Three groups of patients received Althesin, minaxolone or di-isopropyl phenol to supplement 67% nitrous oxide in oxygen. A fourth group receiving halothane to supplement nitrous oxide in oxygen acted as a control. Hepatic function tests were measured before operation and on days 1, 3, 5 and 7 after major vascular reconstructive surgery. There were significant increases to a mean value above the upper limit of normal in aspartate amino-transferase activity by day 3 in all groups. Total lactic dehydrogenase activity increased in the patients receiving Althesin, minaxolone and halothane. No change was seen in the
alkaline phosphatase
in any of the study groups. Gamma
glutamyl transpeptidase
increased in all groups, but the mean value at day 7 was not greater than the upper limit of normal. The mean activity of ornithine carbamoyl transpeptidase showed no change in any group throughout the study period. Two of the patients receiving minaxolone suffered cholestatic jaundice during the first month. These results suggest that anaesthesia with Althesin or di-isopropyl phenol results in enzyme changes similar to those seen in a comparable group of patients receiving halothane to supplement nitrous oxide in oxygen anaesthesia.

Values for
alkaline phosphatase
and gamma
glutamyl transpeptidase
(GGTP) and the prevalence of their elevation was significantly higher in 35 patients with ankylosing spondylitis (AS) than in 35 age and sex matched controls. The abnormal enzyme levels appeared to reflect a non-specific reaction to inflammation and could thus aid in assessment of disease status.

Six-month-old calves were inoculated with 200 metacercariae of Fasciola hepatica over a 72 day period. Their aspartic transaminase,
alkaline phosphatase
, arginase and gamma
glutamyl transpeptidase
(GGT) values were measured at two-week intervals. Infections with a final fluke count of 19 to 87 flukes did not result in an increase of serum
alkaline phosphatase
or arginase compared with those in control calves. Serum aspartic transaminase values were increased significantly (P less than 0.025) above those of controls but were not increased over reference values. Seemingly, measurement of these enzymes would not be useful in assessing the degree of fluke infestation. In contrast, serum values of GGT increased 56 days after preliminary inoculation and remained high for at least 83 days after infection. The initial rise coincided with the penetration of bile ducts by migrating flukes. This relationship may be useful in further studies of fluke-induced biliary damage.

The deliberate ingestion of carbon tetrachloride by a 48-year-old woman provided an opportunity to study the sequential biochemical changes of a severe but self-limited event of hepatocellular damage. The initial phase of cellular injury characterised by high levels of serum aspartate aminotransferase, ferritin and bile acids was followed by a period of partial cholestasis. This was indicated by a marked decrease in secondary bile acids and small rises in
alkaline phosphatase
and 5' nucleotidase. Improving liver function and regeneration became evident during this period and was associated with abnormal levels of gamma
glutamyl transpeptidase
. A late rise in serum ferritin and alphafetoprotein may represent a non-specific inflammatory reaction.

A double blind, cross-over design was used to compare the acute effects of timegadine, naproxen and placebo in rheumatoid arthritis. Timegadine appeared to be more effective than naproxen in those variables related to disease activity although there were no statistically significant differences (apart from morning stiffness) where variables were examined individually. There was a tendency to a rise in
alkaline phosphatase
, and possibly gamma
glutamyl transpeptidase
, during the 2-week timegadine treatment period. No other biochemical or haematological abnormality was observed.

In experimental animals endotoxin administration causes increased levels of thromboxane B2 and prostaglandins. Liver cirrhosis is often complicated by endotoxemia. In sixteen patients with alcoholic liver cirrhosis, we measured plasma thromboxane B2 levels. In twelve patients we found on one or more occasions raised plasma thromboxane B2 levels. Raised plasma thromboxane B2 levels were associated with significantly higher serum levels of urea,
alkaline phosphatase
, gamma
glutamyl transpeptidase
and lower antiplasmin and antithrombin III levels. It is possible that some of the complications in patients with alcoholic liver cirrhosis are mediated by thromboxanes.

