We have isolated a clone from a EMBL3 constructed bovine genomic library using a PCR-generated DNA fragment as a probe covering-660bp to + 158 bp of boving alpha-sl casein gene. The results obtained from hybridization assay, restriction endonucleases map and partial DNA sequence analysis indicated that the cloned fragment spanned 5.7kb of the first part of the gene and 8.0 kb of its upstream flanking region. Some divergent restriction endonuclease recognition sites and a few mutations and deletions were found in exons, introns and 5'-flanking region of the gene we cloned when compared with the PCR-generated fragment and those which had been published [6,10]. All the deletions occurred at repeat sequences.

A positive clone was isolated from a bovine genomic library in EMBL3 with the probe of alpha-s1 casein gene cDNA. The results from both restriction endonuclease digestion and Southern hybridization probed with the upstream and downstream fragments from the termination codon of bovine alpha-s1 casein gene cDNA suggested that the clone contained 3.5 kb downstream and 10.2 kb upstream fragments from the termination codon of alpha-s1 casein gene. Some divergences in restriction endonucleases sites, a few deletions, and many point mutations were found in introns and exons when compared with those which were published.

Free polysomes were isolated from the livers of rats maintained on a 60% casein diet to induce ornithine aminotransferase mRNA. Ornithine aminotransferase-synthesizing polysomes were immunoadsorbed using monospecific, affinity-purified antibody and Staphylococcus aureus cells. Poly(A+)RNA prepared from these polysomes by oligo(dT)-cellulose chromatography was used as a template for the synthesis of double-stranded cDNA by reverse transcriptase. Using the deoxyguanosine-5'-triphosphate-deoxycytidine-5'-triphosphate tailing method and pBR322 as a vector, recombinant molecules were produced and used to transform Escherichia coli. Two clones containing DNA complementary to ornithine aminotransferase mRNA were identified by colony hybridization and hybrid-arrest translation. A partial restriction endonuclease map of the ornithine aminotransferase cDNA inserts was constructed which spanned 1066 base pairs.

Restriction of protein intake in the diet has been shown to produce a change in transcription activity as determined in vitro. Conditions for a reduced transcription activity were investigated with selective digestion of nuclei with micrococcus nuclease (EC 3.1.4.7). Liver nuclei from young male rats fed a diet containing either 20 or 3% casein for 6 days were digested with 1.3 microgram of micrococcus nuclease protein/mg of nuclear DNA (specific enzyme activity 15,000 or 150 units/mg of protein). Part of the chromatin-bound RNA polymerase activity was transferred from the 2,000 x g to the 102,000 x g pellet. Independent of the type of endonuclease use, the specific activity of chromatin-bound and soluble RNA polymerase I plus III was similar in the two groups of rats. After protein restriction RNA polymerase II activity was significantly diminished in the 2,000 x g pellet, and was unchanged in the 102,000 x g pellet. Heparin-stimulated and soluble RNA polymerase II activities were significantly reduced. Number and length of RNA chains synthetized by chromatin-bound RNA polymerase I plus III remained unchanged by dietary treatment. After a low protein diet, RNA polymerase II in the absence and presence of heparin synthesized an unchanged number of RNA chains with reduced length. A selective digestion of chromatin with micrococcus nuclease is needed to show the reduction in RNA polymerase II activity after protein restriction.

Aurintricarboxylic acid (ATA) is an endonuclease inhibitor which has been shown to block apoptotic cell death. We have now demonstrated that ATA is also an inhibitor of the Ca(2+)-activated neutral protease (calpain), a class of cytosolic enzyme that may also be activated during apoptosis. The two major calpain isoforms (mu- and m-calpain) were both inhibited by ATA with IC50's of 22 microM and 10 microM, respectively. The autolysis of purified mu-calpain was prevented by ATA in a concentration-dependent manner. Using casein zymography, it was found that the inhibition of mu-calpain by ATA was reversible. Finally, in a fetal rat cerebrocortical culture model of excitotoxicity, pre- and post-treatment of ATA (50 microM) reduced N-methyl-D-aspartate (NMDA)-induced spectrin breakdown and neuronal death, while application of ATA concurrent to NMDA challenge alone had no effect. This pattern of protection could not be explained by simple NMDA receptor antagonism. We thus propose that the neuroprotective effect of ATA could be in part due to its ability to inhibit calpain.

