Binding region and link protein were prepared from pig laryngeal cartilage proteoglycans after chondroitinase ABC and trypsin digestion. Experiments on gel chromatography showed the purified binding region to interact reversibly with hyaluronate (HA), and this binding was also shown to be stabilized by native link protein. The trypsin-prepared link protein showed properties of self-association in solution that were partially inhibited by oligosaccharides (HA10-16) and abolished by modification of free amino groups (lysine residues) with 2-methylmaleic anhydride. The Mr (sedimentation equilibrium) of the modified link protein was 41 700. Analysis of binding region showed it to contain 25% (w/w) carbohydrate, mainly in galactose, glucosamine, mannose and galactosamine. It contained some keratan sulphate, as digestion with endo-beta-D-galactosidase (keratanase) removed 28% galactose and 25% glucosamine and the Mr (sedimentation equilibrium) decreased from 66 500 to 60 800. After keratanase digestion the interaction with polyclonal antibodies specific for binding region was unaffected, but the response in a radioimmunoassay with a monoclonal antibody to keratan sulphate was decreased by 47%. Preparation of a complex between binding region, link protein and HA approximately 34 showed a single component (5.5S) of Mr (sedimentation equilibrium) 133 500. In this complex the antigenic determinants of link protein appeared masked, as previously found with proteoglycan aggregates. The isolated binding region and link protein were thus shown to retain properties comparable with those involved in the structure and organization of proteoglycan aggregates.

Chick-embryo cartilage contains a unique set of proteoglycans. Type H proteoglycan (PG-H) is the most abundant, constituting over 90% of the total cartilage hexuronate. We previously showed that treatment of PG-H with chondroitinase ACII and keratanase yields a protein-enriched core molecule [PG(-CS,KS)] with enzymically modified linkage oligosaccharides of the chondroitin sulphate and keratan sulphate chains. We report here that further treatment of PG(-CS,KS) with pepsin and N-oligosaccharide glycopeptidase (almond glycopeptidase) released four distinct types of mannose-containing oligosaccharide. Two of them were shown to be: (Formula: see text). Of the mannose-containing glycopeptides formed by pepsin digestion, about 40% (as mannose) were resistant to N-oligosaccharide glycopeptidase. Since the resistant fraction was enriched in keratan sulphate remnants, it is suggest that the mannose-containing oligosaccharides in this fraction represent those located in a keratan sulphate-enriched region of PG-H.

The glycosaminoglycan synthesis in Furth solid mastocytoma tissue has been studied. Approx. 10% of the polysaccharide isolated after incubation in vitro with [(14)C]-glucosamine was digestible with chondroitinase ABC and the product of digestion was identified as 2-acetamido-2-deoxy-3-O-(beta-d-gluco-4-enepyranosyluronic acid)-4-O-sulpho-d-galactose. Similarly, labelling of polysaccharide in vivo with (35)SO(4) (2-) followed by isolation of mast-cell fractions by density-gradient centrifugation on colloidal silica revealed the presence of a polysaccharide which migrated as did chondroitin sulphate on electrophoresis in barium acetate. Chondroitinase ABC produced the same digestion product as before. Finally, the presence of the UDP-N-acetylgalactosamine-chondroitin 6-sulphate hexasaccharide N-acetylgalactosaminyltransferase previously implicated in chondroitin sulphate biosynthesis was demonstrated in microsomal particles from fractions of purified mast cells.

Lysosomal arylsulfatases A and B of peripheral leukocytes from patients with chronic myelogenous leukemia and from healthy subjects were studied. Two enzyme activities of leukemia cells were significantly higher than those of cells from healthy subjects, irrespective of total and differential counts of leukemic cells. Upon anion-exchange chromatography, the arylsulfatases of chronic myelogenous leukemia cells and normal leukocytes were separated into the basic B enzyme and its anionic variant (B1) and A enzyme. However, the amount of B1 enzyme relative to B enzyme or the activity ratio of B1 enzyme to total arylsulfatase B (B + B1) was higher in chronic myelogenous leukemia cells than in normal cells. The anionic property of the enzyme was found to be due to phosphate groups bound to the carbohydrate moiety of the arylsulfatase, based on the following results. When B1 enzyme was treated with alkaline phosphatase followed by isoelectric focusing, it was changed to a less anionic enzyme with heterogeneous components which are ascribed to phosphodiester groups linked to the heterogeneous carbohydrate moiety of the enzyme; no effect was observed by sialidase treatment. Upon treatment of B1 enzyme with endo-beta-N-acetylglucosaminidase H, which cleaves sugar chains of a high mannose type in glycoproteins, the anionic heterogeneous components were converted to the basic component similar to B enzyme. From our present and previous observations, it can be concluded that the increase of phosphorylated forms of the lysosomal hydrolase represents one characteristic of rapidly proliferating neoplastic cells.

