Changes in serum enzyme levels, liver histology and liver function tests have been correlated to determine the usefulness of these tests in assessing liver status. The effects of carbon tetrachloride administration on these parameters has been determined in a group of 20 sheep. Normal levels, elevated levels after injury and the effect of elapsed time after injury are reported for serum glutamic dehydrogenase, sorbitol dehydrogenase, glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase,

, cholesterol and proteins. Variation in the time of elevation of enzyme activities may be useful in determining the elapsed time between acute injury and serum sampling. In comparison to sheep fed an adequate diet, a diet with a restricted protein intake was associated with increased severity of histological lesions and decreased liver function.

The method for the determination of enzymic activity in turbid, lipaemic sera, which involves clearing by polyanion precipitation with heparin and magnesium chloride, was critically reviewed. In the diagnosis of diseases of the liver and pancreas, which are frequently associated with hyperlipoproteinaemia, only residual enzyme activities are measured in the cleared serum after polyanion treatment. In the measurement of glutamate dehydrogenase and in the Phadebas test for alpha-amylase, the enzymes are inactivated by treatment with heparin and magnesium chloride. On the other hand, as a result of polyanion precipitation gamma-glutamyl transferase is transferred, together with lipoproteins and chylomicrons, to the lipid-rich supernatant. Acid phosphatase also exhibits only residual activity in cleared serum. The activity of aspartate aminotransferase, alanine aminotransferase,

Enzyme survey specimens were prepared by spiking portions of a normal serum pool with creatinine, urea, and five enzymes (
lactate dehydrogenase
, creatine phosphokinase,
alkaline phosphatase
, and aspartate and alanine aminotransferases), and preparing admixtures of the spiked pools with the original serum. This admixture technic established linear interspecimen relationships that could be confirmed by analyses for creatinine and urea nitrogen. Both ethylene glycol-stabilized liquid serum specimens and lyophilized specimens were prepared as sets of six to eight samples having six concentrations of each enzyme. The sets were distributed on five occasions to about ten laboratories that were widely separated geographically, and the specimens were analyzed by a variety of methods, by various instrumental systems, and in different reaction conditions, and results were reported in diverse units. This report describes how the analytic data obtained through the use of these specimens that were specifically designed for survey purposes can be analyzed statistically to provide meaningful assessments of laboratory performance in enzyme analyses.

Diethyistibestrol was administered orally to 11 boys with Duchene muscular dystrophy (DMD). Creatine phosphokinase (CPK) and
lactate dehydrogenase
(
LDH
) activities, characteristically high in DMD, and presumably of muscle origin, were reduced significantly (P LESS THAN .05). On ther other hand, the activity of
alkaline phosphatase
, an enzyme not of muscle origin, increased. These enzyme changes were reversible when diethyistibestrol was discontinued. Despite appreciable alterations in totoal serum enzyme activity, no important change was found in the isozyme patterns. Piperazine estrone sulfate was administered to another patient with DMD. The effects of this physiologic hormone were, in part, similar to those of diethyistibestrol. Experimentally, diethylstilbestrol reduced the efflux of CPK and
LDH
from mouse skeletal muscle. This may be the manner by which diethylstilbesterol reduced the serum enzyme levels in DMD, but this has not been proved directly. These studies are the first step in an effort to identify various agents that in combinations may normalize serum enzyme levels.

The activity of the following enzymes in clinically normal newborn calves was investigated: glutamate-oxalacetate transaminase (GOT), glutamate-pyruvate transaminase (GPT),
alkaline phosphatase
(APh), creatine phosphokinase (CPhK),
lactate dehydrogenase
(
LDH
), leucine-aminopeptidase (LAP), aldolase (A), and cholinesterase (ChE). The studies were carried out at the first hour prior to offering colostrum as well as at the 6th, 12th, 24th hr and on the 2nd, 3rd, 4th, 5th, 7th, 10th, 15th, and 20th day following it first intake. Regularly rising values of the enzyme activity up to the 24th hour were observed with APh, GOT, GPT, CPhK, and LAP. The aldolase enzyme (after colostrum had been given for the first time) in all animals showed a statistically significant drop of activity at the 6th hour. The activity of
LDH
displayed a consistently rising trend up to the end of the experimental period. The cholinesterase activity showed high values immediately following birth, reaching those found in the dams by the end of the observation period.

