The percentage of bacteriocin-producing and phage-producing Klebsiella strains was as follows: K. pneumoniae-10%, K. ozaenae-7%, K. rhinoscleromatis-9%. The antimicrobial spectrum of the studied inducible particler was broad and was not limited by the frames of the genus and family. Bacteriocins and bacteriophages from Klebsiella were active to Klebsiella, Enterobacter, Escherichia, Shigella and Proteus representatives significant in medicine. Klebocins and Klebsiella phages exhibited antagonistic effects to phytopathogenic bacteria. Some strains of Erwinia and Pseudomonas were sensitive to phages or bacteriocins from Klebsiella. Bacteriocins protected corn and tomato seeds from contamination by erwinioses agents. All cultures of Agrobacterium, Corynebacterium, Micrococcus, Staphylococcus, Streptococcus were resistant to action of phages and klebocins. Bacteriocins from Klebsiella were assayed for their sensitivity to trypsin, chymotrypsin, lysozyme, ribonuclease, deoxyribonuclease. Action of klebocins was associated with a protein component. Proceeding from data of diffusion through the disc ultrafiltration membranes molecular weight of klebocins was in the range of 30,000 and 50,000 Da.

This study evaluated the Promega Magic Minipreps (MM) (Promega Corporation, Madison, WI) DNA purification system for use in plasmid analysis of common nosocomial bacterial pathogens. The MM system is a kit that includes lysis solutions and buffers and incorporates a minicolumn of DNA binding resin for recovery of plasmid DNA. The MM system was used according to the manufacturer's directions to recover plasmids for agarose gel electrophoresis from clinical isolates of Acinetobacter calcoaceticus, Enterobacter cloacae, and Klebsiella pneumoniae. For Salmonella enteritidis and Staphylococcus aureus, lysozyme and lysostaphin, respectively, were used for pretreatment. Plasmid DNA from ten isolates could be recovered in approximately one hour with very little manipulation and no phenol/chloroform extractions and was suitable for restriction endonuclease digestion. Compared with a standard miniprep protocol, the MM system was much easier to perform and resulted in significant cost savings due to a 50% reduction in technologist time. The authors conclude that the MM system is a convenient and cost-effective method for clinical microbiology laboratories for recovering plasmid DNA from nosocomial bacterial pathogens.

DEODAN is a lysozyme lysate from Lactobacillus bulgaricus for oral administration which has shown antitumor activity in mice and humans. The effects of this preparation on some functions of monocytes/macrophages and on host resistance to experimental infections were examined. The oral administration to mice of DEODAN-150 mg/kg daily (the recommended dose in humans) caused an increase of the spreading ability and phagocytic activity of peritoneal macrophages, which showed morphological signs of cell activation. The level of Interleukin-1 (IL-1) secreted in the culture supernatant of peritoneal macrophages of DEODAN-treated mice was found to be slightly increased only when the mice were treated with 150 mg/kg DEODAN for 10 days. However, the in vitro incubation of human blood monocytes with DEODAN resulted in induction of membrane-bound and cytoplasmic IL-1 and Tumor Necrosis Factor (TNF)-alpha. The oral treatment of mice with DEODAN also caused a decrease in mortality after experimental infections with Klebsiella pneumoniae and Listeria monocytogenes. These results indicate that DEODAN activates the phagocytic and secretory functions of mononuclear cells and increases host resistance to bacterial infections.

Nocardia lysozyme digest (NLD) was administered intraperitoneally (i.p.) at a dose of 500 micrograms/kg to normal and immunosuppressed mice for 3 consecutive days prior to inoculation with Klebsiella pneumoniae. A protective effect was observed when the pathogen was injected subcutaneously and intravenously, as opposed to an aggravating effect obtained in the case of intraperitoneal inoculation The i.p. administration of NLD partially restored the immunosuppression caused by cyclophosphamide but did not change cobra venom-induced deterioration of the infection. The results obtained could be regarded as a consequence of the lowered alternative pathway serum complement activity and the crucial role of the diminished level of complement in the peritoneal cavity.

The constantly increasing number of substances with adjuvant activity outpaces the elucidation of their mode of action. This problem is of great importance as the immunomodulatory action of an adjuvant is time- and route-dependent, which implies that administration at a different moment or site may result in a reduced immune response. In the present work the possibility to achieve dual effect (stimulatory or inhibitory) is regarded in the light of the complement system. The object of the study is a preparation obtained by lysozyme digestion of Nocardia opaca cell walls (NLD). According to the results, the administration of NLD to mice (i.p. at a daily dose of 0.5 mg kg-1) during 3 days prior to the antigen resulted in an inhibition of serum antibody level against sheep red blood cells (SRBC) and lipopolysaccharide (LPS). At the same time, the preparation stimulated the antibody response to SRBC if it was applied after the antigen. The ability of NLD to ensure protection against experimental infection with Klebsiella pneumoniae was comparatively studied in complement-normal mice (strain ICR) and in C5-deficient mice (strain DBA/2). Firstly, it was established that complement-deficient mice were more resistant to infection than complement-normal. Secondly, the preparation expressed a protective effect in C5-deficient animals; nevertheless the inoculation was done s.c. or i.v. The departure of the infection depended on the rate of opsonization of K. pneumoniae. Under certain conditions NLD can provoke excessive C3 activation, which might aggravate the course of the infection. The preparation augmented the host response to second challenge with K. pneumoniae of complement-normal and C5-deficient mice.

