Annual nitrogen (N), phosphorus (P), and potassium (K) flows in agriculture in The Netherlands were identified and quantified in 1990, with special emphasis on pig production. Also, the effects that various management strategies in pig production have on NPK emission in 1990 were compared using a static deterministic simulation model. Ammonia emission from pig production in 1990 (60.9 Gg N) exceeded the defined target for the year 2000 (12.7 Gg N). Measures that affect volatilization of ammonia directly (i.e., introduction of low-emission stables, manure storage facilities, or manure application techniques) reduced ammonia emission most effectively. These measures, however, should be combined with a reduction in application of artificial N fertilizer to avoid an increase in N losses through leaching, run-off, or denitrification. Targets for ammonia emission in the year 2010 require a reduction in the pig population of 24 to 62%, in addition to implications of measures described in this article. National NPK losses in 1990 through leaching, run-off, or denitrification, predicted at 223.5 kg/ha for N, 32.7 kg/ha for P, and 67 kg/ha for K, exceeded government targets for the year 2010 (185 kg N/ha; 8.7 kg P/ha; norm not set for K). Reducing application of artificial NPK fertilizer reduced national NPK losses most effectively. For P, use of phytase and feeding pigs in accordance with their P requirements is required, in addition to limited use of artificial P fertilizer to meet targets for the year 2010. Hence, from an environmental point of view, pig production in The Netherlands is limited primarily by ammonia emission targets for the year 2010.

An experiment was conducted to determine whether broiler litter concentration of N and P and equilibrium NH3 gas concentration can be reduced by reducing dietary CP and P levels and supplementing with amino acids and phytase, respectively, without adversely affecting bird performance. Equilibrium NH3 gas concentration above the litter was measured. The experiment was divided into a starter period (1 to 21 d) and grower period (22 to 42 d), each having two different CP and P levels in a 2 x 2 factorial arrangement. The CP treatments consisted of a control with a mean CP of 204 and 202 g/kg for starter and grower periods, respectively, and a low CP diet with means of 188 and 183 g/kg, respectively, but with similar amino acid levels as the control. The P treatments comprised starter and grower control diets containing means of 6.7 and 6.3 g/kg P, respectively, and low P treatment means of 5.8 and 5.4 g/kg P supplemented with 1.0 g/kg phytase. Reducing starter diet CP by 16 g/kg reduced weight gain by 3.5% and, hence, body weight at 21 d of age, but did not affect feed intake or feed efficiency. Reducing P did not affect feed intake and weight gain, but improved feed efficiency by 2.0%. Responses in feed intake and efficiency to CP depended on the level of dietary P. For the grower period there were no significant differences in feed intake, weight gain, and feed efficiency, nor in body weight at 42 d of age, after correcting for 21-d body weight, between CP and P treatments. There were significant (P < 0.001) reductions in litter N and P concentrations, but not equilibrium NH3 gas concentration, moisture content, or pH, for low CP and P diets. Mean equilibrium NH3 gas concentration was 63 ppm. Litter N concentration was reduced 16.3% with the low CP diets, and litter P by 23.2% in low P treatments. The results suggest that dietary manipulation shows merit for reducing litter N and P concentrations while maintaining acceptable production performance from broilers.

Rice bran protein isolate (RBPI) containing approximately 92.0% protein was prepared from unstabilized and defatted rice bran using phytase and xylanase. The yield of RBPI increased from 34% to 74.6% through the use of the enzymatic treatment. Nitrogen solubilities of RBPI were 53, 8, 62, 78, 82, and 80% at pHs 2.0, 4.0, 6.0, 8.0, 10.0, and 12.0, respectively. Differential scanning calorimetry showed that RBPI had denaturation temperature of 83.4 degrees C with low endotherm (0.96 J/g of protein). RBPI had similar foaming properties in comparison to egg white. But emulsifying properties of RBPI were significantly lower than those of bovine serum albumin. The result of amino acid analysis showed that RBPI had a similar profile of essential amino acid requirements for 2-5-year-old children in comparison to that of casein and soy protein isolate.

Government targets for ammonia emission and for N and P loss per hectare (ha) of agricultural land were used to assess carrying capacity for poultry production in The Netherlands with data from 1990. In addition, the effect of alternative management strategies on carrying capacity was determined. Ammonia emission from poultry production in 1990 [20.5 gigagrams (Gg) N] exceeded the target for 2000 (i.e., 6.9 Gg N). Targets defined for 2000 and 2010 (i.e., 4.6 Gg N) can be achieved, however, without reducing poultry numbers, assuming national introduction of measurements studied. Measures that reduced ammonia emission directly, i.e., introduction of low-emission housing or manure application techniques, were most effective. In 1990, N and P losses equalled 215 kg/ha for N and 31 kg/ha for P. The N loss was slightly lower than the target for 2000 (219 kg N/ha) but exceeded the target for 2010 (144 kg N/ha). Reduction of application of artificial N fertilizer, however, reduced N loss effectively from 215 to 22 kg/ha. National P loss in 1990 exceeded the target for 2000 (15.3 kg P/ha). Reduction of application of artificial P fertilizer reduced P loss most effectively from 31 to 14 kg/ha. To achieve the target for 2010 (8.7 kg P/ha), additional reduction in P excretion by poultry is required. This reduction can be achieved by use of phytase in layer and broiler feed and by use of a coarse Ca source in layer feed. Unlike pig production, carrying capacity for poultry production in The Netherlands is not limited by governmental targets for acidification, eutrophication, or drinking water contamination.

