1) Variations in the serum concentrations of total proteins and the electrophoretic fractions, glycoprotein, mucoprotein, fibrinogen, erythrocyte sedimentation rate, calcium, phosphorus, alkaline, and acid phosphatases were analyzed until the 30th day following uncomplicated fracture of shafts of long bones of the limbs in 25 cases. 2) A significant fall of albumin with concomitant rise of alpha 1, alpha 2, and beta globulins were noted until 30th day. 3) Mucoprotein, glycoprotein, and fibrinogen showed parallel elevations with that of alpha and beta globulins. 4) The peak values of alpha 1 and alpha 2 globulins, mucoprotein, and fibrinogen were registered on the 10th day after trauma. Albumin showed maximum fall on the 10th day in all these cases. 5) Glycoprotein showed a peak value on the 5th day. 6) Total protein and gamma globulin remained almost unchanged throughout the studies. 7) Beta globulin showed higher values and paralleled more closely the fibrinogen and erythrocyte sedementation rates. 8) The elevations of beta globulin, fibrinogen, and erythrocyte sedimentation rate were higher, and persisted beyond 30 days in lower-limb fractures as compared to upper-limb fractures. 9) Serum calcium, phosphorus, alkaline phosphatase, and acid phosphatase were not significantly different following fractures and therefore did not reflect much physiologic variation. 10) The most significant changes in the levels of plasma fractions studied were conspicuous on the 10th day and lasted for about 1 month.

In serial studies of hepatic function in rats after 70% partial hepatectomy, quantitative changes were found in several of the serum components used clinically to assess liver status. The activities of the following enzymes were found to increase: gamma-glutamyl transpeptidase and lactic dehydrogenase were maximal 6 h postoperatively, while glutamic oxaloacetic transaminase and alkaline phosphatase reached peak values at 24 and 48 h respectively. Albumin levels were found to be relatively constant during the study; however, total protein concentration was lowest 6--12 h postoperatively, paralleling a decrease in globulin concentration. Bilirubin levels were elevated to 4x normal within 12 h after surgery. After partial hepatectomy calcium and phosphorus concentrations were significantly decreased at 24 and 12 h respectively. With the exception of alkaline phosphatase, the activities of all serum components measured returned to normal levels by 1 week after surgery; the alkaline phosphatase concentration continued to be elevated 2 weeks postoperatively.

Phenotypes of the cells developing into small colonies after days of primary culture of adult rat hepatocytes in serum-free modified Dulbecco Modified Eagles' medium containing 10 mM nicotinamide and 10 ng/ml epidermal growth factor were analyzed immunocytochemically, cytochemically and ultrastructurally. Albumin, cytokeratin 8 and 18 were seen by immunocytochemical techniques in the cells of the small colonies at Day 6. Transferrin, alpha 1-antitrypsin, ceruloplasmin, and haptoglobin, proteins secreted by mature hepatocytes, were faintly stained in these cells as was alpha-fetoprotein. These proteins were secreted into the culture medium as evidenced by immunoblot analysis. gamma-Glutamyltransferase, alkaline phosphatase and glucose 6-phosphatase were not present in the cells of the small colonies as well as the surrounding hepatocytes at Day 6 of culture. In addition, ultrastructural examinations of the cells in the small colonies indicated that these cells not only had many characteristic mitochondria and desmosomes, but also a few small peroxisomes. Such cells, even after 20 days in culture were proliferating, as evidenced by the intranuclear presence of the proliferating cell nuclear antigen. The potential relation of these cells to hepatocytes which may serve as the principal reserve for replicating hepatocytes is discussed.

A study was carried out to evaluate the role of laboratory investigations for diagnosis of liver abscess from the year 1976 to 1981. Bacteriological, parasitological, serological, haematological, histopathological studies and liver function tests were carried out in 240 confirmed cases of liver abscess during the study period. Out of 219 cases, pus culture for pyogenic bacteria was positive in 68 cases (including 27 cases of secondary bacterial infection of amoebic liver abscess) and sterile in 151 cases. Ent histolytica was demonstrated in pus of 24 cases. The diagnostic titre of 1:128 and above was detected in 137 sera by isohaemagglutination test. The liver function tests were of limited value. Albumin globulin ratio was altered in 91.25% cases and there was moderate increase in the levels of alkaline phosphatase. Histopathological studies demonstrated Ent histolitica in 2 cases only. Changes characteristic of pyogenic liver abscess were observed in 6 cases. Rest showed no specific changes.

