Tissues from the cerebral cortex, liver and myocardium of a patient with Lafora disease were obtained at autopsy and were studied biochemically. 1. Glucose content in the myocardium and liver was almost nil while that in the controls was 0.66 mg/g wet weight in the former and 8.80 mg/g wet weight in the latter.
Glycogen
content in the cerebral cortex and myocardium was about 10 and 3 times more than in controls. 2. Polyglucosan extracted from the cerebral cortex, liver and myocardium had a longer exterior glucose chain than that in the liver of the control but a normal, alpha or beta 1,4-glucosidic linkage was observed. 3. The activities of
glucose-6-phosphatase
and amylo-1,6-glucosidase in the cerebral cortex, liver and myocardium were well preserved. The activities of acid maltase in the three organs mentioned above and of neutral maltase in the myocardium were elevated twice and one and half times more than the control. Phosphorylase levels in the myocardium were extremely small, while in the cerebral cortex and liver normal activities were observed. In light of these findings, glycogen metabolism in Lafora disease is discussed.

In 28 dogs the distal articular cartilage of the femur was removed and the regenerating articular surface on the 70th postoperative day was studied histochemically for hexokinase,
glucose-6-phosphatase
, phosphohexose-isomerase, fructose-1, 6-diphosphatase, aldolase, glyceraldehyde-3-phosphate dehydrogenase, lactate dehydrogenase, lactate dehydrogenase isoenzymes, phosphoglucomutase, phosphorylase, glycogen synthetase, UDP--glucose dehydrogenase, and UDP-glucuronic acid-4-epimerase. The articular surface consisted of fibrous tissue and of cartilage islets. The latter contained cells differentiating into cartilage and young chondrocytes. The glycolytic enzymes reacted positively in the regenerative articular surface. Enzyme activities were higher in the cells (particularly the chondroblasts and young chondrocytes) of the cartilage islets than in the connective tissue. In the cells differentiations into cartilage, beside the LDH isoenzymes characteristic of glycolysis, a significant LDH1 and LDH2 activity was observed. At the same site the presence of fructose-1, 6-diphosphatase-activity could be assumed, but there was no
glucose-6-phosphatase
activity.
Glycogen
synthesis proceeded in the cells of the cartilage islets and UDP-glucuronic acid-4-epimerase activity was observed in the differentiated cells. UDP-glucose dehydrogenase activity was positive in every section of the articular surface.

The experiment was carried out on rats, which were divided into three experimental and one control groups. The experimental animals were intraperitoneally injected with furfural in the dose of 58 mg/kg body weight for 30 days. In the liver samples obtained at autopsy, apart from routine staining with hematoxylin and eosin, estimation of the activity of the following enzymes was made: succinic dehydrogenase. Glycogen
and protein concentrations and the activities of liver glycogen metabolic enzymes were measured in 22 children aged 4 to 15, suffering from extrahepatic portal hypertension. Glucose-6-phosphatase, amylo-1,6-glucosidase, fructose-1,6-diphosphatase, phosphorylases alpha and beta, phosphoglucomutase, and phosphohexose isomerase levels were analyzed. Liver biopsy specimens obtained by surgical marginal biopsy were used in the study. No or drastic reduction of phosphorylase alpha activity and reduction of glycogen concentration and glucose-phosphatase activity were found characteristic of extrahepatic hypertension. Analysis of correlations of the findings has demonstrated a medium correlation in 4 cases and a strong correlation between the findings in 1 case, the possibility being estimated as 0.95-0.99. The highest number of correlations was observed with phosphorylase alpha and
glucose-6-phosphatase
(3 correlations). Liver blood stream impairments result in injury to one of its main biochemical functions, i.e., the maintenance of blood glucose homeostasis, this leading to reduction of the adaptation potential of the body; this should be borne in mind when planning therapeutic measures for patients with extrahepatic hypertension.

storage diseases (GSD) include inborn errors in glycogen synthesis and degradation which, like most metabolic diseases, evades any therapeutical concept up to now. Nevertheless, in a subgroup of glycogenoses, characterized by functional absence of the key glycogenolytic enzyme
glucose-6-phosphatase
(GSD-1), dietary treatment is able to ensure almost normal development of affected patients. The principle of treatment consists of oral application of raw cornstarch, which is hydrolyzed slowly in the intestinum resulting in normoglycemia over a period of several hours. Thus, nighttime nasogastric tube-feeding, the standard dietary regimen in GSD-1, could be avoided. Based on our experiences, this therapeutic approach, which has proven successful up to now only in elder children and adults, can be recommended even in children under 2 years of age, if the individual response to an oral starchload has been examined prior to the starch diet.

The results indicate that utilization of carbohydrate and lipid substrates for thermogenesis is intensified in cold-adapted rats.
