A method is described for the localization and characterization of phospholipases A1 and A2 (EC3.1.1.4) in Krebs II ascites cells, particularly in the plasma membranes. Cells were lysed with a Dounce homogenizer in an isotonic sucrose medium. Plasma membranes sediment with mitochondria and lysosomes during subcellular fractionation and are finally isolated on a continuous sucrose gradient. The membranes are localized at two levels in the gradient, at densities of 1.06 and 1.15, in which 5'-nucleotidase (EC 3.1.3.5) activity exhibits a 9- and 21-fold purification, respectively. Total contamination by endoplasmic reticulum, lysosomes, and mitochondria is 17 percent for the low-density membrane fraction and 25 percent for the high-density fraction. The phospholipases A present in Krebs II cells are active at pH 4.0 and pH 7.5. At the 2 pH values, they have A1 and A2 specificities. The intracellular distribution of acidic forms is comparable to that of acid phosphatase (EC 3.1.3.1), while neutral forms are localized like lactate dehydrogenase (EC 1.1.1.27). A small proportion of neutral phospholipase A2 has the same repartition on the sucrose gradient as nicotinamide adenine dinucleotide diaphorase (EF 1.6.4.3), an endoplasmic reticulum marker, and as 5'-nucleotidase, a plasma membrane marker.

Portions of closed jejunal biopsies from the dog were homogenised and their organelles separated by isopycnic centrifugation on continuous sucrose density gradients. The distributions of marker enzymes for the principal organelles were determined using highly sensitive assay procedures. The following organelles, with assayed marker enzymes and modal densities between brackets were characterised: peroxisomes (catalase, 1.21); brush borders (zinc-resistant alpha-glucosidase, leucyl-beta-naphthyl-amidase, gamma-glutamyl transferase, alkaline phosphatase, 1.20); lysosomes (N-acetyl-beta-glucosaminidase, alpha-mannosidase, 1.19); mitochondria (malate dehydrogenase, 1.18); endoplasmic reticulum (Tris-resistant alpha-glucosidase, 1.16); basal-lateral membranes (5'-nucleotidase, 1.11) and cytosol (lactate dehydrogenase). Homogenisation in isotonic sucrose containing digitonin (0.12 mmol/litre) selectively disrupted lysosomes and increased the equilibrium density of brush border and basal-lateral membranes. This procedure will be used to study the subcellular pathology of naturally occurring intestinal disease in the dog.

A study of the value of serum enzymes in 184 patients with colorectal cancer has been performed. The enzymes studied were gamma glutamyltransferase (gammaGT), alkaline phosphatase (AP), lactate dehydrogenase (LDH), 5'-nucleotidase (5'-NT), glutathione reductase (GR), alanine and aspartate transaminases. In patients without liver metastases, elevated enzyme levels were found in 11-55% preoperatively. 5'-NT showed the least number of elevated activities, while gammaGT activities were increased in 29% and LDH in 55%. The percentage of elevated enzyme levels rose significantly in the early postoperative period. Patients with liver metastases showed increased enzyme activities in 40-60% preoperatively: gammaGT was the most sensitive indicator. Increased enzyme activity was related to the degree of liver involvement with secondary tumor. With extensive liver metastases, gammaGT levels were increased in 82%. It is concluded that serum enzymes are of limited value in the preoperative detection of liver metastases, and particularly when tumor involvement of the liver is small.

1. Homogenates of guinea-pig left ventricle were fractionated by differential pelleting and by centrifugation on continuous sucrose density gradients. 2. The principal subcellular organelles of myocardium, characterized by their marker enzyme content, were resolved by density gradient centrifugation in a small-volume zonal rotor. The equilibrium densities (p) of the principal organelles are (with marker enzymes in parentheses): sarcolemma, 1-12 (5'-nucleotidase); lysosomes, 1-16 (N-acetyl-beta-glucosaminidase); mitochondria, 1-17 (cytochrome oxidase); peroxisomes, 1-18 (catalase); cytosol (lactate dehydrogenase). 3. The subcellular distribution of various adenosine triphosphatase activities and previously unassigned enzymes was determined. Leucyl-beta-naphthylamidase and gamma-glutamyl transpeptidase showed both cytosol and sarcolemma components. Ca2+-dependent adenosine triphosphatase showed dual localization to the mitochondria and to the sarcolemma.

