accessibility to DNA in the nuclei of A431 human carcinoma cells is increased within 90 min by nontoxic nanomolar levels of okadaic acid, known to inhibit protein phosphatase 2A. This genomic hypersensitivity was partly enhanced by joint treatment with epidermal growth factor and okadaic acid but did not appear without the latter. Nuclei with greater DNA susceptibility showed a decrease in M(r) 80,000 DNA binding protein doublet specific for dAT-rich sequences concurrent with the "apparent" hyperphosphorylation of a M(r) 70,000 nuclear

Human endogenous retroviral element S71 had previously been shown to contain gag- and pol-related regions and a 3' LTR-like sequence. The nucleotide sequence of S71 was determined and compared with the corresponding regions of SSV and its helper virus SSAV. The 1.48-kb S71 gag region consists of
matrix protein
p15 (MA)-, capsid protein p30 (CA)-, and nucleocapsid protein p10 (NC)-related sections and the 1.82-kb pol region of tether, RNase H (RH), and
endonuclease
/integrase (IN) sections. The S71 nucleotide sequence contains a 167 amino acid open reading frame encompassing MA. The boundaries of the S71 element are delimited by direct repeats and the entire element is 5.4 kb long. Similarity between S71 and the v-sis-bearing, defective SSV provirus also covers overall structural organization, including the presence of presumably nonretroviral sequences. Both the gag and the pol regions of S71 contain sequences highly conserved in numerous retroviruses. Phylogenetic analysis with conserved CA, RH, and IN sequences showed that of all other (C-type) human retroviral elements available for comparison, S71 is most closely related to infectious primate and murine retroviruses. This suggests that S71 represents a phylogenetic subgroup of its own. In addition we identified short ranges of conserved amino acid sequences within C-type retroviral gag and pol genes sufficient for phylogenetic analysis. Use of these may facilitate large-scale phylogenetic evaluation of C-type retroviral elements and allow rapid classification of new elements.

The purpose of this study was to express the
matrix protein
of rabies virus in baculovirus expression system and prepare its polyclonal antibody. Using the total RNA of RABV strain BD06 as a template, RT-PCR technique was utilized to amplify the sequence of M gene, which were then inserted into shuttle vector pFastbac I to construct the recombinant vector pFastbac I-M. After identification using the double restriction
endonuclease
cleavage method, the recombinant vector pFastbac I-M were transformed into the competent E. coli DH10 Bac to construct the recombinant expression vector Bacmid-M, which were transfected into Sf9 cells mediated by lipofectamine 2000 to obtain the recombinant baculovirus AcMNPV-M. The mice anti-His monoclonal antibody, rabbit anti-RV positive serum and canine anti-RV positive serum were used in Western Blot assays to identify the expression and reactogenicity of the recombinant. The recombinant M protein were purified under denaturing conditions using the nickel iron affinity chromatography column, then used to immunize the New Zealand White rabbit to prepare its polyclonal antibody. Western Blot assay and FAVN assay were used to validate the polyclonal antibody. Our results showed that the M protein of RABV were successfully expressed in baculovirus expression system,of which molecular weight was of about 25kD;the recombinant M protein has a good reactogenicity and immunogenicity; the rabbit polyclonal antibody prepared by purification of M protein could react with the M protein of RABV strain BD06,SRV 9,CVS-24,ERA,PV2061 and aG. Undoubtedly, the successfully preparation of both recombinant M protein and its polyclonal antibody support a material foundation for further study on the properties of M protein of RABV.
