The binding isotherms of native bovine serum albumin with cationic detergents, such as octyl, decyl, dodecyl and tetradecylpyridinium bromides were determined at pH 6.8 and 3.4 at 25 degrees C. The isotherms for dodecyl and tetradecylpyridinium bromides were also determined at 3 degrees C. The average number of detergent cations bound increased with increasing hydrocarbon chain length. At low detergent concentration the binding of all alkylpyridinium bromides was smaller at pH 3.4 than at pH 6.8. Dodecylpyridinium bromide was bound to native beta-lactoglobulin, aldolase, ovalbumin, haemoglobin, myoglobin,
lysozyme
, trypsin and ribonuclease at pH 6.8. No binding occurred to alpha-chymotrypsin and chymotrypsinogen. The free enthalpy change, --delta G degrees, calculated from intrinsic association constants K was determined.

Fourier transform infrared and laser Raman spectroscopies were used to study the effects of dodecylpyridinium bromide on the conformation of haemoglobin, myoglobin, bovine serum albumin, ribonuclease, ovalbumin,

, trypsin and beta-lactoglobulin in aqueous solution. Addition of the cationic detergent caused a decrease in alpha-helix conformation in highly helical proteins. At low detergent concentrations stabilization of beta-sheet conformation was observed.

In 18 patients with bone marrow aplasia with pancytopenia

activity and unsaturated vitamin B12-binding capacity in the serum were determined. These investigations, together with determinations of peripheral blood polymorphonuclear count, were done again after prednisone administration. In all cases a significant fall was found in the NZW vit B12 and LZM activity in the serum. A slight rise in the polymorphonuclear count in the 24th hour of the study was associated with a rise in the NZW wit. B12 in the serum, and decreased LZM activity. This confirmed the previously demonstrated complex character of corticosteroid action on the system of polymorphonuclears. These results point also to the usefulness of determination of unsaturated vitamin B12-binding capacity for evaluating the value of the total granulocyte pool in granulocytopenia.

Two different molecular forms of bacteriolytic endo-N-acetylmuramidase (
EC 3.2.1.17
) were found in the Acanthamoeba castellanii culture. The enzymes were separated by column chromatography on DEAE-Sephadex or by polyacrylamide-gel electrophoresis. In young cultures only one molecular form of the enzyme was selectively secreted by amoeba cells to the environment. The second, more basic enzyme protein was preferentially synthesized by aging Acanthamoeba castellanii cells, and was also liberated to the medium. In amoeba cells the bulk of forms A and B of endo-N-acetylmuramidase are associated with subcellular organelles--lysosomes.

The activity of
lysozyme
and unsaturated binding capacity of vitamin B12 in the serum were determined in hydrocortisone test for calculating the reserve of granulocytes in bone marrow. A significant rise in the count of neutrophils in peripheral blood 3 hours after the beginning of the test was not associated with statistically significant changes in the determined parameters. This in an evidence that prolongation of the survival of cells in the circulation as one of the mechanisms of corticosteriod action on the granulocyte system is without any greater importance. This is connected, most probably, with short duration of the test. The rise of neutrophil count in peripheral blood after hydrocortisone administration may be regarded, therefore, as a measure of the value of one marrow reserve of these cells.

It is a general opinion that the renal transplant patients may show the decrease of immunity which manifests by their increased susceptibility to bacterial, viral and fungal infections. One of the important non-specific defensive factors of the organism participating in the protection against infections is

. It is the purpose of the present paper to estimate the
lysozyme
activity in the un- and diluted serum in the renal transplant patients and to determine the possible mechanism of the observed deviations. 32 renal transplant patients (24 men and 8 women) aged of 16 up to 50 were investigated, being in the period of 1 until 4 years after transplantation. In the investigation the kind of immunosuppressive treatment used was considered which served the purpose of dividing the total group of patients into two subgroups: one treated with cyclosporine (12 patients) and the other treated with azathioprine (20 patients). The control group consisted of 20 healthy persons, matching by age, sex and living place the group investigated. In both groups investigated and control, were determined:

activity in the serum of the undiluted and diluted blood, total leukocyte number, the absolute number of peripheral blood neutrophils as well as their adherence to fibres was evaluated. In the renal transplant patients compared with the control group and in the separated subgroups, no statistically significant differences were found concerning leukocyte count, absolute neutrophil count and adherence of those cells.(ABSTRACT TRUNCATED AT 250 WORDS)

Immunological studies were carried out in 85 male smokers smoking 15-25 cigarettes daily for 2-25 years, and in 49 non-smokers. Cigarette smoking for a period longer than 10 years caused a fall of IgA, IgG, IgM and

concentrations. On the other hand, the levels of C3c and C4 components of complement, alpha 1-acid glycoprotein, ceruloplasmin, haptoglobin and antistreptolysin O were normal. Impairment of immunity to infections and neoplasms in cigarette smokers may be related to deficiency of various proteins responsible for normal course of immune processes of the organism.
