The kinetic characteristics (Vmax and Km) of membrane-bound and serum 5'-nucleotidases from rats of different age groups are studied. The age-related increase of Km parameter in plasma membranes of fatty tissue and liver demonstrated a lower enzyme affinity to the substrate in ageing membranes. The specific activity of 5'-nucleotidase in blood serum of aged animals was below that in young ones, while Km value remained stable. Tissue differentiation of the catalytic properties of the enzyme and their aged-related characteristics may be to a considerable extent determined by the physicochemical state of cell membranes.

Neuropathy target esterase (NTE) is the suggested "target" molecule involved in the initiation of organophosphorus-induced delayed polyneuropathy. Sciatic nerve NTE was separated into particulate (P-NTE) and soluble (S-NTE) fractions by ultracentrifugation at 100,000 g for 1 h in 0.32 M sucrose and compared with the corresponding brain extract. Total sciatic NTE activity was 80-100 nmol/min/g tissue from which 50-60% was recovered in the soluble supernatant fraction and the remaining 40-50% in the pellet fraction. About 90% of brain tissue activity (approximately 1,800 nmol/min/g tissue) was recovered as P-NTE. A similar distribution was obtained when more drastic centrifugation without sucrose was performed. P-NTE and S-NTE were distributed with the membrane and cytosolic markers assayed, respectively, glucose-6-phosphatase, Na+,K(+)-ATPase, 5'-nucleotidase, phospholipids, and lactate dehydrogenase. When the pH during the centrifugation was increased from 6.4 to 11, recovered P-NTE activity decreased from 1,750 to 118 nmol/min/g tissue for brain and from 31 to 12 nmol/min/g for sciatic nerve. However, S-NTE activity and total nonfractionated control activity were only slightly affected by the same pH treatment. The distribution pattern encountered may be better understood as representing two different proteins than an equilibrium between soluble and membrane-bound portions of a single protein, with P-NTE activity depending on a membrane factor from which it is separated through fractionation at high pH.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of insulin on human red blood cells was investigated, both on intact cells and on isolated plasma membranes, testing the responsiveness of membrane-bound enzymes--such as (Na+-K+)-ATPase and 5'-nucleotidase--as well as the ouabain binding and ionic fluxes. It appears that insulin stimulates Na-pumping mechanisms increasing (Na+-K+)-ATPase activity through an enhanced availability of pumping sites, as can be inferred from the increased ouabain binding. The apparent unresponsiveness of fluorescence polarization parameters, following insulin treatment of isolated plasma membranes and intact cells, rules out--at present--an involvement of membrane lipid fluidity in the mechanism of action of insulin on human erythrocytes.

A 30-day ingestion of gamma-hexachlorocyclohexane (lindane) by carp (Cyprinus carpio) induced hypoglycemia without activation of two hepatic gluconeogenesis enzymes (fructose diphosphatase, EC 4.1.2.13, and glucose-6-phosphatase, EC 3.1.3.9) and hyponatremia and variations in muscle plasmic membrane-bound enzymes (especially cholinesterases, EC 3.1.1.7). After 109 days carps exhibited a decrease in natremia but no significant hypoglycemia. There was an activation of gluconeogenesis enzymes. Important changes were observed in the activities of muscle plasmic membrane enzymes (especially 5'-nucleotidase, EC 3.1.3.5, and ATPases, EC 3.6.1.3). Lindane, a lipophilic substance, especially disturbed the activity of membrane-bound enzymes enclosed in a phospholipid matrix.

An improved aqueous two-phase polymer method has been developed for the isolation of sperm plasma membranes by manipulating various parameters that influence markedly the purity as well as yield of the membrane. The method consists of hypotonic shock of intact spermatozoa with 1.25 mM EDTA to dissociate the plasma membrane and dispersion of these cells to a two-phase polymer system consisting of 5.5% 252-Kd dextran and 4.2% 20-Kd polyethylene glycol prior to centrifugation at 9700 X g for 30 min when the two polymer phases are separated; the membrane fraction sediments at the interphase. The resulting membrane fraction was purified further by repeating the two-phase fractionation step. The yield of the membranes was approx. 35-40%, based on the recovery of the membrane-bound marker enzymes alkaline phosphatase and 5'-nucleotidase. The isolated membranes showed a high degree of purity as evidenced by phase contrast and electron microscopic studies and analyses of marker enzymes characteristic of cellular organelles. The yield and purity of the membranes have been found to be markedly dependent on the conditions of the hypotonic shock, obtained as a function of, EDTA concentration and on the molecular sizes of the dextran and polyethylene glycol that constitute the two-phase polymer system, as well as on the centrifugal force used for the sedimentation of the membrane.