The extraction of periplasmic beta-lactamases from Gram-negative bacilli is a necessary preliminary step to analytical isoelectric focusing. Previously described methods are time-consuming and require large amounts of broth. We describe a lysozyme-based method which needs just 5 mL broth and requires less than 24 h to perform. The method was reproducible in extracting beta-lactamases from reference strains containing known beta-lactamases. We applied the method to a collection of more than 70 extended-spectrum beta-lactamase-producing isolates from a multinational study of bacteremic isolates of Klebsiella pneumoniae. Further studies are being undertaken to assess the method's applicability to other bacterial species.

The influence of the exometabolites of the fungus S. boulardii, contained in the probiotic preparation "Enterol", on the biological properties of opportunistic and pathogenic enterobacteria of fecal microflora (inactivation of lysozyme, colicin production, hemolytic activity, antibiotic resistance) was studied. The study revealed that the supernatants of S. boulardii decreased antilysozyme activity (ALA) in lactose positive (lac+) and lactose negative (lac-) Escherichia coli and Klebsiella strains, but produced no influence on ALA in Salmonella. In response to the action of S. boulardii exometabolites colicin production in E. coli (lac+) was found to increase, while in E. coli (lac-) colicin production was suppressed. An increase in the sensitivity of lactose negative E. coli to cefazolin and cefotaxime under the action of S. boulardii supenatants was noted. The results obtained in this study show the probable mechanism of the corrective action of "Enterol" on intestinal biocenosis, which should be taken into consideration in the differentiated selection of probiotics for the treatment of intestinal dysbacteriosis.

The influence of the subinhibitory concentrations of bifidobacterial exometabolites on the capacity for inactivating lysozyme in pathogenic and opportunistic enterobacteria was determined. The supernatant of B. bifidum 1 suppressed antilysozyme activity (ALA) in most Klebsiella strains and in some pathogenic Escherichia coli strains, but increased ALA in some E. coli of indigenous flora and failed to influence ALA in Salmonellae. The stimulating effect of the vital activity products of bifidobacteria with respect to the antilysozyme sign in some S. enteritidis isolates and diarrheagenic E. coli was found. These properties should be taken into consideration while selecting strains for the construction of probiotics.

The influence of vitamins of group B (B1, B6, B12) and vitamin C on the antilysozyme activity (ALA) of pathogenic and opportunistic bacteria (Salmonella, Klebsiella, Escherichia coli) has been determined in a number of in vitro experiments. In E. coli vitamins had different influence on ALA: vitamins B1 and C (at a concentration of 5 mg/ml) increased this activity in E. coli, while vitamins B12 and C (at concentrations of 0.5 and 0.05 mg/ml) decreased it. In Klebsiella vitamins B1, B12 and C (at a concentration of 0.05 mg/ml) decreased ALA, while after the addition of vitamin C at concentrations of 0.5 and 5 mg/ml into the medium ALA increased. In Salmonella vitamins of group B had no influence on ALA, ascorbic acid at a low concentration (0.5 mg/ml) decreased their capacity for inactivating lysozyme. In enterobacteria vitamin B6 produced no changes in ALA.

Klebsiella pneumoniae is a common virulent causative agent for pneumonia. Lysozyme has previously been shown to play an important role in nonimmune host defense of the airways. This study was undertaken to assess the role of lysozyme M, the major isoform of lysozyme in mouse lung, in the killing of K. pneumoniae in lysozyme M(-/-) mice and transgenic mice with increased expression of lysozyme (lysozyme(tg) mice). The airways of lysozyme M(-/-) mice maintained in a pathogen-free facility were colonized by Lactobacilli, a component of the oropharyngeal flora. No lactobacilli were detected in the lungs of wild-type (WT) or lysozyme(tg) mice. Twenty-four hours after intratracheal infection with K. pneumoniae, bacterial killing was enhanced 9-fold in lysozyme(tg) mice compared with WT mice and 43-fold compared with lysozyme M(-/-) mice. In survival studies, no lysozyme M(-/-) mice survived beyond 72 hours after infection, whereas 75% of lysozyme(tg) (p < 0.01) and 25% of WT mice survived to 120 hours (p < 0.01). Deficiency of lysozyme M in the lungs increased susceptibility to K. pneumoniae infection, whereas increased expression of lysozyme conferred resistance to infection and enhanced survival.
