The possibility that pancreatic secretory abnormalities might precede the appearance of pancreatic neoplasms and thus provide clues to early detection of this malignancy has been investigated in an animal model. Syrian golden hamsters were treated with bis-(2-oxopropyl)-N-nitrosamine on two successive weeks (2 mg/100 g body weight/week). Pancreatic secretions from treated and untreated control animals were studied at approximately monthly intervals. The animals were anesthetized, their pancreatic ducts cannulated, and basal pancreatic juice collected for 30 min. Pancreatic secretion was then stimulated by sequential intravenous injection of secretin (50 ng/100 g) and C-terminal octapeptide of cholecystokinin (4 ng/100 g) 1 hr later. Four consecutive 15-min collections of fluid were made following secretin stimulation and four additional collections after CCK administration. Each collection was examined for volume, total protein, trypsin,
chymotrypsin
, elastase, arylsulfatase, beta-D-glucuronidase,
alpha-D-glucosidase
, and leucine naphthylamidase. In addition two trypsinogen variants present in pancreatic secretions were determined. The pancreas and other organs were removed and examined histologically at the end of each experiment. Cytological atypia appeared 3 months, ductal hyperplasia 4 months, and pancreatic neoplasms 6 months after the last injection of carcinogen. Striking decreases in flow rate and output of trypsin and
chymotrypsin
were observed several months prior to the appearance of histologically recognizable pancreatic tumors. By contrast, output of beta-D-glucuronidase and
alpha-D-glucosidase
in pancreatic juice increased markedly in the last 2 months preceding the emergence of neoplasms. The diagnostic significance of these premalignant abnormalities is illustrated most dramatically in the form of ratios of lysosomal to digestive enzymes, such as beta-D-glucuronidase-trypsin or
alpha-D-glucosidase

chymotrypsin
. Highly significant increases in these ratios were observed consistently, not only in hamsters with pancreatic neoplasms, but also in animals with preneoplastic lesions (ductular hyperplasia) which preceded malignancies by about 2 months.

After twenty weeks of continuous dosing with Trichostrongylus colubriformis larvae substantial, but declining, numbers of worms had persisted in most of the lambs examined, although there were wide inter-individual variations. Mucosal lesions were found in the proximal small intestines of all the infected animals, their severity being directly related to worm burden. Representative brush border enzyme activities analysed in intestinal mucosal extracts from the same lambs showed differing responses. Alkaline phosphatase and glycyl-L-leucine dipeptidase were significantly depleted, whereas
maltase
activity was only marginally reduced, and leucine aminopeptidase activity was normal. Mucosal acetylcholinesterase activity was significantly elevated in the parasitised animals and, interestingly in view of the postulated role of this enzyme in nematode pathogenicity, the level of activity was directly correlated with individual worm burdens. chymotrypsin
activities were unaffected and the level of superoxide dismutase, an enzyme associated with the inflammatory response, was normal. There were also no consistent changes in the mucosal activities of several enzymes including lactic dehydrogenase, creatine phosphokinase, aldolase, and glutamic oxaloacetate transaminase, whose leakage from damaged or necrotic tissues has been well defined in terms of the concomitant increase in their activity in the circulation. Lambs treated orally with fenbendazole five and/or ten weeks before slaughter either in the presence or absence of continued larval intake, had negligible worm burdens, and showed little evidence of intestinal damage at post mortem. Brush border enzyme levels, with the exception of alkaline phosphatase and, in two cases dipeptidase, were normal in these animals. The activity of alkaline phosphatase was approximately double that in the continuously infected, untreated lambs, but remained markedly lower than in the uninfected controls. The activities of the other enzymes studied, including acetylcholinesterase, were within the control range. In summary, in chronic trichostrongylosis even relatively low nematode burdens were associated with marked pathological and biochemical damage in the intestine with both lesion severity and mucosal acetylcholinesterase activity being directly related to worm numbers. Although morphological integrity was completely restored after anthelmintic treatment, the persistent low activity of brush border alkaline phosphatase coupled with the enzymological findings in untreated, infected animals suggests that recovery of the full functional capability of the intestinal mucosa may take longer.

completely inhibited giant-cell development, without altering macrophage viability. No alteration of giant-cell formation was observed when 0.1 M of L-fucose, N-acetyl-glucosamine, D-arabinose, D-xylose, melibiose, D-glucose, D-galactose, alpha-lactose, sucrose, D-fructose, or maltose was present during incubation of macrophages with MFF. Giant-cell formation was abolished when 0.1 M alpha-D-mannose was present during macrophage incubation with MFF. These results suggest that the protein moiety of MFF recognizes a specific receptor site on the macrophage membrane, one that is different from those described for other lymphokines and contains alpha-mannose.

