In patients with haptoglobin (Hp) 2-2, the incidence rate of tuberculosis of respiratory organs was found out to be 1.7 times as high as among patients with Hp 2-1 and 1-1; in those persons with aggravated heredity this parameter was 3.4 times as high. Based on the above data, in 414 first diagnosed cases with different phenotype pulmonary tuberculosis a comparative analysis has been done of certain factors of nonspecific bodily reactivity, which showed that in patients with Hp 2-2 versus Hp 2-1 blood levels of total haptoglobin, ceruloplasmin get decreased while total proteolytic activity is noted to be on the increase. The lysozyme concentration is significantly lower than in patients with Hp 2-1 and remains within the normal range. Poorly manifest reaction to the infectious agent of ceruloplasmin, total Hp, lysozyme which have distinct effects and are interrelated with mechanisms of immune defense creates a favourable background in subjects with Hp 2-2 for origination, development and progressing of the tuberculous process.

285 Patients suffering from the respiratory tract organs primary tuberculosis were subjected to the tests on definition of blood groups according to AB0 system, as well as its total proteolytic activity, alpha 1-antitrypsin, ceruloplasmin, general haptoglobin, lysozyme, malonic dialdehyde and diene conjugates levels were estimated. The sick persons as compared with the healthy ones were defined to reveal a tendency to increase of the persons with 0(I)- and B(III)-blood groups and decrease of those ones with A(II)-groups. Independently on the pulmonary tuberculosis patients phenotypic index their tested blood biochemical indices levels increase. As an exception is the proteolytic activity of the persons with B(III)- blood group, and also alpha 1-antitrypsin and lysozyme--with 0(I) and AB(IV) groups, in this case their rates failed to exceed the norm confidential interval. The blood metabolic parameters were defined as independent on its group.

Pseudoparaproteins were observed in 129 (10.5%) of 1,229 high resolution protein electrophoretic fractionations of serum (N = 847), urine (N = 368), or cerebrospinal fluid (N = 14) performed in this laboratory during a 12-month period. The pseudoparaproteins identified in serum electrophoretic patterns included fibrinogen, C-reactive protein, hemoglobin-haptoglobin complex, elevated beta-globulins (transferrin and C3), lysozyme (muramidase), and an extended migration artifact. In the electrophoretic patterns of urine, the pseudoparaproteins consisted of nonspecific gamma zone bands of varying intensity. Gamma zone trace protein in the cerebrospinal fluid was often of sufficient intensity to cause potential confusion with oligoclonal bands. Awareness of the characteristic electrophoretic migration positions of these pseudoparaproteins helps to avoid unnecessary ancillary testing and expense.

The aim of our study was to assess the effects of cigarette smoking on selected indices of immunity. The study comprised 116 men divided into three groups: 37 subjects smoking for not more than 10 yr, 39 subjects smoking for more than 10 yr, and control group consisting of 40 age-matched men who never used to smoke. The following parameters were studied: total number of lymphocytes, B-cells, T-cells subpopulations: (CD3+)T-, (CD4+)T-helper, (CD8+)T-cytotoxic and (CD16+)natural killer (NK)-cells and serum concentration of immunoglobulins A, D, G and M, C3c and C4 complement components, acute phase proteins: alpha(1)-acid glycoprotein, haptoglobin, ceruloplasmin and lysozyme. The (CD4+)/(CD8+) ratio was also calculated. The suppressive effect of tobacco smoke on human immunity was seen as decreased serum concentration of immunoglobulins and lysozyme, especially in men smoking for more than 10 yr, decreased (CD16+)NK-cells absolute number and elevated population of (CD8+)T-cytotoxic lymphocytes entailing a decrease in CD4+/CD8+ ratio.

