The purpose of the trial was to establish the effect of the injection of the lysozyme dimer or vitamins connected with Se on the activity of chosen antioxidant enzymes and the total antioxidant status in pregnant heifers. Examinations were carried out during winter season in one farm on 21 heifers aged 22-24 months. Between the 21st and 14st day before expected parturition, seven heifers were once i.m. injected with antioxidants (Vitamin A-600 000 i.u.; Vitamin D3-200 000 i.u.; Vitamin E-1.5 mg/kg b.w., Selenium-0.022 mg/kg b.w.), and the next seven animals with lysozyme dimer (Lydium-KLP) at a dose of 0.02 mg/kg b.w. versus 7 non-treated control animals. Blood samples were taken before injection and then in hour 24 and 72 after injection, and between, the 7th and 14th day after calving. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSHpx), glutathione reductase (GSHred) and total antioxidant status (TAS) were measured by colorimetric method with the use of Randox kits. The mean value of SOD activity 21-14 days before expected calving was 704.8 +/- 294.6 U/ml of whole blood, GSHpx 59222 +/- 23699 U/l of whole blood, GSHred 110.8 +/- 22.5 U/l and TAS 0.33 +/- 0.15 mmol/l of serum. These indicators did not change in the control group with the exception of a statistically insignificant decrease in SOD activity after parturition. Statistically significant increase in blood SOD activity was noted only in the first day after injection of vitamins combined with selenium. These antioxidants also caused an insignificant increase in blood GSHpx activity in 72 hour following the injection, and in the second week after calving (statistically significant). The injection of antioxidants or lysozyme dimer did not change the activity of blood GSHred. However, an increase in the TAS was found in hour 24 (non significant) and 72 (statistically significant) following the single injection of lysozyme dimer.

The effect of KLP-602 (active substance: lysozyme dimer) on the replication of two animal viruses: the TK900 strain of Aujeszky's disease virus and the Roakin strain of the Newcastle disease virus were investigated. The maximal tolerable dose of the drug was determined for two cell cultures (CECC and GMK) and the effect of the medicine on the titre range of infectious viruses and their adsorption was assayed. The direct impact of KLP-602 on the viral strains used was also determined. And finally the replication dynamics of viruses in the presence of KLP-602 preparation was estimated. KLP-602 showed no direct effect on either the viruses applied in the study or their adsorption. The drug, introduced into the culture 24 hours before its infection, did not affect the replication of the pseudorabies virus, but decreased the titre of the Newcastle disease virus. KLP-602 introduced simultaneously with the infection considerably lowered the final titres of both viruses. The medicine had the greatest inhibitory effect on the replication dynamics of both types of viruses in the CECC and of the pseudorabies virus in the GMK culture upon the maximal tolerable concentrations of drug and low infectious doses of viruses applied.

Immunomodulation is a commonly used method of prophylaxis in humans and animals. Lysozyme dimer (KLP-602) was used at a dose of 50 ug/kg b.w. in order to correct the immunosuppression caused by the action of herbicide glyphosate (Roundup- Monsanto), which was used in a single bath for 10 minutes in a concentration of 100 mg/l of water. The investigations were carried out on 2 species of fish: the carp (Cyprinus carpio L.) and european catfish (Silurus glanis L.). Herbicide glyphosate caused a decrease in metabolic and phagocytic activity (RBA and PKA) and in proliferative response stimulated by Con A and LPS in carp and european catfish. The immunosuppression sustained for about 2 weeks. The results obtained indicate the possibility of correction of immunosuppression applying lysozyme dimmer (KLP-602) after use of which, the level of the studied indexes increased.

In rats exposed for 28 days (5 hours a day) to ozone at a concentration of 0.5 ppm and receiving alpha-tocopherol at doses of 4.5 mg/rat and 15 mg/rat, levels of acute phase proteins (APP)--C-reactive protein (CRP), ceruloplasmin (Cp), total protein, gamma-globulins, and activity of lysozyme in blood serum were studied. The assays were performed in the presence of respective control groups, i.e. rats receiving the same doses of alpha-tocopherol but not exposed to ozone, a group of animals not supplemented with vitamin but exposed to ozone, a group of animals injected with physiological fluid and a control group not subjected to any of the treatments. The study revealed that the ozone-exposed animals had an increased lysozyme activity and a decreased total protein level. However, in rats protected by alpha-tocopherol and exposed to ozone, the concentration of APP, lysozyme activity and total protein were found to be decreased. Similar relationships also occurred in animals receiving alpha-tocopherol and not exposed to ozone.

