The activities of
5'-nucleotidase
, K+, Na+-activated Mg2+-dependent adenosine triphosphatase (ATPase) and leucine-beta-naphthylamidase were determined from 17 rheumatoid synovial fluids and from extracts of the corresponding synovial tissues. There was little correlation between the enzyme activities in the synovial fluids and those in the respective synovial-tissue extracts. In seropositive cases of
rheumatoid arthritis
the activities of
5'-nucleotidase
and leucine-beta-naphthylamidase in the synovial-tissue extract were higher than in seronegative cases. Also, the ratios of the enzyme activities in the synovial fluids to the resepctive activities in synovial tissue were lower in the seropositive cases. The activity of
5'-nucleotidase
in the synovial tissue decreased during gold treatment.

5'-nucleotidase
(5NT) activity in synovial fluid from 159 patients with various diagnoses. The activity of 5NT was compared with activities of nucleotide pyrophosphohydrolase, alkaline and neutral phosphatases, and adenosine deaminase, in the same samples. Higher levels of 5NT activity occurred in synovial fluid from osteoarthritic joints than from joints of patients with gout, pseudogout, or
rheumatoid arthritis
. The highest levels of 5NT activity were found in synovial fluid from patients with Milwaukee shoulder syndrome and from osteoarthritis patients in whom deposition of calcium-containing crystals was also present.

A total of 14 transformed cell clones were obtained by micro-injecting origin-defective SV40 DNA into three types of cloned adherent synovial cells (ASC) (dendritic cells (DCs), macrophage-like cells (MCs), and fibroblast-like cells (FCs)) from two
rheumatoid arthritis
patients (five DC clones (SV40-DCs), five MC clones (SV40-MCs) and four FC clones (SV40-FCs)). All the transformed cell nuclei expressed SV40-specific T antigen. The cells which formed a colony had a few times shorter doubling time than the original cells. IL-1 alpha, IL-1 beta and prostaglandin E2 were detected in the culture supernatant from the unstimulated transformed cells like untransformed cells. The SV40-DCs showed the most potent accessory cell function in oxidative mitogenesis assay among the three types of SV40-ASCs. Granulocyte macrophage colony stimulatory factor (GM-CSF) was detected only in the culture supernatant from the SV40-MCs without stimulation. Extensive phenotypic analysis revealed relatively cell-specific markers. SV40-DCs were HLA-DP+ and glial fibrillary acidic protein positive. SV40-MCs stained positive for

We studied purine metabolism in
rheumatoid arthritis
(RA), adenosine deaminase (ADA),
5'-nucleotidase
(5'NU) and purine nucleoside phosphorylase (PNP) activities by measuring the circulating mononuclear cells of patients with RA and healthy controls. Patients had significantly lower levels of ADA and 5'NU but not of PNP than controls. The decreases could not be related to age, antiinflammatory therapy, decreased percentages of T cells or imbalance between major T cells subsets. Differences in cell maturation or traffic could account for our observation. Alternatively, abnormalities of purine metabolism are not definitely excluded in RA if the lower enzyme activity is not sufficient to perform the metabolic steps.

The aim of this work was to discover if Mycoplasma fermentans, which is known to infect B cells, could be the cause of the raised ecto-5'-nucleotidase observed in the synovial fluid of
rheumatoid arthritis
patients. The ecto-5'-nucleotidase activity in the patients' serum has been shown to correlate with the erythrocyte sedimentation rate and DNA from the mycoplasma has been found in the synovial fluid. B lymphoblastoid cell lines were exposed to 16 strains of Mycoplasma fermentans and their ecto-5'-nucleotidase, CD73, was measured both biochemically and by mouse antibodies to human ecto
