Utilization of natural immunostimulants in fish culture offers a wide range of attractive methods for inducing and building protection against diseases. Lysozyme is an enzyme with bacteriolytic properties and is ubiquitous in its distribution among living organisms. This enzyme has antiviral, antibacterial and anti-inflammatory properties. In nature,
lysozyme
is found as a monomer. Lysozyme dimer is significantly less toxic than its monomer, and its high biological activity has been ascertained in cases of both viral and bacterial infections. In our study, we examined the influence of dimerized
lysozyme
(KLP-602) on the nonspecific cellular and humoral defense mechanisms and protection against furunculosis in rainbow trout (Oncorhynchus mykiss). We have analyzed the immunomodulatory effects of
KLP
-602 after experimental infection by Aeromonas salmonicida. Application of dimerized
lysozyme
(KLP-602) by injection stimulated the cellular and humoral defense mechanisms and provided protection against furunculosis. By contrast, mortality rate was reduced to 45% (one injection) and 25% (three injections) using 10 or 100 microg/kg
KLP
-602. Mortality in the untreated control group was 85%.

This study examined the effects of the dimerized
lysozyme
(
KLP
-602) on the immunocompetence cell activity in sheatfish (Silurus glanis) and its influence in vivo on the non-specific defence mechanisms and protection against motile aeromonad septicaemia (MAS). The in vitro study showed that the
lysozyme
dimer (
KLP
-602), at concentrations between 5 and 50 micrograms/mL of medium significantly (P < 0.05) increased the respiratory burst activity and potential killing activity of pronephric macrophages, as well as the proliferative ability of pronephric lymphocytes stimulated by ConA and LPS. The in vivo study showed that injecting
lysozyme
dimer (Lydium-
KLP
) intraperitoneally at doses of 50 micrograms/kg bw stimulated cell-mediated and humoral-mediated imunity. On day 5, after application of Lydium-
KLP
in vivo, a statistically higher (P < 0.05) respiratory burst activity and potential killing activity of blood and pronephros phagocytes were observed. A higher proliferative ability of blood and pronephros lymphocytes stimulated by Concanavaline A (ConA) or lipopolysaccharide (LPS) was also observed. At the same time, the myeloperoxidase activity in the PMN cells and the
lysozyme
activity and total Ig levels in serum were significantly higher (P < 0.05), compared to the control group. A challenge test with Aeromonas hydrophila showed that dimerized
lysozyme
increased the protection against MAS. Dimerized
lysozyme
stimulates non-specific cellular and humoral mechanisms and protection against MAS in sheatfish.

The immunostimulating effect of Methisoprinol and
KLP
-602 on the development of organs of the immunological system, the reactivity of lymphocytes and selected biochemical parameters of allantoic fluid (the activity of ceruloplasmin and
lysozyme
) during embryogenesis was tested. A positive effect of these drugs, especially
KLP
-602, was found.
KLP
-602 positively affected the reactivity of lymphocytes and the development of immunocompetent organs of chicken embryos as well as it increased the activity of ceruloplasmin in allantoic fluid 6 hours after injection.

The purpose of this study was to determine the possibility of modulation of the cellular immune response with the dimer of
lysozyme
KLP
-602 in fish after suppression induced by atrazine. In the in vivo studies fish were intoxicated with atrazine at a dose of 1/4 LD50, then after 48 hours the dimer of
lysozyme
KLP
-602 was administerd at a dose of 0.02 mg/kg of body weight. After 2, 3, 5, 7 and 10 days after intoxication cells from the blood and pronephros of carp were isolated. In the study we determined the metabolic activity of phagocyte cells and the proliferative ability of T and B lymphocytes. This study showed that administration of the dimer of
lysozyme
KLP
-602 stimulated and corrected the handicapped immunity.

The purpose of the trial was to establish the effect of the injection of the
lysozyme
dimer or vitamins connected with Se on the activity of chosen antioxidant enzymes and the total antioxidant status in pregnant heifers. Examinations were carried out during winter season in one farm on 21 heifers aged 22-24 months. Between the 21st and 14st day before expected parturition, seven heifers were once i.m. injected with antioxidants (Vitamin A-600 000 i.u.; Vitamin D3-200 000 i.u.; Vitamin E-1.5 mg/kg b.w., Selenium-0.022 mg/kg b.w.), and the next seven animals with
lysozyme
dimer (Lydium-
KLP
) at a dose of 0.02 mg/kg b.w. versus 7 non-treated control animals. Blood samples were taken before injection and then in hour 24 and 72 after injection, and between, the 7th and 14th day after calving. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSHpx), glutathione reductase (GSHred) and total antioxidant status (TAS) were measured by colorimetric method with the use of Randox kits. The mean value of SOD activity 21-14 days before expected calving was 704.8 +/- 294.6 U/ml of whole blood, GSHpx 59222 +/- 23699 U/l of whole blood, GSHred 110.8 +/- 22.5 U/l and TAS 0.33 +/- 0.15 mmol/l of serum. These indicators did not change in the control group with the exception of a statistically insignificant decrease in SOD activity after parturition. Statistically significant increase in blood SOD activity was noted only in the first day after injection of vitamins combined with selenium. These antioxidants also caused an insignificant increase in blood GSHpx activity in 72 hour following the injection, and in the second week after calving (statistically significant). The injection of antioxidants or
lysozyme
dimer did not change the activity of blood GSHred. However, an increase in the TAS was found in hour 24 (non significant) and 72 (statistically significant) following the single injection of
lysozyme
dimer.