Numerous granular eosinophilic cells corresponding to Paneth cells were unexpectedly revealed in a moderately differentiated adenocarcinoma of the hepatic flexure of the colon in a 76-year-old man. These cells were clearly seen in routine slides and their nature was confirmed by Masson's trichrome stain, by immunohistochemical reaction for lysozyme, and by electron microscopy. The tumor contained also cells intermediate between Paneth and goblet cells (with both supranuclear granules and mucous globules), observed in Masson's trichrome stain and in electron microscope. Additionally, immunohistochemical reaction for chromogranin A was performed and irregularly scattered neoplastic endocrine cells were visualized in the tumor. The light microscopic and ultrastructural features of this rare neoplasm are described together with detailed clinical data. The incidence and significance of the Paneth cell and endocrine differentiation in colorectal carcinomas are discussed with the review of the literature.

Successful results of earlier studies on testing the immune-modulating and anti-microbial properties of 1,2,4-triasole derivative as well as benefitial application of natural bio-stimulators in animal's prophylaxis and treatment inspired us to undertake further investigations. The aim of the present study was to evaluate and compare the prophylaxis effects of two natural immune-modulating agents, garlic (Allium sativum) extract and Echinovit C preparation and synthetic derivative of 1,2,4-triasole on non-specific immunity indices in slaughter turkey-hens. The results obtained suggest that the strongest stimulatory effect is exerted by garlic followed by 1,2,4-triasole derivative and Echinacea juice. Immune-modulating action of the substances applied was manifested by an increase in cellular immune parameters: the percentage of phagocyting cells, NBT-positive granulocytes and lysozyme level.

Bacteria from genera Chlamydia (Ch.) and Chlamydophila (Chl.) are very pathogenic and may infect humans and animals. They also may cause latent infection, especially in animals. In this paper we discuss the non-specific and specific cellular and humoral immunity in farm animals, after infection or immunisation with Chlamydia sp. and Chlamydophila sp. bacteria. It has been shown, that the infection or immunisation with the microorganisms influenced the activity of polimorphonuclear cells (PMN) and mononuclear cells (MN) in the process of phagocytosis. It has also been shown that the bacteria influenced the amount and activity of lysozyme, activities of myeloperoxidase and lysosomal enzymes. Infection or immunisation with the microorganisms was demonstrated to affect numbers of lymphocytes T and B and those of their subpopulation as well as the activity of cytokines and levels of serum and secreted immunoglobulins. The changes were detected just a few hours after infection or immunisation and persisted for a few days to a few decades.

Cells of a mutant of Listeria monocytogenes lacking functional PBP5, an enzyme with DD-carboxypeptidase activity, make thicker cells walls. In this study we show that the muropeptide profile of the mutant, obtained after HPLC analysis of a muramidase digest of cell wall murein, differs from that for the wild type strain. The main differences embrace strongly reduced disaccharide-tripeptide content, strongly increased amounts of pentapeptide-containing muropeptides and a shift in profile from less cross-linked muropeptides (monomers, dimers) towards more highly cross-linked ones.

NaCl-induced aggregation of hen egg white lysozyme (HEWL) was monitored by NMR spectroscopy. Small, but significant, changes induced by salt addition in TOCSY spectra were attributed to the effect of local reorganization of protein backbone upon ion binding. Salt-induced variations in HN and H alpha chemical shifts were mapped on the HEWL 3D structure which allowed the construction of a scheme of the spatial localization of potential ion binding sites. It was found that in a 0.5 M NaCl solution six chloride anions and at least one sodium cation are bound to preferred sites on the HEWL surface.

The role of protein kinase A (PKA) in the humoral immune response of the greater wax moth Galleria mellonella larvae to live gram-positive bacteria Micrococcus lysodeikticus and gram-negative bacteria Escherichia coli was investigated. The immune challenge of larvae with both kinds of bacteria caused an increase in fat body PKA activity depending on the injected bacteria. Gram-positive M. lysodeikticus was a much better inducer of the enzyme activity than gram-negative E. coli. The PKA activity was increased about 2.5-fold and 1.5-fold, after M. lysodeikticus and E. coli injection, respectively. The in vivo inhibition of the enzyme activity by a cell permeable selective PKA inhibitor, Rp-8-Br-cAMPS, was correlated with considerable changes of fat body lysozyme content and hemolymph antimicrobial activity in bacteria-challenged insects. The kinetics of changes were different and dependent on the bacteria used for the immune challenge of G. mellonella larvae.