The pathology and enzymology of the intestinal mucosae of lambs dosed daily with 2500 Trichostrongylus vitrinus larvae and killed at five, nine or 14 weeks were compared with worm-free animals. The proximal small intestines of the infected lambs exhibited extensive mucosal damage at five and nine weeks, but only isolated lesions were found at 14 weeks. Activities of the brush border enzymes alkaline phosphatase, leucine amino-peptidase,
maltase
and glycyl-L-leucine dipeptidase were all significantly depleted during infection, although the magnitude, time of onset and duration of the individual enzyme responses varied. Mucosal activities of the pancreatic enzymes, trypsin and to a lesser extent
chymotrypsin
were also markedly decreased particularly during the first nine weeks of infection. Specific acetylcholinesterase activity was significantly increased throughout the study, maximal levels being observed at five weeks. In contrast 'pseudo'-cholinesterase levels were consistently within the control range. During the early stages of infection (five weeks) glutamine-oxaloacetate transaminase activity was significantly decreased, while aldolase and creatine phosphokinase levels were significantly elevated. At nine weeks low glutamine-oxaloacetate transaminase activities were again detected and lactate dehydrogenase activity was also markedly reduced. At 14 weeks the mean activities of all four enzymes were within the normal range as were superoxide dismutase levels throughout. Thirty-eight second parity sows were either immunized (IMM) against somatostatin (SRIF) and/or injected with growth hormone-releasing factor (GRF) during gestation. Treatment effects on pancreatic, gastric and duodenal development as well as on digestive enzyme activity of piglets at birth (before suckling) or at 24 h postpartum were investigated. Birth weights of piglets were similar across treatments (p > 0.1). Weight, DNA, RNA, total protein content and enzyme activity for all three organs increased between birth and 24 h postpartum (p < 0.01), except for pancreatic RNA and
chymotrypsin
which decreased (p < 0.01), and protein content of the pancreas which was unaltered (p > 0.1). Gastric RNA, pancreatic weight:DNA, RNA:DNA and amylase:DNA ratios were increased in 1-day-old piglets from SRIF-IMM sows (p < or = 0.05). GRF only had significant effects (p < 0.05) on the
maltase
:DNA ratio, which it decreased. Yet, there were tendencies (p < 0.1) for duodenal weight, DNA and total protein content to be increased in piglets from GRF-injected dams. It is therefore apparent that major developmental changes of the pancreas, stomach and duodenum of piglets take place during the first 24 h postpartum. Injections of GRF and/or immunization against SRIF during gestation in swine also have several effects on digestive enzyme activity of neonatal pigs. Yet, the physiological implications of these early changes are not clear at the present time.