We examined whether inter-individual differences in correlation coefficients previously found among subjects truly reflect consistent inter-individual differences or are time-related within an individual. The consitutents studied in this investigation were (a) the enzmes aspartate aminotransferase,
alkaline phosphatase
, and
lactate dehydrogenase
; and (b) the non=protein nitrogen-containing constituents urea, uric acid, and creatinine. Ten healthy women were each subjected to 15 venipunctures over a five-week period (Series I), and, after a two-month interval, were again subjected to 15 venipunctures over a second five-week period (Series II). Before statistical analysis, the data were corrected for the batch-to-batch (day-to-day) arnalytical variation. There was a signiificant (P less than .05) change in the covariance structure (variances or correlation coefficients, or both) between the two series in four of the 10 subjects for the combination of enzymes, and in three other subjects for the combination of nonprotein nitrogen constitutents. Although we found a significant (P lees than .05) average intra-individual variation in the mean values from series to series in the cases of the three enzymes and urea, the magnitude of the inter-series variation in means was relatively small. We report the effect of exercise on the activity values of five enzymes in sera as studied in four healthy male volunteers. The underlying purpose of this present study was to produce an increase in the activity values in the sera of selected enzymes found in muscle. Then by observing the decay rate of these enzymes, we computed the inter-individual differences in clearance rates serum half-life) of these enzymes. Blood specimens were collected just prior to exercise, 1 h after excerise, and on eight additional times up to 93 h after exercise. All specimens were assayed on one occasion for activity values of creatine kinase, asparate aminotransferase, alanine aminotransferase,
alkaline phosphatase
, and
lactate dehydrogenase
. We found increases in the three muscle enzymes with average increases being: creatine kinase, +116%; asparate aminotransferase, +41%; and
lactate dehydrogenase
, +32%; all of which remained above baseline values for 53 h or longer. In the case of creatine kinase, a monoexponential decay curve depicted the data (from the 19-h specimen to the 67-h specimen). The calculated "apparent serum half-life" for creatine kinase varied from 38 h to 118 h in the subjects tested.

. The most relevant observations are: (1) A relative lack of polymorphism, a characteristic feature of the Amerindian populations studied up to now. These data are consistent with the hypothesis of a 'common ancestral background' in Indian populations whatever the degree of sociocultural and linguistic diversity, and the geographical distances. (2) Specific traits due to the frequency of alleles in some systems confer to that tribe a particular position among Amerindians. The effects of genetic drift may be postulated in order to explain the high rate of PGM and 6PGD polymorphism. Furthermore, in that small community, the disappearance of some alleles (pa gene) can plausibly be explained in terms of a balanced influence of mutational and selective pressure.

The adsorption of 8 enzymes to polyaminomethylstyrene was studied. While

alkaline phosphatase
and glucose-6-phosphate dehydrogenase exhibit a relatively low affinity to the carrier, alcohol dehydrogenase, glutamate dehydrogenase and urease were found to form stabile complexes with the polymer that are enzymatically active. Adsorbed urease and beta-hydroxybutyrate dehydrogenase, are still active after several weeks; the other preparations lose their activity soon. It can be shown by the example of yeast alcohol dehydrogenase that the activity loss following adsorption is caused possibly by a process of reorientation of already bound enzyme molecules or by the increasing enzyme coverage of the carrier, with the active centres becoming more and more inaccessible for the substrate. During the substrate conversion catalysed by the alcohol dehydrogenase-polyaminomethylstyrene complex, a small amount of the enzyme is again detached from the carrier. The activity rises to a certain extent in the supernatant but drops to zero again. The stability of the adsorbed urease is distinctly increased compared with the dissolved enzyme. For the pH optimum and the KM value there are no differences between the two preparations. Continuous application of polyaminomethylstyrene-bound beta-hydroxybutyrate dehydrogenase and urease, respectively, in a column shows that both preparations have unchanged enzymatic activities even after running times of 5 and 24 days, respectively.
