Blood samples from 109 Siriono (Eastern Bolivia) belonging to the Tupi-Guarani group were investigated for enzyme variants in the following systems: glucose-6-phosphate dehydrogenase, phosphogluconate dehydrogenase, adenylate kinase, phospho-glucomutase (locus 1 and 2), acid phosphatases, lactate dehydrogenase,
NADH diaphorase
, pseudocholinesterase (E1 and E2 locus), and serum
alkaline phosphatase
. The most relevant observations are: (1) A relative lack of polymorphism, a characteristic feature of the Amerindian populations studied up to now. These data are consistent with the hypothesis of a 'common ancestral background' in Indian populations whatever the degree of sociocultural and linguistic diversity, and the geographical distances. (2) Specific traits due to the frequency of alleles in some systems confer to that tribe a particular position among Amerindians. The effects of genetic drift may be postulated in order to explain the high rate of PGM and 6PGD polymorphism. Furthermore, in that small community, the disappearance of some alleles (pa gene) can plausibly be explained in terms of a balanced influence of mutational and selective pressure.

Zinc content of testes, bones, esophagus, kidneys, and muscles was decreased, whereas iron content was increased in the testes of zinc-deficient rats compared to restrictedly fed control rats. Histochemical enzyme determinations revealed reduced activities of certain enzymes in the testes, bones, esophagus, and kidneys. In the testes, lactic dehydrogenase (LDH), malic dehydrogenase (MDH), alcohol dehydrogenase (ADH), and
NADH diaphorase
; in the bones, LDH, MDH, ADH, and
alkaline phosphatase
; in the esophagus, MDH, ADH, and
NADH diaphorase
; and in the kidneys, MDH and
alkaline phosphatase
were decreased in zinc-deficient rats compared to restrictedly fed controls. Succinic dehydrogenase (SDH) revealed no significant changes under the conditions of our experiments in various groups of rats that were investigated. In a "repleted" group of rats, content of zinc in testes and bones increased significantly, compared to the deficient group. The iron content of the testes decreased after repletion with zinc. In the testes, bones, esophagus, and kidneys, the activities of various enzymes increased after repletion with zinc. Inasmuch as the major manifestations of zinc deficiency syndrome in the rat include growth retardation, testicular atrophy, and esophageal parakeratosis, our results suggest that the content of zinc in the above tissues most likely controls the physiological processes through the formation of zinc-dependent enzymes.

Endogenous enzyme activity can be readily and routinely demonstrated in ultrathin, frozen sections for electron microscopy. The procedure employed to obtain the best structural preservation as well as enzyme activity in thin sections involved fixation in glutaraldehyde, embedding in thiolated gelatin or pure gelatin, partial dehydration in glycerol, and sectioning in a cryostat at -35 degrees C with a slightly modified Porter-Blum microtome on which the tissue is maintained at -70 degrees C and the knife at -23 degrees C. Kidney cortex was used as test tissue, but a few other organs were occasionally used. Thin sections were floated on the surface of several incubation media routinely employed for enzyme cytochemistry. Positive, specific reactions were obtained for
alkaline phosphatase
in kidney brush border, for adenosine triphosphatase in brush border and in basal membranes of distal tubules, for acid phosphatase and esterase in lysosomes, and for
NADH diaphorase
in mitochondria. Mitochondrial ATPase was sporadically evident only in the distal tubule of the kidney. Localizations of enzyme activity reported by other technical approaches were confirmed and in some cases somewhat improved.

Brush border membrane vesicles from rat small intestine were isolated by a Mg/EGTA precipitation method. Further fractionation either by free flow electrophoresis or by sucrose density gradient centrifugation leads to subfractions which differ with respect to enzyme enrichment factors, transport properties for D-glucose and protein pattern analyzed by sodium dodecyl sulfate/polyacrylamide gel electrophoresis. A relative enrichment of (Na+ + K+)-ATPase is found in one fraction, whereas in another fraction maltase, aminopeptidase M and
alkaline phosphatase
are relatively enriched. The fractions show different properties of D-glucose transport under tracer exchange conditions and a different inhibition of D-glucose transport by phlorizin and phloretin. These results indicate that the vesicles obtained from rat small intestine by this cation precipitation method are not homogeneous. The inhomogeneity cannot be due to a crosscontamination by membranes other than from the cell envelopment, as none of the fractions show a significant enrichment of succinate--cytochrome c oxidoreductase, KCN-resistant
NADH oxidoreductase
or glucosaminidase. The inhomogeneity might be due either to a crosscontamination by basal-lateral membranes or to membranes derived from epithelial cells not yet fully differentiated.

The activities of
alkaline phosphatase
and reduced nicotinamide adenine dinucleotide (NADH) diaphorase in the principal cells of the guinea pig epididymis were studied histochemically. Alkaline phosphatase activity was absent from the principal cells but was present in the basement membrane of the epididymal epithelium.
NADH diaphorase
activity was distributed throughout the cytoplasm of the principal cells in each epididymal segment. There was a gradual increase in
NADH diaphorase
activity from segments 1 through 7. Possible functions of
alkaline phosphatase
and
NADH diaphorase
in the epididymis are discussed.

NADPH diaphorase is a histochemical activity which, in formaldehyde-fixed tissue, is rather specific for nitric oxide synthase. Recently, it was shown that NADPH diaphorase activity is inhibited by ethylenediaminetetraacetic acid (EDTA) in neurons but not in the choroid plexus epithelium. The present study, while confirming these results, demonstrates that the apparent sensitivity of NADPH diaphorase for EDTA reflects only the dependence of malic enzyme, which is used as the source of reduced cofactor, on Mg2+ or Mn2+ ions. Furthermore, evidence is provided that the apparent EDTA-insensitive NADPH diaphorase activity in the choroid plexus reflects the activity of
alkaline phosphatase
in conjunction with
NADH diaphorase
. Apart from these pitfalls, the use of the indirect, malic enzyme based method for NADPH diaphorase was found to cause much higher background staining compared to the direct method using NADPH, and is therefore proposed to be abandoned.

Histochemical examination of tissues from chickens maintained from hatching on experimental zinc-deficient diets, showed that enlargement of the nucleoli of the crop epithelium was associated with increased amounts of RNA in that organelle.
