Herpes simplex virus type 1 isolates from 63 patients with herpetic keratoconjunctivitis were investigated. For the purpose of detecting strain differences neurovirulence was determined in mice, and viral DNA's were analyzed by digestion with EcoRI and Hind III restriction endonucleases. Neurovirulence differed by a factor up to 10(5) between individual strains and proved to be independent of the ratio between infectious and noninfectious particles in the stocks used. The endonuclease cleavage patterns revealed differences of the viral DNA structure which permitted us to distinguish seven clusters of strains. Correlations between neurovirulence and DNA markers could not be established nor could correlations be found between these markers and features of disease. The main reason for not finding a relationship in the latter case may be attributed to the significant role of host factors in the course of herpetic diseases.

An outbreak of epidemic keratoconjunctivitis occurred at the department of ophthalmology of a hospital in Yokohama, involving 14 inpatients, 12 outpatients, and 2 doctors. Adenovirus type 37 (Ad-37) was isolated from the conjunctival swab in 12 of 18 cases. In neutralization tests, the isolates showed some cross-reaction with adenovirus type 19 (Ad-19). The Ad-37 isolates were indistinguishable from each other and from the prototype Ad-37, but distinct from the prototype Ad-19 in the restriction endonuclease analysis of viral DNA.

The inverse dependence of activity of restriction endonuclease SsoII preparations on the number of low molecular mass plasmids of Shigella sonnei transforming Escherichia coli recipient cells producing the enzyme has been shown. Escherichia coli strain producing efficiently one of two Shigella sonnei 47 restriction endonucleases SsoII has been isolated. The producer strain harbours two of the nine Shigella sonnei 47 plasmids. One of them P4 codes for SsoII+ phenotype while the other P9 determines the plasmids conjugation transfer. Biochemical and physiological characteristics of the producer strain XS13 are identical to the ones of the recipient Escherichia coli strain PS200. XS13 is unable to induce keratoconjunctivitis in guinea pigs in pathogenicity test.

DNA restriction endonuclease analysis for genome typing of adenovirus (Ad) DNA was carried out on a total of 65 Ad isolates including serotypes Ad4, Ad7, Ad8, Ad11, Ad19, and Ad37 from patients with epidemic keratoconjunctivitis and acute conjunctivitis obtained in Japan from 1982 to 1986, Australia from 1973 to 1986, and the Philippines in 1984. All 4 isolates of Ad7 in Australia were Ad7b. Four of 6 Ad11 isolates obtained in Japan were typed as Ad11 prototype (Ad11p), and the remaining were identified to be new genome types, designated tentatively as Ad11c and Ad11d. An isolate of Ad11 obtained in Australia was typed as Ad11c. Nine Ad8 isolates in Australia and in the Philippines were typed as Ad8p, but 11 Ad8 isolates in Japan were Ad8b. Thirteen Ad19 isolates were identified as Ad19a. All 3 isolates of Ad37 in Japan and three isolates in Australia before 1982 were typed as Ad37p, however, 5 isolates in Australia after 1983 were identified as a new genome type, designated as Ad37d. In Japan, 10 isolates of Ad4 were identified as Ad4a.

Forty-one strains of adenovirus type 19/37 (Ad19/37) mainly isolated from patients with keratoconjunctivitis or conjunctivitis between 1974 and 1984 were re-evaluated by serum neutralization (SN), haemagglutination inhibition (HI) and DNA restriction analysis. Of 19 isolates which were neutralized to high titre by antiserum prepared against prototype Ad19, 5 showed cross-reactivity with 32-64 units of Ad37 antiserum, while of 22 strains neutralized by high titre by Ad37 antiserum, 3 showed cross-reactivity with 32 units of Ad19 antiserum. By DNA restriction analysis, all Ad19 isolates were identical to each other and to Ad19A virus. Using endonuclease Bgl 1, three variants were observed among the Ad37 isolates.

A new human adenovirus has been isolated from patients with keratoconjunctivitis and/or genital infection since 1976. This adenovirus, designed candidate adenovirus 37 (Ad 37) is serologically distinct but related to Ad 10, 13, 19, and 30 (see the accompanying paper by de Jong et al). SDS-Polyacrylamide gel electrophoresis of Ad 37 virion polypeptides showed that this adenovirus is a member of subgroup D. DNA restriction endonuclease analysis of DNA from Ad 37 and related serotypes belonging to subgroup D showed that Ad 37 is a new genome type belonging to subgroup D but clearly distinct from the 20 serotypes classified into this subgroup.

We studied the restriction endonuclease cleavage patterns of DNAs of adenovirus type 8 (Ad 8) isolated from epidemic keratoconjunctivitis cases. DNAs of 25 Ad 8 isolates collected during the period from 1975 to 1981 in Sapporo were subjected to enzymatic cleavage with PstI, BamHl , HindIII and SalI. On the basis of the cleavage patterns, the isolates were divided into two subtypes, A and B, both different from Trim strain used as the Ad 8 prototype strain. Subtype A was prevalent in the period from 1975 to 1978, while subtype B in the period from 1976 to 1981.

An outbreak of acute keratoconjunctivitis in an ophthalmology department affected 15 patients and seven members of staff and necessitated temporary closure of the unit. Adenovirus (Ad) was isolated from eye swabs taken during the outbreak, but typing of the isolates by virus neutralisation assay proved unsatisfactory, wrongly assigning the isolate to serotype 10. A robust and rapid method for preparing microgram amounts of Ad DNA from infected cells was devised and used as the basis of a non-radioactive method for routine genotyping of adenovirus clinical isolates. All isolates associated with the outbreak, and one from a patient presenting at a nearby hospital during the outbreak, were found to have restriction endonuclease digestion patterns characteristic of Ad 37.

Adenovirus strains of a new type were isolated from patients with epidemic keratoconjunctivitis. They were not completely neutralized by any antiserum against adenovirus prototypes. PCR followed by restriction endonuclease analysis demonstrated that they were type 8. PCR followed by sequencing revealed a high homology rate between them and type 9.

Adenovirus are important pathogen primarily associated to respiratory infections of children and military personnel, even though it is also associated to cases of conjunctivitis and keratoconjunctivitis. We analyzed respiratory secretion collected from subjects with and without respiratory infection symptoms, being 181 civilians and 221 military subjects. The samples were inoculated in HEp-2 and/or A549 tissue cultures for viral isolation. Samples presenting cytopathogenic effect (CPE) in any tissue culture were tested by a polymerase chain reaction (PCR) assay to confirm adenovirus isolation. The isolates confirmed as adenovirus were further analyzed by restriction endonuclease assay for determination of viral species. Three isolates were identified as specie A (two from civilian and one from military), one isolate from military was identified as specie C, and one isolate from civilian was identified as specie D. For two isolates the specie could not be identified.
