1. A total of 320 1-d-old Arbor Acres broiler chicks were used to investigate the effect of Cu(2+)-loaded montmorillonite (CM) on the growth performance, intestinal morphology and activities of brush border enzyme in the intestinal mucosa and digestive enzyme in the intestinal digesta of broilers. 2. The chicks were assigned randomly into 4 groups with 80 chicks per treatment. The 4 dietary treatments were: basal diet only (control group), basal diet + 2 g montmorillonite/kg, basal diet + 1 g CM/kg, and basal diet + 2 g CM/kg. The chicks were raised in cages and feed and water were provided ad libitum for a period of 42 d. 3. The addition of CM to the diet of broilers significantly increased body weight and feed efficiency. Similarly, birds receiving montmorillonite had higher feed efficiency than the control after 42 d of feeding. 4. Data on villus height and crypt depth for duodenum, jejunum and ileum indicated that treating the diet of broilers with either CM or montmorillonite improved the mucosal morphology of the small intestine. 5. The presence of CM in the diet of broilers significantly increased the activities of
maltase

The effects of supplementing a barley-based diet for weaned piglets withexogenous beta-glucanase and xylanase on gastrointestinal digestiveenzyme activities were investigated. Thirty-six cross-bred weaned pigletswere randomly assigned to two groups with three pens based on sexand mass. Each group was fed on the diet based on barley with or withoutadded beta-glucanase and xylanase (0.15%) for a 4-week period. Theresults showed that enzyme supplementation improved growth performanceof piglets significantly (p < 0.05), but had no effect (p = 0.091)on average daily feed intake. The results also showed that supplementationof beta-glucanase and xylanase had no effect on pepsin activity in gastriccontents but slightly decreased (p = 0.092) the pepsin activity ingastric mucosa. Meanwhile, no effect of enzyme supplementation ontrypsin activity in duodenal contents was observed. However, the activitiesof amylase and

in duodenal contents were significantly(p < 0.05) decreased, whereas the activities of
maltase
, sucrase andgamma-glutamyl transpeptidase (gamma-GT) in jejunal and ileal mucosa wereenhanced significantly (p < 0.05). Lipids extracted from tsao-ko were separated into three fractions with silica gel column chromatography and fed to mice (3 mo old) for 90 d to clarify their inhibitory activity on digestive enzyme activity. The diets contained the following: control--no tsao-ko, 0.05% total lipid of tsao-ko (TL), 0.0109% chloroform fraction (CF), 0.0245% acetone fraction (AF), or 0.00365% methanol fraction (MeF). Although CF and AF slightly inhibited the activities of
alpha-glucosidase
, alpha-amylase, and
lipase
, intakes of these fractions had little influence on plasma and liver lipid concentrations when compared with the control diet. MeF did not inhibit
alpha-glucosidase
but had DPPH radical scavenging activity and the mice fed this fraction had the most marked reduction in plasma glucose and TBARS concentrations compared with the other diet groups. These results suggest that the fat-soluble polar components of tsao-ko contain an active component that might be associated with decreased plasma glucose and TBARS concentrations in mice.

Two luminous marine bacterial strains, LC2-005(T) and LC2-102, were isolated from seawater at Kuroshio Region and Sagami Bay in Japan, respectively. These bacteria were Gram-negative, oxidase-positive, catalase-positive, motile and rod-shaped. On the basis of 16S rRNA gene sequence analysis, strains LC2-005(T) and LC2-102 formed a cluster within the Vibrio harveyi species group. However, multilocus sequence analysis using five loci (pyrH, ftsZ, mreB, gyrB and gapA) and DNA-DNA hybridization experiments indicated that these strains were distinct from the currently known Vibrio species. Additionally, these strains differ from related Vibrio species in utilization of glucose, mannitol, inositol, sorbitol, rhamnose, sucrose, melibiose and arabinose, production of lysine decarboxylase, ornithine decarboxylase, tryptophan deaminase, esterase (C4),

