The intestinal dipeptidase and disaccharidase activities were investigated in 120 male albino rats of the Wistar strain after administration of 21 mumol.kg-1 body weight phosalone, 14.8 mumol.kg-1 body weight lindane and 10.5 mumol.kg-1 body weight phosalone combined with 7.4 mumol.kg-1 body weight lindane. The dipeptidase activity under the effect of these comparatively low doses of pesticides reveals slightly to moderate changes. The activity of intestinal disaccharidases after a 90-day phosalone and lindane treatment is markedly decreased, particularly that of sucrase. The mechanism of these changes remains unknown.

Experiments on male rats showed that following subcutaneous injection of alloxan in a dose of 15 mg per 100 g of body weight amylolytic, invertase, glycyl-l-tyrosine dipeptidehydrolase activity of the mucosa of the small intestine altered differently. As a result, there occurred a deviation of the enzymatic spectrum from the normal characterized by a sharp increase (with the development of diabetes) of the activity of the carbohydrate hydrolysing enzymes (amylase and invertase) with a simultaneous reduction of the activity of the enzymes participating in protein (dipeptidase) and fat (monglyceridlipase) hydrolysis.

On the basis of comparative determinations of the activities of dipeptidases and disaccharidases of the mucous membrane of the small intestine (proximal jejunum) clear correspondences between the morphological findings and the biochemical parameters were the result. L-alanyl-L-prolin-dipeptidase and glycyl-L-valin-dipeptidase as well as lactase, saccharase, maltase and trehalase were determined in altogether 45 children with various malabsorption syndromes of different age in different stages of disease. Diminutions of the activity of the dipeptidases were to be proved analogously to maltase, saccharase and lactase, too, in most cases of subtotal or total villous atrophy. From the results conclusions may be derived to the restricted ability of protein absorption in chronic disease of the small intestine.

Acute experiments showed that daily 12-hr heating leads to different changes in fermentative activity (amylase, invertase, glycin-1-leycin-dipeptidase, monoglyceridiypase) in mucous membrane of the small intestine in white rats. These changes are due to not only the decrease of food consumption, but evidently are the result of functional changes in different organs and systems.

It was revealed in acute experiments that exposure to high temperature changed the invertase, dipeptidase, lipolytic and alkaline phosphatase activity of the small intestinal mucosa in rats. The direction of these changes and their degree differed in the thyroidectomized and sham-operated animals. It is suggested that the thyroid hormones took part in the reaction of the fermentative systems of the small intestine to the action of the thermal factor.

The effect of tetramethylthiuramdisulphide on the activity of the small intestine mucosa in rats was studied. When administered in different doses the compound was found to bring about, in the main, a fall of the monoglyceride-lipase and dipeptidase and a rise of the invertase alkaline-phosphatase and amylolytic activity. A protracted introduction of the compound brings down the weight of the small intestine mucosa. One month after stoppage of the drug's administration in a dose of 1/50 DL50 practised for 6 months and 15 days following its single introduction in a dose of 1/3 DL50 the enzymatic and the weight of the intestinal mucosa returned back to normal.

Six litters of newborn crossbred piglets were utilized to examine 1) the effects of substituting 20% of the protein of an all-milk protein liquid diet with a soy protein isolate (milk-soy diet) on small intestinal variables and 2) the effects of supplementing this milk-soy diet with 25 g of either putrescine or ethylamine per kilogram diet on small intestinal variables. Small intestinal xylose absorption tended to increase from wk 1 to wk 2 of age in pigs fed the milk, putrescine and ethylamine diets, but not in pigs fed the milk, putrescine and ethylamine diets, but not in pigs fed the unsupplemented milk-soy diet. Crypt depth in pigs fed the milk-soy diet tended to be less (9.4%; P greater than .10) than the crypt depth in pigs fed the other diets, but mitotic index was not different (P greater than .10) among diets. Mucosal protein, DNA and RNA concentrations and mucosal brush border sucrase and cytosolic dipeptidase activities tended to be least in pigs fed the putrescine and ethylamine diets. Concentration of mucosal putrescine was greatest (P less than .002) in the distal regions of the small intestine of pigs fed putrescine. Mucosal ornithine decarboxylase activity was inhibited by putrescine (P less than .02), but it was not affected by the soybean protein isolate used in this study. Supplementing soy protein isolate diets with amines may enhance intestinal absorption and enterocyte proliferation.

Full-value diets of similar composition were given to male rats weighing 207-230 g, by intravenous (group 1) or intragastric (group 2) routes. The proportion of amino acids, fats and carbohydrates was 9.9:15.7:74.4 (with regard to their calorific value). The diet calorific value comprised 60.6 kcal/rat/day. An average mass increase in group 1 was 2.44 +/- 0.14 g/day, in group 2 - 1.75 +/- 0.11 g/day. The protein content and activities of alpha- and gamma-amylase, invertase, maltase, and glycil-L-leucine dipeptidase were assayed in the intestinal mucosa of the proximal portion of the small intestine in group 1 rats, while a decreased alpha-amylase activity in the distal portion of the small intestine was recorded in the animals of group 2. The mass of the pancreas in the rats of group 1 and 2 was authentically lower than in the control rats which received oral feeding with natural foods. The lowest mass of the pancreas was observed in the rats of group 1. Specific activity of trypsin, lipase and RNase in the pancreatic tissues of rats in groups 1 and 2 was similar. The results of the study have evidenced a lowered function of the digestive system under conditions of artificial feeding, especially in case of intravenous nutrition.

Some intestinal enZymes were assayed which were related to: (i) Cellular proliferation, for example, aspartate carbamoyltransferase, thymidine kinase, uridine kinase, and dihydroorotase; (ii) cellular differentiation, for example, lactase, invertase, maltase, alkaline phosphatase, and dipeptidase; and (iii) lysosomes, for example, beta-glucuronidase, acid beta-galactosidase, and acid phosphatase. These enzymatic determinations can be used to distinguish the crypt from the villus during healthy or diseased states.