Chronic ingestion of ethanol (5 g/kg/day) for 6 weeks increased the hepatotoxicity of a single injection of D-galactosamine (330 mg/kg) in rats. Plasma transaminases,
alkaline phosphatase
, gamma
glutamyl transpeptidase
and sulphobromophthalein retention were consistently high in alcohol-fed rats compared to sucrose-fed controls, 25 hours after galactosamine administration. Liver histology in sucrose-fed rats revealed typical inflammatory changes and cytoplasmic vacuolation without cell necrosis was seen. Propylthiouracil treatment had no beneficial or protective effect in alcohol-fed rats in this animal model of hepatitis.

Eleven male patients with rheumatoid arthritis were treated with the anabolic steroid nandrolondecanoate and parameters of calcium and bone metabolism were studied. No changes were found in the concentrations of calcium, phosphorus or hydroxyproline in serum and urine (corrected for the creaturia). A significant decrease (P less than 0.01) was observed in the levels of
alkaline phosphatase
, but this was attributed to a change in liver enzymes, because the gamma
glutamyl transpeptidase
fell. Acid phosphatase levels showed an increase, possibly as a result of an effect of nandrolondecanoate on the prostate. No change was found during treatment in the serum levels of parathyroid hormone, calcitonin, 25 hydroxyvitamin D, 24, 25 dihydroxyvitamin D and 1.25 dihydroxyvitamin D. A seasonal fluctuation was observed for 25 hydroxyvitamin D and 24, 25 dihydroxyvitamin D, but not for 1,25 dihydroxyvitamin D. It was concluded that short-term treatment with the anabolic steroid nandrolondecanoate did not result in changes in parameters of calcium and bone metabolism.

Reference (normal) ranges for
alkaline phosphatase
, total bilirubin, urea nitrogen, chloride, cholesterol, total carbon dioxide, creatine phosphokinase, gamma
glutamyl transpeptidase
, glucose, lactic dehydrogenase, aspartate and aminotransferase, alanine aminotransferase, sorbitol dehydrogenase and triglycerides were established for cattle, sheep, pigs, ponies and ducks by use of the ABA-100 and A-Gent reagents (Abbott Laboratories). Most of the results agreed with those in the literature where methods were similar. Results were divided according to age for all species except ducks. Differences were noted between age groups with certain tests. The day-to-day reproducibility of control sera results are also presented.

The numerous physiological and nutritional factors which influence the concentration of serum calcium are considered. The causes of hypercalcaemia and hypocalcaemia are briefly discussed, with particular reference to the clinical symptoms and pathology. The effect of the acid-base status on the serum-ionized calcium level is stressed. The causes of changes in the serum concentrations of phosphorus and magnesium are briefly reviewed, along with the abnormalities of lactate, pyruvate, and hydrogen ion concentrations. The kidney function tests, blood urea nitrogen, serum creatinine, and the renal clearance tests are discussed, with emphasis placed on correlating their results with the findings from repeated urinalyses. The important physiologic influences and pathological processes which result in changes in the concentrations of these parameters are delineated. The causes of increases in the serum enzymes,
alkaline phosphatase
, alanine transaminase, asparate transaminase, lactic dehydrogenase, sorbitol dehydrogenase, glutamic dehydrogenase, gamma
glutamyl transpeptidase
, creatinine phosphokinase, amylase and lipase are discussed. The changes in serum bilirubin concentration and its components are fully described, with emphasis placed on the correlation of the findings with urinalysis data and the complexities resulting from the numerous pathologic conditions causing jaundice. These conditions are listed for each of the domestic animals. The other liver function tests, bromosulphthalein dye retention or excretion, serum uric acid and blood ammonia concentration are briefly considered. All the tests described are very useful, and frequently essential, in aiding the veterinary practitioner to arrive at a diagnosis and prognosis, but they never replace clinical acumen.