PCR-RFLP technique was applied to analyze correlation between the polymorphisms of CSN1 S2 (alpha(s2) casein), CSN3 (kappa casein) and beta-lg (beta-lactoglobulin) genes and milk performance in 69 individuals of Xinong Saanen dairy goat. The results showed that there was significant correlation between different genotypes of CSN1 S2 locus and milk yield:average milk yield of individuals with genotype FF was less than that of genotype NN (P < 0.05); the second lactation milk yield of individuals with genotype NN was over 90 kg higher than that with genotype FF (P < 0.01). This indicates that allele F of CSN1 S2 locus probably has significant negative effect on milk yield. The results of CSN3 gene digested with endonuclease Hind III cleavage showed that no significant difference of milk yield between genotype DE and genotype EE was detected in first, second, third and fourth lactation milk yield and average milk yield (P > 0.05). The results of CSN3 gene with endonuclease Taq I cleavage showed that no significant difference of milk yield among individuals with genotype TT, TC and CC was detected (P > 0.05). No polymorphism was detected in PCR products of CSN3 gene digested with endonuclease Hae III. The analysis of beta-lg gene's 5' flanking region (710 bp) by PCR-RFLP in Xinong Saanen dairy goat showed that milk yield of individuals with genotype AA was higher than that with genotype AB in second, third lactation milk yield and average milk yield (P < 0.05). The results implied that allele A of beta-lg gene's 5' flanking region is probably related to high milk yield.

K-casein gene was regarded as a candidate gene for milk production traits of cows. In this study, a 779 bp fragment of k-casein gene of Chinese Holstein was amplified by polymerase chain reaction (PCR), the polymorphisms of three loci of k-casein gene were detected by PCR-RFLP with restriction endonuclease Taq, Hind, Pst. After sequencing, T/C single nucleotide polymorphism (SNP) was identified at nucleotide 10 891C/A SNP was identified at nucleotide 10 927 and G/A SNP was identified at nucleotide 10 988 in exon4 of k-casein gene. Both alleles (A and B) of three loci were found in the population that showed low polymorphism. The gene frequencies of A and B were 86.03% and 13.97%, respectively. The genotype frequencies of AA, AB, and BB were 73.71%, 24.63%, and 1.66%, respectively. Statistical results of c2 test indicated that three polymorphism sites in the population fitted with Hardy-Weinberg equilibrium (P > 0.05). Meanwhile, the effect of polymorphism of k-casein gene on milk production traits was analyzed. The results indicated that in the three loci, the different genotype of k-casein gene had no significant influence on milk yield and milk protein percent (P > 0.05). The cows with genotypes BB and AB showed higher milk fat percent than those with genotype AA ( P < 0.05 ) ; with genotype AB showed higher fat protein ratio than those with genotype AA ( P < 0.05 ). The polymorphism of the three loci in the experimental population is closely linked. The conclusion is that k-casein B allele can be used as the molecular genetic markers of modifying milk fat percent in Chinese Holstein cows.

PCR-RFLP was used to genotype 87 Sarabi native cattle of north-western Iran for A and B alleles of kappa casein gene. A 350 bp length of exon 4 and intron 4 was amplified and digested with HinfI endonuclease. Samples were loaded on agarose gel (2%) and genotyped under UV light. Allele frequency of desirable B allele was 0.57. Stochastic simulation was used to generate milk yield trait for a population of 4950 females and 50 males for 15 overlapping generations. Population parameters included 1100 and 436 kg for average milk yield and phenotypic deviation, respectively; with heritability of 0.27. Additive and dominance effects of Kappa Casein gene were considered as 187.63 and 50.37 kg, respectively. Two methods were considered for selection of males based on the first phenotypic record of their dams (PAS) or molecular information of each male, individually (GAS). Females were always selected on their first phenotypic record. Although, there was a significant difference between polygenic and major gene genetic response between two methods after the 5th generations, but there was no significant difference for the sum of polygenic and major gene response. After 15 generations of selection there was no significant difference between inbreeding coefficient under two methods. Selection plan for males based on one single major gene had no advantage over the conventional selection based on dam record in native Sarabi breed.

The genetic polymorphism of the goat Kappa casein was investigated in Tunisian goats. Blood samples were collected from local goat breeds. Samples of genomic DNA were obtained from leukocytes of 175 dairy goats and regions of interest in the gene were amplified by Polymerase Chain Reaction (PCR) and then evaluated in agarose gel. For a better characterization of the single nucleotide polymorphism, a PCR-Restriction Fragment Length Polymorphism was performed employing the endonuclease DNA amplification using 459 bp primers. The PCR products of primers (459 bp) digested by restriction enzyme Alw44I produced two fragments of 459 and 381 bp. The Kappa casein allelic variants in tested animals revealed different genotypes, two of them were homozygous: AA or BB, AC or BC and CC. Genotypic frequencies were 12.5, 60.5 and 27% for AA or BB, CC and AC or BC, respectively. Identification of different variants of the Kappa casein can be used for the improvement and conservation of Tunisian local goats.