The changes in the biochemical parameters of peritoneal macrophages and their coupling to the secretory and phagocytic functions in CH3A mice during the growth of the reinoculated solid hepatoma 22a were studied. The DNA and RNA synthesis during the active tumour growth was more intense than that in resident macrophages. The activity of uridine kinase increased up to 156.0 +/- 12.0 nmol/hour/10(8) but was absent in resident macrophages. This was accompanied by a 7.2-fold increase of interleukin-1 synthesis as determined by the [3H]thymidine incorporation into thymocyte DNA in response to concanavalin A administration to C3H mice. Similar changes were observed in peptone-stimulated macrophages. A specific feature of macrophages from tumour-bearing mice was the impairment of activity of purine exchange enzymes and the efficiency of phagocytosis that were unobserved in peptone-stimulated macrophages. The activity of adenosine deaminase and purine nucleoside phosphorylase was inhibited as a result of their preincubation with zymosan, a phagocytosis-stimulating agent. This was accompanied by a significant decrease of the first chemiluminescence peak resulting from disturbances in Fc-reception. Macrophages of tumour-bearing animals possessed an increased 2.2-fold activity of membrane-bound AMP 5'-nucleotidase concomitant with the lack or decrease of the amplitude of the second chemiluminescence peak reflecting the disturbances in digestion resulting from phagocytosis.

The effect of oral ingestion of different fractions of onion (Allium cepa)--extract, residue, and whole--at a dose level equivalent to intake of 50 g of onion per day for a 70-kg man, for 30 days, to male adult, normal, albino rats was studied on blood and erythrocyte membrane lipids and certain membrane-bound enzymes. Onion extract and residue showed hypercholesterolemic effect, while whole onion showed hypocholesterolemic effect in blood. In erythrocyte membranes, all the fractions had hypocholesterolemic and hypolipidemic effect, which was accompanied by changes in the erythrocyte membrane enzymes studied, i.e., alkaline and acid phosphatase, 5'-nucleotidase, total and Mg2+ ATPase. The above study indicated that it is safer to take whole onion rather than onion residue or extract, because whole onion could lower the blood cholesterol level even in normal condition and has a less pronounced effect on the micro-environment of the cells.

The tick Boophilus microplus contains a nucleoside phosphate-hydrolysing enzyme which, in many respects, resembles the well characterized 5'-nucleotidase from mammalian tissue. The tick enzyme has been purified to homogeneity. It is a membrane-bound glycoprotein with an apparent Mr of 67,000 and, although it fails to hydrolyse a range of nucleoside 2'- or 3'-monophosphates, it has broad specificity for the 5' derivatives. Further investigation of the enzyme's substrate specificity, however, shows some important differences from the mammalian nucleotidases. It hydrolyses both bis-p-nitrophenyl phosphate and p-nitrophenyl phenylphosphonate, typical substrates for phosphodiesterases. However, the tick enzyme is most strikingly different from the mammalian enzymes in that it hydrolyses not only AMP but ADP and ATP as well. Further, the products of the hydrolysis of ATP are adenosine and tripolyphosphate, a reaction which has not been reported previously. The products of ADP hydrolysis are adenosine and pyrophosphate.

The relationship between undernutrition-induced alterations in some myelin membrane-bound enzyme activities and phospholipid fatty acid composition of this membrane was ascertained in developing rat brains. Undernutrition was imposed in pregnant dams through gestation and lactation, (last 10 days of gestation, and through lactation) by feed restriction. Experimental groups of animals received 50% of the amount of diet consumed by controls. Pups born to these mother rats were killed at day 7, 14 or 21 of postnatal age. Myelin membrane was isolated from the major regions of the brain, and analysed for phospholipid fatty acid profiles, phosphoinositides and ganglioside species. While there were no diet-related differences in the activities of 5'-nucleotidase (EC 3.1.3.5), myelin phospholipids from cerebella and brain stems of experimental rats exhibited lowered proportions of the long-chain polyunsaturated fatty acids, C20:4 (n - 6) and C22:6 (n - 3) concomitant with elevated activities of ouabain-sensitive Na+/K+-ATPase (EC 3.6.1.4). Levels of diphosphoinositide, triphosphoinositide and trisialogangliosides also decreased in myelin from brains of experimental animals. These results suggest a relationship between myelin phospholipid fatty acid profiles as indicators of membrane unsaturation, and the possibility of allosteric modification of Na+/K+-ATPase activity.

The gene encoding the membrane-bound 5'-nucleotidase of Vibrio parahaemolyticus was cloned and expressed in Escherichia coli. Cells of E. coli harboring a plasmid, pNUT5, which carries the 5'-nucleotidase gene were able to grow on ATP as the sole source of carbon, although the original cells were not. The 5'-nucleotidase activity was detected in whole cells of E. coli harboring pNUT5 and in membrane vesicles prepared from these cells. Most properties of the 5'-nucleotidase produced in E. coli, that is, its requirements for Cl- and Mg2+, substrate specificity, and inhibition by Zn2+, were similar to those observed in V. parahaemolyticus, but some alterations in properties were observed: The 5'-nucleotidase was partially inducible in V. parahaemolyticus, but its expression in E. coli was completely constitutive. The specific activity of the 5'-nucleotidase in membrane vesicles of E. coli harboring the plasmid was 30 times that observed in whole cells, whereas the specific activities in membrane vesicles and in whole cells of V. parahaemolyticus were almost the same. A new, dense band of protein with an apparent molecular mass of 63 kDa was detected when membrane proteins of E. coli harboring the plasmid were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.