A significant mechanism of arsenate toxicity to Pisum sativum is interference with its mineral nutrient balance. This conclusion is supported by assessments made after exposing P. sativum L. cv. "Phenomen" for 12 days to 12.5, 20.8, and 33.3 mg, and for 32 days to 7.5, 22.1, 36.7, and 73.3 mg of sodium arsenate/kg dry wt soil in the greenhouse. At 20.8 mg of arsenate, mobilization of manganese from the cotyledons was significantly increased and that of zinc decreased. Nitrogen accumulated in the roots. On Day 32, at 22.1 mg of arsenate, magnesium, zinc, and manganese contents of the roots increased, but that of phosphorus of the shoot decreased. The distribution pattern and the ratios between individual elements were severely altered. Relatively more arsenic accumulated from the low than the high soil concentrations. Growth of the shoot was more affected than that of the roots. After a 32-day exposure, chlorophyll content of the leaves increased, but the chlorophyll a/b ratio decreased. On Day 12, at 12.5 mg and 20.8 mg of arsenate, in vivo phytase activity was 64 and 66% that of the controls, respectively.

Three experiments were conducted to evaluate P bioavailability, growth performance, and nutrient balance in pigs fed high available P (HAP) corn with or without phytase. The bioavailability of P in normal and HAP corn relative to monosodiumphosphate (MSP) for pigs was assessed in Exp. 1. In a randomized complete block design, 96 pigs (average initial BW 9.75 kg) were fed eight diets for 28 d. The reference and test diets were formulated by adding P as MSP, HAP, or normal corn at 0, 0.75, or 1.5 g/kg to a corn-starch-soybean meal basal diet (2.5 g/kg P) at the expense of cornstarch. Plasma inorganic P concentration responded linearly (P < 0.05) to supplemental P intake. Estimates of P bioavailability from HAP andnormal corn when plasma P was regressed on supplemental P intake were 46 and 33%, respectively. In Exp. 2 and 3, pigs were fed corn-soybean meal-based diets containing HAP corn or normal corn and 0 or 600 units of phytase per kilogram in a 2 x 2 factorial arrangement (two corn sources and two levels of phytase). In Exp. 2, 48 crossbred pigs (barrow:gilt, 1:1) averaging 9.25 kg were used to evaluate growth performance. There were no detectable interactions between corn source and phytase for any of the performance criteria measured. Pigs receiving normal corn had the lowest (P < 0.05) BW and rate of gain. Feed efficiency was lower (P < 0.05) in pigs fed normal compared with those fed the HAP corn phytase-supplemented diet. In Exp. 3, 24 crossbred barrows averaging 14.0 kg were used to evaluate nutrient digestibility. There were no detectable interactions between corn and phytase for any of the N and Ca balance criteria. Nitrogen and Ca retention were improved in pigs receiving HAP corn with phytase (P < 0.05). Retention and digestibility of P was lowest (P < 0.01) for pigs on normal corn diet without phytase. The percentage of P digested and retained was improved and fecal P excretion lowered (P < 0.05) by feeding HAP corn. The results of this study indicate that the bioavailability and balance of P in HAP corn is superior to that of normal corn. The addition of 600 phytase units (Natuphos 600, BASF) to HAP corn-based diets further improved P digestibility and reduced P excretion in pigs.

Experimental data on phytate phosphorus utilisation by ruminants are scarce. The aim of this study was to estimate the phytase activity of rumen micro-organisms when phytate phosphorus supply is high. A semi-continuous culture system fermentor (RUSITEC) was used. The inoculum was obtained from eight goats fed on either high or low forage level diets. Experimental buffers only differed by the nature of phosphorus monosodium phosphate vs. corn sodium phytate. The nylon bags containing 15 g DM of substrate were removed after a 48-hour incubation period. The system was maintained for 15 days: 5 days for adaptation, in order to obtain a steady state, and 10 days for sampling and recording. No significant differences were observed for DM digestibility, gas production, pH, N-NH3, and SCFA for the different treatments. Bacterial efficiency of phytate phosphorus utilisation was significantly higher (p < 0.001) with organic P, but remained lower than the data usually reported in the literature. These results may be explained by the relative saturation of bacterial phytase activity when the buffer contains a high level of phytate phosphorus.

