The purpose of the present study was to elucidate the metabolic requirements of autophagocytosis. Two model systems were used for this purpose: a) glucagon-induced autophagocytosis in the rat liver, and b) the wave of autophagocytosis which occurs when isolated flounder kidney tubules are incubated in vitro. In the rat liver, protein synthesis was inhibited by the administration of cycloheximide (1.5 mg/kg) to rats 2 hours prior to glucagon injection. In flounder kidney tubules, protein synthesis was inhibited at least 90% by adding cycloheximide, actinomycin D, pactamycin and puromycin to the medium. In both systems the inhibition of protein synthesis failed to inhibit the formation of autophagic vacuoles or their subsequent transformation into autolysosomes, as depicted from electron microscopic histochemical preparations. In flounder kidney tubules no differences were found in the levels of p-nitrophenylphosphates, beta-DL-
glycerophosphatase
, N-acetyl-beta-D-glucosaminidase,
arylsulphatase
, beta-D-galactosidase or acid proteinase when tubules were incubated up to 5 hours in the presence or absence of protein synthesis inhibitors. When ethionine was administered to rats 2 hours before glucagon injection, a decrease of approximately 75% in the ATP levels was observed. After ethionine administration, glucagon failed to induce the formation of autophagic vacuoles. The incubation of flounder kidney tubules in the presence of cyanide or in a nitrogen atmosphere decreased the ATP levels to less than 10% of controls and blocked autophagy. On the other hand, cyanide had little effect on acid hydrolase levels at 1 hour of incubation. A wide variety of other inhibitors were also shown to block autophagy. These results further support the hypothesis that, in the formation of antophagic vacuoles, preexisting enzyme and membrane pools are utilized. On the other hand, the esotropy-exotropy membrane conformational changes occurring in the formation of autophagic vacuoles seem to be energy dependent and can therefore be blocked by lowering intracellular ATP levels.

activity was detected in R-, R'-D- and B-cells, with labelling being observed on the apical plasmic membrane level in all four, and in B-cells on the pinocytotic vesicle membranes. Acid phosphatase and aryl-sulphatase activities were only detectable in B-cells; the most frequently labelled structures were located in the vacuolar system, dictyosomes and Golgi vesicles, although Golgi structures occasionally reacted to acid phosphatase. Nome of the dense bodies observed in B-cells reacted to
arylsulphatase
. Similarly they were unevenly labelled during acid phosphatase tests. Hence it may be assumed that dense bodies are not involved in hydrolases. It is possible that these enzymes originated from vesicles generated by the Golgi saccules surrounding and joined to the vacuoles, thus bypassing the lysosome I stage.

A protective immune response without liveweight loss can be induced in sheep against T. colubriformis but results depend on the anthelmintic used and duration of immunizing infections. More than 90% protection was achieved in sheep immunized by three 15- or 7-day oxfendazole abbreviated infections or three 21-day nonabbreviated infections. Only 41% protection was induced by 3-day oxfendazole abbreviated infections. Significantly higher worm burden and faecal egg counts were present after challenge in sheep immunized by 7-day levamizole abbreviated infections compared to 7-day oxfendazole abbreviated infection. Liveweight gains of sheep immunized by 15- and 7-day abbreviated infections were not significantly different than non infected controls. Liveweight loss seemed to be associated with high activity of mucus peroxidase and high numbers of eosinophils in the intestinal lumen. High parasite numbers seemed to be associated with low activity of
alkaline phosphatase
in mucus. Mucus peroxidase,

Tapes philippinarum is a bivalve mollusc of the Pacific Ocean, successfully imported for human consumption into the northern Adriatic Sea (Europe). For better knowledge of its considerable adaptive ability in comparison with similar autochthonous species, a morpho-functional characterisation of its haemocytes was carried out with the establishment of short-term cell cultures (60 min at 25 degrees C). Various methods of cytochemical staining identified four cell types in the haemolymph: granulocytes (48.05% +/- 1.43), hyalinocytes (32.18% +/- 0.99), haemoblasts (18.97% +/- 0.63) and serous cells (0.8% +/- 0.19). The granulocytes, possessing cytoplasmic granules with differing dye affinity, included basophils, neutrophils and acidophils. Such granules stained vitally with Neutral Red, and correspond to lysosomes. Hydrolytic and oxidative enzymes were mainly detectable after stimulation in the presence of yeast cells. Both granulocytes and hyalinocytes were positive for