(C4), chymotrypsin, acid phosphatase,
alpha-glucosidase
, beta-glucosidase and N-acetyl-beta-glucosaminidase and the ability to reduce nitrate to nitrite. The major fatty acids were C(15 : 0) iso 2-OH and/or C(16 : 1)omega7c, C(16 : 0), C(18 : 1)omega7c and C(14 : 0). The DNA G+C contents of strains LC2-005(T) and LC2-102 were 45.2 and 45.5 mol%, respectively. On the basis of the polyphasic taxonomic evidence presented in this study, it can be concluded that strains LC2-005(T) and LC2-102 belong to the same genospecies and represent a novel species of the genus Vibrio, for which the name Vibrio azureus sp. nov. is proposed. Pathogenicity of fungi is connected with their ability to easily penetrate the host tissues, survive in the infected host organism and use the elements of the host tissues as nutrients. Hence, the co-occurrence of pathogenic properties with the high enzymatic activity, which is manifested through the production of various enzymes including extracellular enzymes, was observed. It can be expected that it is possible to decrease fungal pathogenicity by lowering their enzymatic activity. The aim of the study was to determine the effect of nicotinamide on enzymatic activity of the fungi, which are most frequently isolated in cases of skin infection. Enzymatic activity was analysed using 15 Candida albicans, 15 Trichophyton rubrum and 15 Trichophyton mentagrophytes strains. The strains used for the study were collected from the current diagnostic material. API ZYM tests were used in diagnostic analysis. MICs of nicotinamide were determined by the macrodilution method in liquid medium. In the case of Candida strains, the presence of nicotinamide in the broth had a significant effect on the decrease of enzymatic activity (P < 0.05) of esterase (C4), esterase
lipase
(C-8), valin-arylamidase, acid phosphatase and alpha-glycosydase. A considerably stronger effect of nicotinamide was observed in the case of dermatophytes (P < 0.005). Its action led to a decrease in the activity of all the enzymes under study except
alpha-glucosidase
produced by T. rubrum strains. Thus, nicotinamide exhibited biological activity towards C. albicans, T. rubrum and Trichophyton mentagrophytes, which resulted in a decrease in the activity of enzymes produced by the fungi.

The carbohydrases (

(tributyrinase) of the Welch bacillus retain their hydrolytic activities in sterile solutions of the bacterial cells. The demonstration of the carbohydrases and detection of their hexose products constitute experimental proof that Welch bacilli attack complex carbohydrates by way of a preliminary hydrolysis. The result of experiments on the oxidation of the enzymes show that the inhibitory effect of air upon the initiation of growth of the living Welch bacillus does not include a comparable effect upon the activity of its hydrolyzing enzymes.

The study of bacterial communities in microbially-mediated water treatment systems is becoming increasingly popular. Aquatic bacterial communities are often found in fixed-film environments, residing within a matrix of extracellular polymeric substances commonly referred to as a biofilm. A method for detaching the biofilm is required to either enumerate or characterize these bacterial communities. There are a variety of detachment methods including scraping, swabbing, shaking, sonication, blending, and digestion. The objective of this work was to develop an agitation-based protocol for detachment of culturable bacterial communities from the biofilm surrounding pea gravel from constructed wetland mesocosms. Three different protocol factors were systematically investigated using a triplicated 2(3) factorial design to determine the most effective detachment protocol. Factors studied included: the use of either tap water or phosphate buffer as the shaking/detachment solution; the use of either manual-shaking at room temperature or mechanical shaking at 30 degrees C; and the presence or absence of an enzyme cocktail consisting of
lipase
, beta-galactosidase and
alpha-glucosidase
. The resulting suspensions were evaluated for organics, inorganics, culturable bacteria, community level physiological profile (CLPP) and several BIOLOG ECO plate substrate related diversity indices. Using these metrics, the most effective shaking/detachment protocol was identified as mechanical shaking for 3h at 30 degrees C using a phosphate buffer with an enzyme cocktail.

The purpose of this study was to evaluate the effect of high hydrostatic pressure (HHP) on the enzyme activities in Saccharomyces cerevisiae (ATCC 16664) and Escherichia coli (ATCC 11229). Enzyme activities before and after HHP treatment were determined using an APIZYME enzyme assay kit. Thirteen active enzymes were detected in S. cerevisiae and E. coli. Pressure treatment at 448 MPa for 30s at 23 degrees C resulted in different effects on enzymes in S. cerevisiae and E. coli. HHP completely inactivated

Our previous study showed that tsao-ko intake can lower plasma and liver triacylglycerol (TG) concentrations and has hypoglycemic and antioxidant activity in mice. This study involved separating two major fractions (A and B) from the methanol extracts (MeX) of tsao-ko using silica gel column chromatography, and then determining the effect of the fractions in vivo and in vitro to clarify the most effective components of tsao-ko. An intake of MeX and A fraction statistically significantly reduced body lipids and plasma thiobarbitutic acid reactive substances (TBARS) concentrations compared with the control and inhibited
lipase
and
alpha-glucosidase
activities. These reductions were not observed in mice fed the B fraction and these inhibitions of B fraction were mild compared with MeX and A fraction. The plasma and liver TG concentrations of each fraction group did not show significant differences compared with the control. The [M-H](+) and maximum UV absorption of the A fraction were 291 m/z and 279 nm, respectively. The peak of A fraction appeared at a similar time to the epicatechin standard in the LC/MS/MS analysis and the MS/MS spectrum of the A fraction was similar to that of the epicatechin standard. It was concluded that the most effective component of tsao-ko for body lipid reduction and hypoglycemic and antioxidant activity was contained in the polar fraction and the evidence suggested that this component could be epicatechin. However, the strongest TG lowering components of tsao-ko may be methanol insoluble.