Activity of enzymes catalizing bioenergetic processes of substance transport through cell membranes, adenosine triphosphatase and para-nitrophenyl phosphates, activity of certain enzymes of carbohydrate metabolism, lactate dehydrogenase and glucose-6-phosphate dehydrogenase, as well as to 5'-nucleotidase taking part in nucleic metabolism were determined in the pancreas of thyreoidectomized rats. Simultaneously the content of lactic acid, phosphorus, potassium and sodium, which immediately related to activation of the mentioned enzymes, was determined in the pancreas. In thyroidectomized rats the activity of Mg2+, Na+, K+-ATPase, Na+, K+-ATPase and lactate dehydrogenase in the pancreas increases, that of glucose-6-phosphate dehydrogenase, para-nitrophenylphosphatase and 5-nucleotidase decreases, the content of lactic acid, potassium, sodium and phosphorus increases.

In 30 patients with histologically verified malignant liver processes, the authors have examined the value of the serum activity of liver lactate dehydrogenase (LDH), alkaline phosphatase and gamma-glutamyltranspeptidase and in a number of patients, 5'-nucleotidase. These values were compared with the findings received from scintigraphy of the liver. Based on the results obtained, the authors have concluded that the activities of the gamma-glutamyltranspeptidase (gammaGT), 5'-nucleotidase and liver lactate dehydrogenase are more significantly increased in malignant processes of the liver than scintigraphy of the liver could register.

The microsomal fraction was isolated from homogenate of 125I-labeled leukemia L 1210 ascites cells by filtration of postmitochondrial supernatant through a Sepharose 4 B column. It was found that the particles are labeled with iodine and show 5'-nucleotidase activity suggesting the presence of cell membranes in the fraction. The soluble proteins fraction were retarded on the column showed lactate dehydrogenase activity, and low activity of soluble beta-D-glucuronidase.

Serum enzymes have not proved useful in evaluation of patients with early colon cancer, but certain enzymes such as transpeptidase, phosphohexone isosomerase, or 5'-nucleotidase have been of assistance in following the course of the disease, particularly in patients with metastatic spread to the liver. Attempts have been made to improve the utility of enzyme analysis in colon cancer by examination of enzyme patterns in colon biopsy specimens, feces, and colon washings. These studies, which will be summarized, are of importance in the possible development of diagnostic tools and as probes in the understanding of the etiology of colon cancer. The technical problems in carrying out these assays in humans, as well as the significance of the activity of aryl sulfatase, beta-glucuronidase, beta-glucosidase, lactic dehydrogenase, glucose-6-p-osphate dehydrogenase, and other enzymes will be considered.

Human erythrocyte ghosts were solubilized in a low ionic strength medium containing 1% Triton X-100 and subjected to electrophoresis in polyacrylamide gels containing Triton X-100. Five major bands were stained with Coomassie Blue, all except one band being heterogenous when re-electrophoresed in gels containing sodium dodecyl sulphate. It was possible to detect acetylcholinesterase, non-specific esterase, ATPase, alkaline phosphatase, 5'-nucleotidase, glyceraldehyde-3-phosphate dehydrogenase, lactate dehydrogenase, and aldolase activities on the Triton-containing polyacrylamide gels. Two of the enzymes, ATPase and 5'-nucleotidase, showed substantial inhibition by Triton X-100 in quantitative studies. This appears to be a useful method for studying membrane enzymes in normal and pathological red cells.

The effect of active schistosomiasis on the activity of some serum enzymes of hepatic origin has been studied in 46 male Egyptian patients with S. mansoni infections. These enzymes included total lactic dehydrogenase and its isoenzymes, 5'nucleotidase, B-glucuronidase, ornithine carbamyl transferase and sorbitol dehydrogenase. Samples were collected on admission, a week after the end of treatment and one month later. B-glucuronidase showed no change whereas ornithine carbamyl transferase, sorbitol dehydrogenase and 5'-nucleotidase showed elevated levels throughout the study. Total LDH was not significantly different on admission but increased significantly after treatment and returned to normal at follow up. Of the five LDH isoenzymes only LDH4 showed elevated levels. The significance of these findings in relation to conventional liver function tests is discussed.