Urinary proteins from 14 patients with tubulointerstitial nephritis were analyzed by cellulose acetate membrane electrophoresis. Urinary total protein concentrations were measured, and urinary 15 proteins (prealbumin, albumin, alpha(1)-microglobulin, alpha(1)-antitrypsin, alpha(2)-macroglobulin, haptoglobin, retinol binding protein, transferrin, beta(2)-microglobulin, IgA, IgG, kappa- and lambda-light chains, cystatin C, and lysozyme) were identified by the use of a rapid and highly sensitive colloidal silver staining reagent suited for use with cellulose acetate membranes, as reported previously by Matsuda et al. (J Clin Lab Anal 15:171-174, 2001; Clin Chem47:763-766, 2001) and Hiratsuka et al. (J Clin Lab Anal 10:403-406, 1996). We also analyzed urinary total protein concentration and urinary protein fractions according to the presence of acute or nonacute interstitial nephritis. In addition, the relationship between urinary protein fraction and complications of interstitial nephritis was analyzed. The goal of this work was to find a useful index for the diagnosis of tubulointerstitial nephritis.

Linoleic (18:2n-6) and alpha-linolenic acids (18:3n-3) have many important physiological functions including immunomodulation. We tested how immunization influences the metabolism of 18:2n-6 and 18:3n-3 in the neck muscle of pigs. At 35 d old, pigs received either an intramuscular neck injection containing hen egg white lysozyme (HEWL), killed Mycobacterium tuberculosis, and Freund's complete adjuvant (immunized) or PBS (control). At 49 d old, immunized pigs received a booster injection of HEWL and Freund's incomplete adjuvant, and the control pigs received PBS into the neck. At 56 d old, all pigs received an intradermal injection of Mycobacterium bovis into the hind leg to induce a delayed-type hypersensitivity (DTH) reaction. At 57 d old, immunized pigs had a twofold increase in serum haptoglobin, a 10-fold increase in antibodies to HEWL, and the skinfold at the DTH reaction site was 10 times thicker than the controls. Both 18:2n-6 and 18:3n-3 (% composition) were approximately 25% lower in muscle TG, 40% lower in FFA, 50% lower in phospholipids, but not different in cholesteryl esters of the neck muscle of immunized pigs. The antigens in this model induce an increased response in the innate (haptoglobin), humoral (antibodies), and cellular (DTH) immune systems as well as a preferential decrease of 18:2n-6 and 18:3n-3 in the inflamed neck muscle. It appears that 18:2n-6 and 18:3n-3 are preferentially metabolized (possibly beta-oxidized) in response to antigens.

Alpha-linolenic acid (18:3omega3) has many important physiological functions including being beta-oxidized, serving a precursor to the synthesis of other lipids and it has immunomodulation properties. The objective of the present study was to test the effects of immunization and dietary 18:3omega3 on immune function and the fatty acid profile of immunized pig tissues. Piglets suckled from sows consuming either a control or high 18:3omega3 diet until 14 days old when they were weaned onto a similar diet as the sow and were moved to a segregated nursery for the remainder of the study. At 35 days of age, pigs on both diets (2 x 2 factorial design) received either an injection containing hen eggwhite lysozyme (HEWL), killed Mycobacterium tuberculosis and Freund's complete adjuvant (immunized) or phosphate buffered saline (PBS) (non-immunized) into the neck followed by a booster injection 2 weeks later and induction of delayed-type hypersensitivity (DTH) one week later. Immunization increased (compared to non-immunized) while the high 18:3omega3 diet decreased haptoglobin by 30% compared to pigs consuming the control diet. Immunized pigs had a seven-fold increase in antibodies to HEWL and pigs consuming the high 18:3omega3 diet also had transiently higher levels of serum antibodies. There was a diet by immunization interaction on the DTH reaction such that immunized pigs consuming the high 18:3omega3 had the largest DTH reaction. The neck muscle proximal to the site of injection of immunized pigs had 10-30% lower levels of triglyceride and phospholipid linoleic (18:2omega6) and 18:3omega3 compared to non-immunized pigs. Thus, a high 18:3omega3 intake in pigs modulates immune function and tissue fatty acids in response to immunization.