Bovine nasal cartilage proteoglycan monomer which had been digested with chondroitinase ABC to form the keratan sulfate-protein core was injected into rabbits. High titer antiserum was obtained as judged by the binding of 125I-labeled keratan sulfate-protein core. Native proteoglycan did not inhibit the interaction of the antiserum with labeled keratan sulfate-protein core. However, the disaccharide obtained from chondroitin 4-sulfate by the action of chondroitinase ABC, 2-acetamido-2-deoxy-3-O-(beta-D-gluco-4-enepyranosyluronic acid)-4-O-sulfo-D-galactose, inhibited the interaction 50% at a concentration of 500 microM. The corresponding 6-sulfated and nonsulfated disaccharides at the same concentration gave 15% and 10% inhibition, respectively. Chondroitinase-digested proteoglycan from Swarm rat chondrosarcoma was strongly inhibitory, while the hyaluronidase-digested chondrosarcoma proteoglycan exhibited no detectable inhibition. Evidently, the antiserum raised against chondroitinase ABC-digested bovine nasal cartilage proteoglycan contains antibodies which recognize the unsaturated uronic acid residue linked to N-acetylgalactosamine 4-sulfate. These antibodies will be valuable for identifying and quantitating chondroitin 4-sulfate-containing proteoglycans in tissues.

This work provides data demonstrating a stimulatory effect of insulin on macromolecular events occurring in cultured regeneration blastemata and demonstrates a synergistic interdependence between nerves and insulin in newt limb regeneration. The current experiments provide evidence for the following: (1) Insulin is paramount for expression of the mitogenic effect of nerves on cultures blastemata. (2) Insulin stimulates the incorporation of (3H)uridine into the acid-insoluble fraction of blastemal homogenates, but it does not alter the turnover rate of incorporated labeled uridine. (3) Insulin also stimulates the incorporation of 35SO4 and (3H)leucine into both chondroitinase-sensitive and chondroitinase-resistant blastemal proteoglycans. (4) Insulin increases the uptake of radiolabeled precursors by the blastemata, namely, (3H)leucine, (3H)uridine, 35SO4, (3H)alpha-aminoisobutyrate, and (3H)2-deoxy-D-glucose. The importance of insulin in the regulation of newt limb regeneration is discussed.

An explant culture of 15 cynomolgus monkey corneas was incubated with [35S]sulfate and [2-3H]mannose as labeling precursors. A 4 M guanidine HCl extract of the corneal stromas was prepared and combined with a 4 M guanidine HCl extract of stromas from 300 unlabeled corneas. The keratan sulfate proteoglycans in the combined extracts were purified by a combination of DEAE-cellulose chromatography, chondroitinase ABC digestion to remove chondroitin-dermatan sulfate proteoglycans, and elution from immobilized concanavalin A. The purified keratan sulfate proteoglycan was digested with papain and the digest was eluted on DEAE-Sephacel. The unbound fraction contained 59% of the 3H activity and consisted of intact oligosaccharide-peptides. The bound fraction, consisting of keratan sulfate chains linked to peptides, eluted during a linear 0-0.75 M NaCl gradient as a peak centered at approximately 0.6 M NaCl and contained 41% of the 3H and all of the 35S activity in the original proteoglycan. The chains were digested with endo-beta-galactosidase, and the digest was eluted on DEAE-Sephacel with a linear 0-0.75 M NaCl gradient. Most of the sulfated digestion fragments from the chains eluted as several distinct peaks during the gradient. All the 3H activity eluted in the unbound volume along with a small proportion of 35S activity. This unbound fraction was eluted on Bio-Gel P-10 to give a 3H peak (Kav = 0.46) well resolved from the remaining 35S activity which eluted near the total volume. This 3H peak contained the oligosaccharide-peptides derived from the linkage region between the keratan sulfate chains and the core protein. Structural analyses of the linkage region oligosaccharides and the intact oligosaccharides (Nilsson, B., Nakazawa, K., Hassell, J.R., Newsome, D.A., and Hascall, V.C. (1983) J. Biol. Chem. 258, 6056-6063) in combination with the 3H-labeling data suggest that the intact keratan sulfate proteoglycans contain an average of about one intact oligosaccharide per keratan sulfate linkage site.