Ammonia (NH3) losses from swine manure contribute to odor problems, decrease animal productivity, and increase the risk of acid rain deposition. This study was conducted to determine whether aluminum chloride (AlCl3) or dietary manipulation with phytase could decrease relative NH3 losses from swine manure. Twenty-four pens of nursery pigs were used in two trials, and the pigs were fed normal or phytase-supplemented (500 IU/kg) diets. Aluminum chloride was added to manure pits (1.9 x 1.2 x 0.5 m) under each pen at 0, 0.25, 0.50, or 0.75% (vol:vol) of final manure volume. Manure pH and NH3 losses (measured by relative NH3 flux) were determined twice weekly. The addition of AlCl3 at 0.75% decreased (P < 0.05) manure pH from 7.48 to 6.69. Phytase decreased (P < 0.05) manure pH to 7.07 compared with 7.12 in the normal diet manure. Aluminum chloride administered at 0.75% without phytase reduced (P < 0.05) relative NH3 losses 52% for the entire 6-wk period. Relative NH3 losses were decreased (P < 0.05) from 109 mg of NH3/(m2 x h) in pens containing pigs fed the normal diet without AlCl3 to 81 mg of NH3/(m2 x h) in pens housing pigs administered the phytase diet, a 26% reduction. When the phytase diet and 0.75% AlCl3 additions were used in combination, relative NH3 losses were reduced (P < 0.05) by 60% compared with pens of pigs fed the control diet without AlCl3. Decreases in manure pH were likely responsible for the observed reduction in NH3 losses. Multiple regression was performed with relative rates of NH3 losses as the dependent variable and rate of AlCl3 addition, diet, and manure pH as independent variables. The model was tested using a stepwise regression (P < 0.001), and results indicated that the most important factors determining NH3 losses were manure pH and diet. However, the contribution of AlCl3 cannot be discounted. When manure pH was regressed against AlCl3 and dietary phytase, AlCl3 levels accounted for 64% of the variation in manure pH (P < 0.001). Dietary manipulation with phytase and application of AlCl3 to manure are promising management practices for the reduction of NH3 from swine facilities.

In our previous study, it was determined that phytase produced by Aspergillus oryzae plays an important role in supplying phosphate to yeast in the process of making sake. During koji making, two types of phytase (Phy-I and Phy-II) are produced. The purified phytases have high thermal and pH stability, in comparison to phytase purified from a submerged culture (ACP-II). In the present study, Phy-I and Phy-II retained their activities for 45 h. The NH2-terminal sequence of Phy-1, which is eight amino acids in length, was identical to that of ACP-II, but the molecular weights of these two forms, as estimated by SDS-PAGE, were quite different from each other (Phy-I, 120 kDa; ACP-II, 58 kDa). From the NH2-terminal amino acid sequence analysis of the predominant phytase (Phy-II), a molecular weight of 116 kDa was expected to reflect a new type of phytase produced only in koji culture. The substrate specificity of Phy-II was sufficiently broad that it hydrolyzed not only phytic acid and p-nitro phenyl phosphate, but also glucose 6-phosphate and glycerol 1-phosphate. In the process of making koji, Phy-I was produced at an early stage, followed by Phy-II; with both phytases being thought to function to hydrolyze phytic acid cooperatively.

Three experiments were conducted to evaluate the compositional and nutritional values of corn grains [phytase transgenic corn (PTC) and isogenic conventional corn (CC)] and compare the efficacy of corn-based phytase and extraneous microbial phytase for enhancing the utilization of phytate phosphorus (P) in single corn or corn-soybean mixed meals (corn:soybean = 2.5:1, wt:wt) fed to roosters. Following a 48-h fasting period, 16 roosters were given 50 g of each sample via crop intubation and excreta were collected for 48 h. Nitrogen-free and phosphorus-free diets were used to evaluate endogenous amino acid and endogenous P losses, respectively. Chemical composition was not different between PTC and CC, whereas the phytase content for PTC was greater than CC (8,047 vs. 37 FTU/kg of corn, DM basis; P < 0.001). No difference was observed in the TME and true amino acid availability values between the PTC and CC in roosters. The true P utilization for PTC was greater than CC (37.92 vs. 55.85%; P < 0.001), and CC and PTC contained 0.13 and 0.19% available P (AP, DM basis; P < 0.001), respectively. There was no difference in P utilization (72.76 vs. 70.23%; P > 0.05) between roosters fed PTC and extraneous microbial phytase in equivalent FTU/kg of diets. The results of this study indicated that the chemical composition, TME, and true amino acid availability in PTC are essentially equivalent to that in CC, and the true P utilization for roosters is higher in PTC than in CC. Corn expressing phytase is as efficacious as equivalent microbial phytase when supplemented in corn-soybean diets for chickens.