An epidemiologic study of Pasteurella haemolytica serovar 1 (Ph1) in market-stressed feeder calves from 7 farms in eastern Tennessee was conducted. The nasal mucus of each calf was cultured sequentially at the farm of origin (day 0), at an auction market (day 133), and at a feedyard in Texas (days 141, 148, 155, and 169). Of the 103 calves tested, 77 were culture-positive, including 1 on day 0, 1 on day 133, 20 on day 141, 57 on day 148, 50 on day 155, and 14 on day 169. From the 143 Ph1 isolates, 20 enzyme profiles were determined by use of a commercial enzyme system that detects 19 enzymatic reactions; 4 antimicrobial susceptibility profiles were obtained, using the disk-diffusion method, which evaluated susceptibility to 11 antibacterial drugs. All isolates were positive for acid phosphatase and alkaline phosphatase, but were negative for alpha-galactosidase, alpha-mannosidase, beta-glucosidase, beta-glucuronidase, cystine aminopeptidase, N-acetyl-beta-glucosaminidase, and trypsin. Other positive enzyme reactions included: leucine aminopeptidase, 140 Ph1 isolates; phosphohydrolase, 90 isolates; alpha-fucosidase, 63 isolates; esterase (C4), 59 isolates; valine aminopeptidase, 30 isolates; esterase lipase (C8), 24 isolates; beta-galactosidase, 2 isolates; and
alpha-glucosidase
,
chymotrypsin
and lipase (C14), 1 isolate each. Thirty-four Ph1 profiles were identified, using combined enzyme and antimicrobial susceptibility profiles. The enzymatic activity of 70 feline and canine Microsporum canis isolates was determined by the Api-Zym test. The liquid phase of cultures, inoculated into Tryptic Soy Broth, was used to examine 19 enzymes. Considerable differences were observed among the extracellular enzymatic patterns. All the isolates produced alkaline phosphatase and beta-glucosidase, while lipase (C14), trypsin,
chymotrypsin
, beta-glucuronidase, and alpha-fucosidase activity was never revealed. The effect of supplementing a cornsoybean diet (C) with glucose (G) or maltose (M) on young broilers (from hatch to 3 wk of age) affected by stunting syndrome (SS) was studied. Stunting syndrome was induced by orally administering an inoculum prepared from the intestines of SS broiler chicks. Relative to the M diet, the G diet improved growth and feed utilization and increased feed intake in naive (NA) control chickens. The C diet was intermediate in this respect. In contrast to the NA chickens, diet did not affect growth or feed utilization in SS chicks. Changes in the relative weights of the gastrointestinal tract segments were evident by 1 wk of age and hypertrophy of these segments persevered to 3 wk of age. Stunting syndrome infection was accompanied by a significant increase in pancreatic trypsin-specific activity during Weeks 1 and 2, and in
chymotrypsin
activity at 1 wk. During this time, amylase-specific activity was not affected. At 3 wk of age, the specific activities of amylase, trypsin, and
chymotrypsin
in the pancreas were lower in the inoculated vs control birds. Whereas no significant effect of SS was observed with activities of amylase in the intestinal contents, trypsin activity was higher in SS chicks at 1 wk, and that of
chymotrypsin
lower during Weeks 2 and 3. Relative to NA chicks, the
maltase
and saccharase activities of SS chicks were much lower during Week 1, but increased later on and were similar to NA chick values at 2 and 3 wk. Whereas the level of blood plasma proteins did not vary from 1 to 3 wk in the NA chicks, it increased gradually in SS chicks to a level that significantly exceeded that in their NA counterparts. Blood plasma glucose and triglyceride levels were slightly lower in the SS chicks (NS), and the blood plasma cholesterol level was significantly reduced during Week 2. Relative to NA chicks, SS infection caused a significant increase in plasma calcium during Weeks 2 and 3, accompanied by a significant reduction in blood plasma phosphorus at 2 wk only. No difference was observed in the blood plasma level of uric acid, which peaked in both treatments during Week 2, or in D-beta-hydroxybutyric acid level, which was quite stable during the experimental period. Stunting syndrome infection was accompanied by a dramatic increase in amylase and alkaline phosphatase activities in the blood plasma, and by a slight but significant decrease in activity of lactic dehydrogenase. Stunting syndrome was concluded to be an affliction not only of digestion but also of metabolism. The main depression in growth caused by SS inoculation is probably due to metabolic alterations beyond those of digestion and absorption.

Responses to stunting syndrome (SS) infective material obtained from affected broilers and administered per os were monitored for 3 wk in a fast-growing commercial broiler population, in slow-growing Leghorn chicks, and in turkey poults. At 2 and 3 wk, the size of the gastrointestinal tract (GIT) segments, the pH of the GIT contents, and the activities of digestive enzymes in the intestinal contents and of disaccharidases on the jejunum mucosae were determined. Inoculation affected the genetic stocks differently. In broiler chicks, growth and feed utilization were markedly reduced. In contrast, inoculation of Leghorns was accompanied by improved feed intake and growth rate. Performance of poults was affected only slightly, albeit significantly. The effect of inoculation on the pH of crop and intestinal contents in Leghorn chicks was opposite to that found in broiler chicks, i.e., a significant increase in the crop and small intestinal pH in the former vs a significant decrease in inoculated broilers. Although inoculation of the broiler chicks did not affect the pH in the proventriculus, in Leghorn chicks it was reduced by 25%. In poults, inoculation did not significantly affect GIT contents pH. The GIT segments were markedly enlarged in broiler chicks, whereas in Leghorn chicks the opposite trend was observed; namely, intestinal segment weights were significantly reduced. In poults, inoculation caused a reduction in the intestinal segments and gizzard weight at 3 wk. During this same period, the liver and pancreas relative weights were dramatically increased in broiler chicks. A higher relative heart weight at 2 wk was observed in broilers and poults; this trend persisted to Week 3 in poults but not in broiler chicks. In broiler chicks, a nonsignificant reduction was observed for all enzymes assayed at 3 wk and for
chymotrypsin
at 2 wk. In Leghorn chicks, inoculation was accompanied by a marked and significant increase in the activity of
chymotrypsin
during both periods. In poults, inoculation caused a marked increase in the activities of amylase during Week 2 and 3, and trypsin at 3 wk. Maltase and saccharase activities in the jejunum of broiler chicks were slightly depressed a t 2 and 3 wk, the depression being significant at 2 wk for
maltase
and at 3 wk for saccharase. In the Leghorn chicks, inoculation caused a twofold increase in the activities of both enzymes. As in Leghorns, inoculation of poults with SS infective material caused a marked increase in the activities of the disaccharidases. The different responses to SS inoculation in the different genetic stocks are discussed.
