Desulfated form of the 3Y1 secreted fibronectin was prepared by treatment with arylsulfatases. Under optimal conditions, the degrees of tyrosine-desulfation of [35S]sulfate-labeled fibronectin by arylsulfatases from Helix pomatia (Type H-1), Patalle vulgata (Type V) and Abalone entrails (Type
VIII
) were determined to be 55.7%, 54.9% and 76.4%. Upon desulfation of [3H]leucine-labeled fibronectin by Type H-1 or Type V
arylsulfatase
, gelatin-binding affinity remained unchanged; while heparin-binding affinity increased nearly 50%. Treatment with Type
VIII
arylsulfatase
caused a considerable decrease in gelatin-binding and a slight decrease in heparin-binding affinities. Nevertheless, desulfation by all three enzymes consistently resulted in a dramatic decrease of fibrin-binding affinity, ranging from 42.1% to 64.4%.

Three mammalian and eight non-mammalian arylsulfatases were investigated for their activities toward tyrosine-O-sulfate (TyrS) in peptides. None of the mammalian arylsulfatases exhibited detectable activities toward TyrS-containing peptides. Of the non-mammalian arylsulfatases tested, Types VII,
VIII
, and H-1, 2, and 5 displayed strong activity on endo-TyrS residues. The prokaryotic
sulfatase
, Type VI, was active only on free TyrS and N-terminal TyrS of Leu-enkephalin. All the sulfatases were active on p-nitrophenyl sulfate and p-nitrocatechol sulfate.

Sulfation plays a major role in regulating the activity of various neurosteroids, including pregnenolone and dehydroepiandrosterone. The present report describes the immunohistochemical distribution of two enzymes involved in the control of neurosteroid sulfation, hydroxysteroid sulfotransferase (HST) and unconjugated steroid enzyme
sulfatase
(
STS
), in the brain of the European green frog Rana esculenta during development. HST and
STS
immunoreactivity were detected from stage
VIII
-XII. At this early stage, HST-positive fibers were seen in the glomerular layer and the basal rhombencephalon. Subsequently, at stage XIII-XV, HST- and
STS
-immunoreactive fibers were vizualized in the accessory olfactory bulb. At stage XVI-XVIII,
STS
-positive cell bodies were observed in the periventricular region of the diencephalon. These observations indicate that the enzymes controlling sulfation of hydroxysteroids are expressed in the frog brain during development.
