The effects of added polyamines on carbamylphosphate (carbamyl-P):glucose phosphotransferase and glucose-6-phosphate (Glc-6-P) phosphohydrolase activities of rat hepatic D-Glc-6-P phosphohydrolase (
EC 3.1.3.9
) of intact and detergent-treated microsomes have been investigated. With the former preparation, in the presence of 1.4 mM phosphate substrate and 90 mM D-glucose (phosphotransferase), 1 mM spermine, spermidine, and putrescine activated Glc-6-P phosphohydrolase 67%, 57%, and 35%, respectively. Carbamyl-P:glucose phosphotransferase, under comparable conditions, was activated 57%, 34%, and 18%. NH+4 (0.25--5.0 mM) produced at best but a minor activation (0--14%), while poly(L-lysine) (Mr = 3400; degree of polymerization 16) equimolar relative to other polyamines with respect to ionized free amino groups activated the hydrolase 358% and the transferase 222%. Treatment of microsomes with the detergent deoxycholate reduced, but did not abolish, polyamine-induced activation. The stimulatory effects of polyamines persisted in the presence of excess catalase, indicating their independence from
H2O2
formation; and were eliminated in the presence of Ca2+. Kinetic analysis revealed that all tested polyamines decreased the apparent Michaelis constant values for carbamyl-P and Glc-6-P, but had no effect on the Km for glucose. Poly(L-lysine) increased the V value for both Glc-6-P phosphohydrolase and apparent V values for phosphotransferase extrapolated to infinite concentrations of either carbamyl-P or glucose. The other tested polyamines elevated only this last velocity parameter. It is proposed that a major mechanism by which polyamines activate
glucose-6-phosphatase
-phosphotransferase is through their electrostatic interactions with phospholipids of the membrane of the endoplasmic reticulum of which this enzyme is a part. Conformational alterations thus induced may in turn affect catalytic behavior. It is suggested that polyamines, or similar positively charged peptides, might participate in the cellular regulation of synthetic and hydrolytic activities of
glucose-6-phosphatase

A developmental block is induced by phosphate in rat embryos at the late two-cell stage. The present study was designed to examine the energy metabolism of rat two-cell blocked and non-blocked embryos. Enzyme activity was measured in individual embryos by histochemical techniques. The activities of malate dehydrogenase, isocitrate dehydrogenase, lactate dehydrogenase, pyruvate dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase, glutamate dehydrogenase, glucose-6-phosphate dehydrogenase,
glucose-6-phosphatase
, and phosphorylase did not differ among non-blocked and blocked embryos. However, the activity of succinate dehydrogenase was significantly decreased in blocked embryos compared with non-blocked embryos. In blocked embryos, cytochrome oxidase activity was distributed homogeneously, but was located at the perinuclear region in non-blocked embryos. Active mitochondrial organization was visualized using the fluorescent probe rhodamine 123 and laser scanning confocal microscopy. In both non-blocked and blocked embryos, mitochondria were distributed homogeneously. The concentration of
H2O2
measured fluorometrically in embryos cultured without phosphate did not change significantly during the culture period, but decreased in embryos cultured with phosphate. The timing corresponded to the occurrence of the two-cell block. In summary, these results suggest that the developmental block in rat two-cell embryos is induced by disturbance of mitochondrial energy metabolism.

In the present study, we evaluated the protective activity of an aqueous extract of Artemisia absinthium against CCl4-induced hepatic damage in rats. The protective activity of this extract at three doses (2.5, 5, and 10 ml/ kg, once orally) against CCl4-induced oxidative damage (1.5 ml/kg, once intraperitoneally) in rats was analyzed. Various blood and tissue biochemical studies were performed, and the administration of the toxicant significantly altered blood biochemical variables. Hepatic lipid peroxidation (LPO) was significantly elevated, whereas glutathione (GSH) level was considerable depleted after intoxication. Remarkable decreases in the activities of adenosine triphosphatase (ATPase) and
glucose-6-phosphatase

G-6-Pase
) after intoxication were observed. Treatment with all three doses reversed altered tissue biochemical values, but the greatest protection was observed at the lowest dose (2.5 ml/kg). The results of this study show that A. absinthium induces strong hepatoprotective activity. It decreased the hexobarbitone-induced sleep time and improved cholerectic activity (bile flow and bile solids) and excretory capacity, and it also stimulated bile secretion. The potent antioxidant activity of A. absinthium was indicated by scavenging effects on 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and hydrogen peroxide (
H2O2
). Thus, be considered for use in reducing hepatic damage and may serve as an alternative medicine in hepatic etiologies.
