as the amount of SI mRNA dropped to reach the low level found in control sucklings when 48 h-starved pups were refed by returning them to their dams. During the refeeding period, the enterocytes that were committed to produce SI by starvation continued to express the enzyme while migrating up the villi. However, the new epithelial cells arising from the crypts no longer synthesized the disaccharidase. The starvation-evoked appearance of SI was preceded by a transient burst of expression of the protooncogene c-fos, an event that may be correlated to the ontogenic rise of c-fos mRNA observed before weaning. However, in contrast to the normal weaning condition, SI induction by starvation occurred without obvious increase of epithelial cell proliferation and turnover. During the starvation and refeeding period, patterns of sucrase activity and SI mRNA paralleled the serum level of glucocorticoids.(ABSTRACT TRUNCATED AT 250 WORDS)

The extent of positional variation in mucosal enzyme activity along the small intestine was investigated in 14-day-old suckling rats. Samples were taken from ten equally spaced sites along the intestine in 11 rat pups and the activities of the enzymes alkaline phosphatase, neutral aminopeptidase, gamma-glutamyl transferase, lactase and sucrase were measured. All the enzymes except sucrase were subject to considerable positional variation. Alkaline phosphatase and aminopeptidase activities were distributed throughout the intestine, with a broad maximum in the distal intestine. Lactase was also broadly distributed but with greatest activity in the mid intestine. gamma-glutamyl transferase exhibited a novel profile, with a very high proportion of the total activity (78%) present in the distal intestine. Sucrase was essentially absent throughout the intestine.

This study has identified a naturally occurring, specific deficiency of a brush border aminopeptidase N (ApN) in the small intestines of five clinically healthy dogs. ApN activity in mucosal homogenates of dog small intestine was reduced significantly in deficient animals (13.4 (1.1) nmol min-1 mg-1 protein, n = 5, P < 0.002) compared to healthy control dogs (95.1 (6.7), n = 22). Alkaline phosphatase, gamma-glutamyl transferase, zinc-resistant alpha-glucosidase, maltase, sucrase and lactase in the ApN deficient dogs exhibited comparable activities to those in the control dogs. Microvillar membranes were analysed by one- and two-dimensional electrophoresis. ApN was represented by a single 145kDa band in all control dogs, identified by immunoblotting and immunoprecipitation. Protein maps from deficient dogs were normal apart from the virtual absence of an ApN spot and there were no apparent abnormalities in the glycosylation of microvillar proteins. The findings suggest that intestinal ApN deficiency in these dogs is a primary lesion involving diminished expression of an otherwise normal enzyme protein.