, non-specific esterase, peroxidase, and cytochrome C oxidase, whereas only granulocytes were positive for beta-glucuronidase, acid esterase, and

. Both cell types were competent phagocytes towards yeast and plasma had an opsonising effect. Moreover, the respiratory burst accompanied phagocytosis with superoxide anion production, recognisable through cytoplasmic deposits of formazan after treatment with nitro blue tetrazolium. Haemoblasts were small undifferentiated cells which, due to their morphology and positivity to the anti-CD34 antibody, show the typical features of stem cells. Serous cells, probably arising from Keber's gland and belonging to another differentiation pathway, contained non-sulphate acid mucopolysaccharides and play an important role in early defence mechanisms, taking part in the formation of clots.

The effects of the application as fertilizer during ten years of two sewage sludges (aerobically and anaerobically digested, at rates of 400, 800, and 1200 kg of N/ha yr), on the aggregate stability and contents of related organic matter components, microbial biomass and levels of five enzymatic activities (
alkaline phosphomonoesterase
, phosphodiesterase, urease,
arylsulphatase
and dehydrogenase) were investigated. The application of both sludges at mid and high rates gave rise to significant increases of organic matter, humified substances and humic acids, but no effects on carbohydrates, microbial gums and aggregate stability were observed. As for biological activity in soils, the high variability of data led to a general absence of statistical significance despite the large differences between treatments observed. Significant increases of phosphodiesterase activity were nevertheless produced by the high rate of aerobic sludge and the mid and high rates of the anaerobic sludge.

), phoshodiesterase (PDE), esterase, lipase and alanine- and leucine aminopeptidases (AP) over the pH range from 4.0 to 8.0 in modified universal buffer (MUB). Stability of the substrates for lipase (4-MUF-heptanoate) and esterase (4-MUF-acetate) measurements was poor, especially at the higher pH values. Chitinase substrate, 4-MUF-N-acetyl-beta-D-glucosamide, was unstable at high pH values whereas the substrate for

activity measurement (4-MUF-phosphate) disintegrated at low pH. The other substrates and MUF and AMC standard solutions were stable over the pH range studied. In this study, the frequency of hepatitis B (HBV) and/or hepatitis C (HCV) in cases of chronic human fascioliasis is studied. Egg count, indirect haemagglutination test (IHAT), haemoglobin level, total leucocyte and eosinophil counts, serum bilirubin, serum aspartate aminotransferase (AST), alanine aminotransferase (ALT),

The objectives of this study were to count and culture Azotobacter spp. in sampled soils, to determine the nitrogen (N) fixing capacity byAzotobacter spp. in pure culture and different soils, and to explore the relationships between N fixation capacity of Azotobacter spp. and microbiological properties of soils in Northern Anatolia, Turkey. Statistically significant relationships were found between the population of Azotobacter spp. in soils and microbial biomass C (Cmic), dehydrogenase (DHA), beta-glucosidase (GA),

) activities. However, relationships between the population of Azotobacter spp. and basal soil respiration (BSR), urease (UA) and catalase (CA) activities were insignificant. The N fixation capacities of native 3 day old Azotobacter chroococcum strains added to Ashby Media varied from 3.50 to 29.35 microg N ml(-1) on average 10.24. In addition, N fixation capacities of Azotobacter spp. strains inoculated with clayey soil, loam soil, and sandy clay loam soil during eight week incubation period were 4.78-15.91 microg N g(-1), 9.03-13.47 microg N g(-1) and 6.51-16.60 microg N g(-1), respectively. It was concluded that the most N fixation by Azotobacter spp. was in sandy clay loam soils.