The structure of adult bovine articular cartilage high density proteoglycans (PG-I) was studied by degradation with Pronase, chondroitinase ABC, and alkaline borohydride treatments and fractionation and analysis of the products. The keratan sulfate (KS) peptides were rich in glutamic acid, proline, and serine and had a low glycine content. The chondroitin sulfate (CS) peptides had a high content of serine, glycine, and glutamic acid and a much lower proline content than the KS peptides. The data indicate that the KS and CS chains occur in more distinct regions of the protein core(s) than in bovine nasal cartilage PG. After alkaline borohydride treatment there was an almost quantitative conversion of xylose to xylitol and galactosaminitol was the only hexosaminitol detected in KS fractions. The results obtained indicated that the alkali-labile bonds linking the CS and KS chains are the same as those reported to occur in other cartilage PGs. The Mr of the KS chains calculated from the glucosamine and galactosaminitol contents gave values of 6,000-7,000, although gel chromatography and light scattering measurements indicated considerable heterogeneity. The KS and CS chains were quantitatively precipitated by cetylpyridinium chloride and the KS and a portion (15%) of the CS chains were found to be soluble in 1% cetylpyridinium chloride. The abnormal solubility properties of the CS chains in the presence of 1% cetylpyridinium chloride is thought to be due to their low sulfate content. The molecular weight of the remainder of the CS chains, based on the ratio of xylitol to galactosamine, varied from 6,500 to 16,000. The low Mr CS chains were rich in 6-sulfated disaccharides whereas the higher Mr chains had a higher content of 4-sulfated disaccharides. The ratio of galactose to xylitol also varied with Mr. These results indicate similarities in the structure of the adult bovine articular cartilage PG-Is to other cartilage high density PGs. The heterogeneities observed in the composition of the KS and CS chains, and their occurrence in relatively distinct regions of the protein core(s) indicate, however, that there is still much to be learned about the structure of these complex macromolecules.

Human lung cancer transplanted into athymic mice contains predominantly an acidic variant (designated B1) of lysosomal arylsulfatase B. B1 enzyme was suggested to be phosphorylated and sialylated (Gasa, S., Makita, A., Kameya, T., Kodama, T., Koide, T., Tsumuraya, M., and Komai, T. (1981) Eur. J. Biochem. 116, 497-503). In order to determine the localization of phosphate in B1 enzyme, we labeled in vivo the transplanted tumor with [32P]H3PO4 or [3H]glucosamine and purified B1 enzyme by immunoprecipitation. Bio-Gel chromatography of the labeled B1 enzyme treated with endoglycosidase H demonstrated that both the excluded and included materials were labeled with 32P and 3H. From acid hydrolysate of the excluded materials, phosphorylated serine and threonine were detected. Protein phosphorylation of arylsulfatase was confirmed by in vitro labeling experiments with [gamma-32P]ATP. By incubation of the tumor homogenate with ATP followed by isolation of the enzymes, B1 enzyme had a significant amount of radioactivity, whereas the B enzyme had little; by exogenous protein kinase, partially purified B enzyme was phosphorylated 35 times more than B1 enzyme. Acid hydrolysate of the included materials in the Bio-Gel column demonstrated mannose 6-phosphate and an unknown phosphorylated compound which migrates more than Man-6-P on electrophoresis and chromatography.

Rat ovarian granulosa cells were isolated from immature female rats after stimulation with pregnant mare's serum gonadotropin and maintained in culture. Proteoglycans were labeled with [35S]sulfate, [3H]glucosamine, [3H]serine, or [3H]mannose as precursors. A low buoyant density dermatan sulfate proteoglycan was separated from a larger hydrodynamic size, high buoyant density dermatan sulfate proteoglycan and from a heparan sulfate proteoglycan using DEAE-Sephacel and Sepharose CL-4B chromatography under dissociative conditions in the presence of detergent. This low buoyant density dermatan sulfate proteoglycan, which constituted approximately 30% of the 35S-labeled proteoglycans in the culture medium, has a relatively small hydrodynamic size (Kd = 0.45 on Sepharose CL-4B) and shows a broad buoyant density distribution in CsCl density gradients, primarily due to the heterogeneity in glycosaminoglycan composition. The average molecular weight of the protein coreoligosaccharide complex obtained by chondroitinase ABC digestion of the proteoglycan was estimated to be approximately 230,000 by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. After digestion with chondroitinase ABC, the dermatan sulfate chains (average Mr = 33,000) yielded 81% 4-sulfated disaccharides and 17% disulfated disaccharides (sulfate groups on the 6-position of the galNAc and probably on the 2-position of the iduronic acid). Alkaline borohydride treatment of this proteoglycan released three distinct size species of oligosaccharides; a species of N-linked oligosaccharide which contains mannose, glcNAc, and sialic acid, and two species of O-linked oligosaccharides. Trypsin digestion of the proteoglycan generated fragments which contain (a) glycosaminoglycan-peptides with an average of 2 dermatan sulfate chains, (b) clusters of O-linked oligosaccharide-peptides, and (c) N-linked oligosaccharide-peptides.
