, the main proteinase component of the juice of pineapple fruit, has been purified and characterized. 1. Efficient extraction of this enzyme from the crude material was possible using "Cellulosin AP," a microbial polysaccharidase preparation containing cellulase, hemicellulase, and
pectinase
. The enzyme was purified mainly by successive applications of anion-exchange chromatography, yielding an apparently homogeneous protein as judged by several physical, chemical, and immunochemical criteria. Properties of
FA2
include: molecular weight, 31,000; isoelectric point, pH 4.6; absorbance at 280 nm of a 1% solution at pH 7.0 per cm, 19.2. 2.

An isozyme of
pectinase
from Aspergillus niger with
polygalacturonase
activity caused chitosanolysis at pH 3.5, resulting in low-molecular weight chitosan (86%), chitooligosaccharides (COs, 4.8%) and monomers (2.2%). HPLC showed the presence of COs with DP ranging from 2 to 6. Charcoal-Celite chromatography and re-N-acetylation of the COs followed by CD, IR, MALDI-TOF-MS and
FAB
-MS analyses revealed an abundance of chitobiose, chitotriose and chitotetraose. The COs-monomeric mixture showed a bactericidal effect towards Bacillus cereus and Escherichia coli more efficiently than native chitosan. Among the chitooligomers, the hexamer showed maximum antibacterial effect followed by the penta-, tetra-, tri- and dimers. Of the two monomers, only GlcN showed slight bacterial growth inhibition. SEM revealed bactericidal action patterns of COs-monomeric mixture towards B. cereus and E. coli.

In the present study, a biosurfactant was synthesized by using a bacterial strain of Pseudomonas aeruginosa in minimal media provided with n-heptadecane as sole carbon source under shake-flask conditions. The biosurfactant was isolated (by acid precipitation, solvent extraction, and rotary evaporation), purified (by column chromatography and TLC), identified (by
FAB
-MS, FTIR, and 1D-(1)H NMR), and chemo-physical characterized (by tensiometry). Two principal rhamnolipid congeners were identified as dirhamnolipid RRC10C10 and monorhamnolipid RC10C10 with a CMC of 50mg/L. The biosurfactant, hence produced, was applied in sole and in combination with
pectinase
in scouring of cotton fabric in contrast to conventional scouring agents of NaOH and anionic surfactant SDS. The scoured cotton fabric was investigated for its weight loss, residual oil and grease, wettability, whiteness, and tensile strength. The results were compared both for conventional and biological approaches. The scouring with biosurfactant plus
pectinase
was equivalent to or better in efficiency than conventional alkaline scouring. The former process is additionally environmentally friendly and bio-compatible. Scanning electron microscopy of cotton fabric showed that the alkaline scouring deteriorates the fabric texture whereas bioscouring with biosurfactant plus
pectinase
gently removes hydrophobic impurities from the cotton fabric.