Isolation procedures employed by various laboratories to obtain cerebral microvessels generally utilize meshes to sieve and collect the microvessels from homogenized brain. This is followed in some cases by further purification using density gradients of Percoll or sucrose, or albumin flotation. We have evaluated microvessels prepared by these methods in terms of ATP content and ATP/ADP ratio, which reflect the cellular energy state, and enrichment of the marker enzymes, alkaline phosphatase and gamma-glutamyltransferase. Albumin flotation generally increased the enrichment of marker enzymes; however, preparations using albumin flotation or a Percoll gradient exhibited considerable variability in ATP content and ATP/ADP ratio with the mean ATP/ADP ratio significantly lower than that observed in microvessels isolated by sieving through meshes. More uniformly high values for both ATP (approximately 1.6 nmole ATP/mg protein) and the ATP/ADP ratio (approximately 2.3) were obtained with meshes alone. Use of a sucrose gradient consistently resulted in preparations with a much lower ATP content and ATP/ADP ratio, compared with preparations obtained with the other methods. Values using the other methods were higher than those previously reported, yet were still lower than the ATP content of about 23 and ATP/ADP ratios of 18 and 7 we found in cultured microvascular endothelium and pericyte, respectively. These low values were not improved by supplying additional fuel to the microvessels during isolation, suggesting they were not the result of fuel deprivations during isolation. Despite the probable damage incurred during isolation, microvessel preparations are a useful in vitro model in which fuel metabolism appears to reflect the prior hormonal/nutritional state of donor animals. However, our data indicate the advisability of measurements of ATP content and ATP/ADP ratio for quality control of preparations used for metabolic studies, especially after Percoll density gradient or albumin flotation steps.

Twelve blood parameters were studied in five groups of women totalling 120 subjects--group I: 26 blood donors (average age 45.2 years, range 23-66); group II: 18 patients with various cerebral, cardiovascular, or infectious illnesses (average age 79.9 years, range 66-92); group III: 28 patients with femoral neck fractures (average age 79.4 years, range 56-95); group IV: 12 patients with hip osteoarthrosis (average age 71.7 years, range 60-87); group V: 36 patients with vertebral crush fractures associated with postmenopausal and involutional osteoporosis (average age 63.0 years, range 51-75). The parameters measured were total proteins, albumin, total, alpha 1, alpha 2, beta, and gamma globulins, total calcium, phosphates, alkaline phosphatase, bilirubin, and haemoglobin. Statistical analysis showed that each group differed from the others even with adjustment for age. Among the discriminant parameters, serum albumin had a distinctive position. Significantly high concentrations of serum albumin in the group with osteoarthrosis raise the question of the possible existence of a population prone to osteoarthrosis in whom the serum albumin level may reflect a special nutritional state associated with the well known bone density in subjects with hip osteoarthrosis. Albumin values in patients with femoral neck fractures are lower than normal but non-significantly. The difference between the group with vertebral crush fractures and that with femoral neck fractures seems to be due to age.

1. Zinc deficiency was produced experimentally in guinea-pigs fed on a diet containing 1.25 mg Zn/kg diet over a period of 60 d. In addition, the response of the Zn-deficient (ZnD) animals to Zn supplementation was studied for 15 d. 2. In the ZnD group a significant reduction was found in serum Zn and protein concentrations and in alkaline phosphatase (EC 3.1.3.1; AP) activity from day 24 onwards. 3. Paper electrophoretic studies on serum revealed a significant decrease in relative values, as well as absolute values, of albumin and gamma-globulin and an increase in beta-globulin. 4. Albumin:globulin increased on day 24 but decreased significantly from day 48 onwards. 5. The kidney and testis of the ZnD group showed a reduction in Zn and protein contents, and AP activity. 6. Zn supplementation of the previously ZnD group resulted in marked although incomplete improvement in the biochemical indices studied.

A sensitive and simplified competitive binding enzyme linked immunosorbent assay (ELISA) has been developed for quantification of urinary albumin. It was found that, probably because of a high antibody dilution, a conventional ELISA with three incubation steps could be simplified to an assay with only one incubation step without losing sensitivity or precision. The technical conditions of the assay are described. Albumin was coated to the walls of polystyrene microtitre plates. Diluted urine was mixed with rabbit antiserum against albumin and then with alkaline phosphatase conjugated anti-rabbit immunoglobulins. The mixture was then incubated in the microtitre plate for 3 h. After washing the plate, substrate was added and the enzyme activity was measured. Detection limit of the assay was 15 micrograms/l or 1.5 ng. The intra-assay and inter-assay coefficients of variation were 6 and 9%, respectively. The range of 24-h excretion of urinary albumin in apparently healthy subjects was 0.6-27.2 mg.

Albumin, total protein, total bilirubin, glucose, cholesterol, triglycerides, blood urea nitrogen, alkaline phosphatase and glutamic oxaloacetic transaminase levels were determined in the serum of cord blood of neonates born to mothers previously treated with the betamimetic drug fenoterol. The results were compared with those of untreated controls. More deviations from the control were found if the interval between termination of treatment and delivery was shorter than 48 hours. A longer drug-free interval seems more favourable for the metabolic balance of the newborns. Newborns whose mother has received beta-sympathomimetic tocolytic treatment need careful supervision.

Hematological, biochemical, histoenzymological, and histopathological changes in serum and tissues were studied in chickens during outbreaks of nephritis. Hematological studies revealed normocytic-normochromic anemia characterized by increased total erythrocyte counts, hemoglobin, packed cell volume, and erythrocyte sedimentation rate. Albumin-to-globulin ratio and sodium levels in serum, glucose in blood, and alkaline phosphatase and glucose-6-phosphatase in liver and kidneys were decreased. Glutamate pyruvate transaminase, uric acid, non-protein-nitrogen, and potassium levels in serum were increased. No significant change in the calcium, phosphorus, and total protein levels in serum was observed. These changes were directly related to the severity of the nephritis.