The effect of
KLP
-602 (active substance:
lysozyme
dimer) on the replication of two animal viruses: the TK900 strain of Aujeszky's disease virus and the Roakin strain of the Newcastle disease virus were investigated. The maximal tolerable dose of the drug was determined for two cell cultures (CECC and GMK) and the effect of the medicine on the titre range of infectious viruses and their adsorption was assayed. The direct impact of
KLP
-602 on the viral strains used was also determined. And finally the replication dynamics of viruses in the presence of
KLP
-602 preparation was estimated.
KLP
-602 showed no direct effect on either the viruses applied in the study or their adsorption. The drug, introduced into the culture 24 hours before its infection, did not affect the replication of the pseudorabies virus, but decreased the titre of the Newcastle disease virus.
KLP
-602 introduced simultaneously with the infection considerably lowered the final titres of both viruses. The medicine had the greatest inhibitory effect on the replication dynamics of both types of viruses in the CECC and of the pseudorabies virus in the GMK culture upon the maximal tolerable concentrations of drug and low infectious doses of viruses applied.

Immunomodulation is a commonly used method of prophylaxis in humans and animals. Lysozyme dimer (
KLP
-602) was used at a dose of 50 ug/kg b.w. in order to correct the immunosuppression caused by the action of herbicide glyphosate (Roundup- Monsanto), which was used in a single bath for 10 minutes in a concentration of 100 mg/l of water. The investigations were carried out on 2 species of fish: the carp (Cyprinus carpio L.) and european catfish (Silurus glanis L.). Herbicide glyphosate caused a decrease in metabolic and phagocytic activity (RBA and PKA) and in proliferative response stimulated by Con A and LPS in carp and european catfish. The immunosuppression sustained for about 2 weeks. The results obtained indicate the possibility of correction of immunosuppression applying
lysozyme
dimmer (
KLP
-602) after use of which, the level of the studied indexes increased.

The dimer of
lysozyme
was administered to carp (Cyprinus carpio L.) in an attempt to reduce the suppression of response in fish exposed to the cypermethrin. Experimental studies were carried out on cells isolated from blood and pronephros of carp. In the in vivo studies fish were intoxicated with cypermethrin (synthetic pesticide) at a dose of LD5. After 24 h fish were injected intraperitoneally with a modulator-the dimer of
lysozyme
(
KLP
-602) at a dose of 0.02 mg/kg of body weight. In the study we determined the metabolic activity of phagocyte cells and the proliferative ability of lymphocytes stimulated by ConA and LPS. The results indicate that this modulator is useful for stimulation of cellular and humoral immunity after experimentally induced suppression by selected pesticide.

The natural compounds which affect defensive mechanisms of organism are important in prophylaxis and therapy of diseases in human and animals. Lysozyme is an enzyme which originates from chicken egg proteins. It modulates non-specific and humoral immunological mechanisms. A higher bioactivity has a
lysozyme
dimmer. The aim of study was assessment of influence of the
lysozyme
dimmer on chosen blood parameters, morphology and functions of internal organs and healing processes of experimental wounds in rabbits. Lysozyme dimmer (
KLP
-602) was used obtained process polymerisations enzyme lyzosyme from chicken egg white. The experimental group were on New Zealand White rabbits. Systemic reactions were investigated in animals after two injections of
lysozyme
dimer in dose 0,02 mg/kg b.w. during 21 days. Blood was collected before and after administration of
lysozyme
dimer in 4, 6, 24 h and in 3, 7, 21 day after first and in 4, 24 h and 3, 7 day after second injection. The following parameters were evaluated of red blood cells number (RBC), hemoglobin concentration (HGB), haematocrit value (HCT), red blood cell indices (MCV, MCH, MCHC), white blood cells number (WBC) and leukogram. Total serum proteins, components C3 and C4 of complement, immunoglobulins G and M, concentration were determined in the serum. Activated partial thromboplastin time (APTT), prothrombin time (PT) and fibrinogen concentration were evaluated in the plasma. Simultaneously, the activity of alkaline phosphatate, GPT and GOT were assessed. Administration rabbits with
lysozyme
dimmer caused slight decrease in RBC number, Hb concentration, HCT and the neutrophils percentage and increase in the lymphocytes percentage. The concentrations of TSP, immunoglobulins, the components of complement ware increased too. APTT an PT were normal but the fibrinogen concentration was increased. The activity of GOT and GPT were unchanged. Changes are in range of value normal and step out more quickly after two injection than after firste injection. The morphology of organs (liver, lungs, heart and kidneys) was normal.
