(5'-N) activity in 17 human fetuses was systemically studied in order to get basic values of the enzyme activity. A wide spectrum of 5'-N activity among the organs was revealed. Very strong activity was found in placenta (42.62 +/- 19.24 nmol adenosine/mg protein/min), skeletal muscle (31.80 +/- 11.10) and skin (27.31 +/- 13.00); strong activity in liver (18.96 +/- 7.60), pituitary (15.06 +/- 6.13) and thyroid (12.69 +/- 4.47); moderate activity in lung (8.43 +/- 3.68), pancreas (8.20 +/- 2.26), small intestine (8.07 +/- 2.28), lymph node (7.26 +/- 2.22),
thymus
(7.06 +/- 2.52), spinal cord (5.86 +/- 1.96) and spleen (5.65 +/- 2.52), and weak activity in testis (4.02 +/- 0.79), heart (3.95 +/- 1.22), adrenal (3.46 +/- 1.68), kidney (3.16 +/- 0.83), ovary (3.16 +/- 0.72), aorta (2.98 +/- 1.20), cerebellar hemisphere (2.28 +/- 1.09) and frontal lobe of the cerebrum (1.49 +/- 0.54). These data are the first to be reported on specific 5'-N activity in the human fetus. The significance of the wide range of 5'-N activity among the organs, and future aspects on the study of 5'-N activity with respect to cell differentiation, maturation, development, aging or malignant transformation were discussed.

A microassay requiring as few as 2 X 10(5) cells per assay was developed for systematic analysis of 9 purine enzymes in lymphocytes from equine peripheral blood, spleen, lymph node,

and bone marrow. The activities of adenosine deaminase (ADA), purine nucleoside phosphorylase (PNP), adenosine kinase (AK), deoxyadenosine kinase (dAK), deoxycytidine kinase (dCK),
5'-nucleotidase
(5'-N), AMP deaminase, hypoxanthine-guanine phosphoribosyl transferase (HGPRT or HPRT), and adenine phosphoribosyl transferase (APRT) were measured by this microassay in lymphocytes from peripheral blood from four different breeds of horses (Arabian, Quarter Horse, Thoroughbred and Shetland Pony). There were no significant differences in the enzyme activities among the various breeds. Peripheral blood lymphocytes (PBL) from foals exhibited enzyme activities similar to those observed for adult animals. All lymphoid tissue contained similar levels of activity for each kinase (AK, dAK and dCK). Spleen had the highest activity for ADA, PNP, 5'-N, and HGPRT. The lowest activity for ADA, APRT, PNP and AMP deaminase was found in
thymus
. Enzymatic activities that varied the most among the tissue were 5'-N, ADA, APRT, HGPRT and AMP deaminase.

The present studies concern the effects of triiodothyronine (T3) on rat thymic and splenic NaK-ATPase activities. When hypothyroid rats were given a single injection of T3 (250 micrograms/100 g body weight), thymic NaK-ATPase activity increased by 18% at 24 h (p less than 0.005), and remained at this peak at 72 h. Injection of T3 (250 micrograms/100 g body weight, on alternate days X 3) significantly augmented NaK-ATPase activity from the
thymus
and spleen of hypothyroid rats by 19 and 49%, respectively. Two weeks of daily injections of T3 (1 microgram/100 g body weight) to hypothyroid rats significantly increased NaK-ATPase activity in the
thymus
and the spleen by 14 and 28%, respectively. Reverse T3 had no significant effect of thymic and splenic NaK-ATPase activities. Mg-ATPase and
5'-nucleotidase
activities were not significantly affected by T3 in either tissue under different thyroid states. The lack of response of thymic and splenic NaK-ATPase to reverse T3, and Mg-ATPase and
5'-nucleotidase
to T3, substantiates the specificity of the hormone action on NaK-ATPase. To investigate whether the T3-dependent increase in NaK-ATPase activity is a result of an increase in the number of enzyme units, the effect of T3 on the quantity of phosphorylated intermediate of NaK-ATPase was assessed in the partially purified membrane fraction. In the euthyroid state, NaK-ATPase activity and phosphorylated intermediate units were 33 and 43% higher, respectively than in the hypothyroid state, implying that the T3-dependent increase in NaK-ATPase activity is a result of an increase in the number of NaK-ATPase units.

Activities of key enzymes of purine metabolism [adenosine deaminase (AD); purine nucleoside phosphorylase (PNP);

Immunological properties of lymphocytes were also investigated measuring antibody formation and the reaction of blasttransformation in response to phytohemagglutinin, concanavalin A and lipopolysaccharide. Within the first 48 hrs after the tumor implantation and immunization certain nonspecific biochemical mechanisms of lymphocytes activation (elevated AD activity, decreased activity of

are reviewed. The optimal conditions for the cytochemical localization of
5'-nucleotidase
(
AMPase
) in the mouse lymphocyte have been established. Quantitative monitoring of the effects of fixation and the components of the cytochemical medium showed that cytochemistry can be performed under conditions that do not lead to loss of
AMPase
activity. The cytochemical reaction product was seen only on the surface of a proportion of splenic lymphocytes, regardless of the fixative used. The splenic cell population included a distinct population of lymphocytes with readily demonstrable
AMPase
activity and another population with no cytochemically demonstrable
AMPase
activity. The number of positive cells varied, but was usually between 20 and 30% when cells were fixed in glutaraldehyde. Lymphocytes purified from
thymus
were always negative for cytochemically demonstrable
AMPase
activity. Biochemically, it was shown that the
AMPase
activity of spleen lymphocytes is more than six times higher than that of
thymus

and spleen lymphocytes in the latent period of DMBA-induced mammary carcinogenesis. The adenosine deaminase activity was established to increase while
5'-nucleotidase
and AMF-aminohydrolase activity to decrease in the thymocytes of intact animals treated with thymostimulin; the adenylae kinase activity of spleen lymphocytes decreased as compared with that in the rats not treated with the preparation. The dynamics of changes in the investigated enzymes in activities in lymphocytes was of wave-like pattern in the latent period. The treatment of animals with thymostimulin in this period normalized adenosine deaminase activity and decreased the activity of the other enzymes in these cells.