During the periparturient period, some impairment of immune defences were observed. Reference values for the different non-specific immune parameters in cows are not generally available, thus limiting the application of these parameters in dairy practice. This paper reports the data on the measurements of different parameters in the blood, and explores the possible influence of the herd on the non-specific immune status of the cow. Five herds located in Northern Italy were selected and overall 39 heifers were enrolled in the trial. Blood samples were taken 14 and 7 days before the expected date of calving, then at 7, 14, 21, 28, 45, 60, and 75 days after calving. The parameters assessed were N-acetyl-beta-glucosaminidase (NAGase), lysozyme, nitric oxide, superoxide dismutase, haptoglobin, respiratory burst, and serum protein profile. After calving, a significant decrease of respiratory burst and nitric oxide concentration were observed in comparison with the pre-calving values but not with the post-calving samplings. Total proteins, beta- and gamma-globulins showed a progressive and significant increase in concentration after calving, in comparison with pre-calving values. The results of the study confirmed that a decrease of immune functions can be observed in commercial dairy herds in the first four weeks after calving. The amplitude of this phenomenon is not common to all animals and all herds, suggesting the possibility to reduce the impairment by improved management and genetic selection.

Studies that blend a comparative approach to immunology with an appreciation for physiological ecology are defining an important new field in biology--ecological immunology. However, a panel of assays that permits a comparative approach to immunology is not yet available. In this paper, we describe several assays of innate immunity that do not require species-specific reagents and therefore ideal for use in comparative immunology studies. We optimized the assays for use in small birds, where sample volumes are limiting. The bactericidal assay measures the capacity of whole blood to kill microorganisms, and integrates many important components of constitutive immunity. The phagocytosis assay measures the phagocytic capacity of macrophages in whole blood. Bioassays for mannan binding protein and lysozyme can be used to measure inflammation-induced levels of these acute phase proteins in the plasma. Species differences in bactericidal and phagocytic activities against Staphylococcus aureus and Escherichia coli were observed in populations of captive and in free-living birds, demonstrating the assays' utility for multi-species comparisons. However, clay-colored thrushes (Turdus grayi) that were stressed by prolonged capture and handling had diminished phagocytic and antibacterial activities, indicating the need to conduct these assays soon after capture. When birds were challenged with lipopolysaccharide (LPS), levels of mannan-binding protein, lysozyme, and haptoglobin were elevated and bactericidal and phagocytic activities of blood were altered, indicating that these measurements are sensitive to the current infection status of the animal. All assays could be done on as little as 10 microL of blood or plasma and should be useful in field studies of comparative immunity.

Broilers are typically raised commercially in dim lighting. It has been suggested that providing brighter light intensity could improve health and provide opportunities for more normal behavioral rhythms. We examined the effects of 3 photophase light intensities (5, 50, and 200 lx) on activity patterns, immune function, and eye and leg condition of broilers (n = 753; 6 replicate pens/treatment). Broilers were reared with one of these intensities from 1 to 6 wk of age; photoperiod consisted of 16L:8D with 1 lx intensity during the scotophase. Broilers reared with 5 lx were less active (P = 0.023) during the day than 50 or 200 lx and showed less (P < 0.0001) change in activity between day and night than 50 or 200 lx. There was no difference between treatments for final BW (2.30 +/- 0.02 kg) or for most immune parameters (IgG primary and secondary responses to keyhole limpet hemocyanin, B and T lymphocyte proliferation, plasma lysozyme, haptoglobin, NO, whole blood killing of Escherichia coli and Staphylococcus aureus), but there was a trend (P = 0.072) for a greater IgM response in 50 lx (6.21 titer) than 5 lx (5.78 titer), with 200 lx (5.92 titer) intermediate. There was no effect of light intensity on back-to-front (1.13 +/- 0.01 cm) or side-to-side (1.48 +/- 0.01 cm) diameter of the eyes or on corneal radii (0.82 +/- 0.01 cm), but 5 lx (2.33 +/- 0.07 g) had heavier eyes (P = 0.002) than 50 lx (2.09 +/- 0.04 g) or 200 lx (2.11 +/- 0.04 g). There were no differences in gait score, although 200 lx broilers had more hock and footpad bruising (P = 0.038) but fewer erosions (P = 0.006) than 5 or 50 lx. Increased daylight intensity had little effect on broiler health but resulted in more pronounced behavioral rhythms.
