The present study evaluated the potential of archaesomes, prepared from the total polar lipids extracted from Methanobrevibacter smithii, as adjuvants for combination (multivalent) vaccines. Groups of Balb/c mice were immunized subcutaneously at day 0 and 21 with one of the following vaccines: trivalent vaccine formulated by the simultaneous co-encapsulation of bovine serum albumine (BSA), ovalbumin (OVA) and hen egg lysozyme (HEL) into archaeosomes (CEC vaccine); an univalent archaeosome vaccine (UVE vaccine) containing either BSA, OVA or HEL; or an admixture vaccine (AMC vaccine) consisting of the three UVE vaccines. Serum specific antibody (IgG + M) responses were determined at day 32, 112 and 203, and specific IgG1 and IgG2a responses were determined at day 112. Mice immunized with the CEC of AMC vaccine developed strong and sustained specific antibody responses to all three antigens at a magnitude similar to those seen in control mice immunized with UVE vaccines. Moreover, the serum BSA-, OVA-, and HEL-specific IgG1 and IgG2a levels in the CEC and AMC immunized mice were overall comparable to those of the UVE immunized control mice. Boosting CEC and AMC vaccinated mice with antigens alone at day 203 elicited strong antibody memory responses, comparable to those in the UVE vaccinated groups. These results show that archaeosomes could be used as adjuvants in developing combination vaccines.

The separation of four proteins including RNase A, cytochrome C, lysozyme and myoglobin was investigated by reversed-phase gradient pressurized capillary electrochromatography (p-CEC) with 1.5 microm non-porous silica C18 stationary phase. This mode was compared with micro-high performance liquid chromatography (mu-HPLC) and the effects of applied voltage, stationary phase and concentration of ion-pairing agent (trifluoroacetic acid, TFA) on the gradient p-CEC were also studied. This separation was performed rapidly on a new CEC instrument Trisep 2010 GV. The results showed that the retention mechanism of proteins in p-CEC mode is based on both chromatographic partitioning and electrophoretic migration. The results also demonstrated that p-CEC may have great potential for fast and efficient separation of proteins.

Novel polyelectrolyte multilayer (PEM) coatings for enhanced protein separations in open tubular CEC (OT-CEC) are reported. Use of four cationic polymers (poly-L-lysine, poly-L-ornithine, poly-L-lysine-serine, and poly-L-glutamic acid-lysine), and three anionic molecular micelles, sodium poly(N-undecanoyl-L-leucyl-alaninate) (poly-L-SULA), sodium poly(N-undecanoyl-L-leucyl-valinate) (poly-L-SULV), and sodium poly(undecylenic sulfate) (poly-SUS) were investigated in PEM coatings for protein separations. The simultaneous effects of cationic polymer concentration, number of bilayers, temperature, applied voltage, and pH of the BGE on the separation of four basic proteins (alpha-chymotrypsinogen A, lysozyme, ribonuclease A, and cytochrome c) were analyzed using a Box Behnken experimental design. The influence of NaCl on the run-to-run reproducibility was investigated for PEM coatings containing each cationic polymer. All coatings exhibited excellent reproducibilities with a %RSD of the EOF less than 1% in the presence of NaCl. Optimal conditions were dependent on both the cationic and anionic polymers used in the PEM coatings. Poly-L-glutamic acid-lysine produced the highest resolution and longest migration time. The use of molecular micelles to form PEM coatings resulted in better separations than single cationic coatings. Chiral poly-L-SULA and poly-L-SULV resulted in higher protein resolutions as compared to the achiral, poly-SUS. Furthermore, the use of poly-L-SULV reversed the elution order of lysozyme and cytochrome c when compared to poly-L-SULA and poly-SUS.
