An alkaline phosphatase which binds to hyaluronate with a high affinity has been extracted from the bovine periodontal ligament and purified. The procedure consisted of extraction with a 10 mM Tris/Mg buffer containing 0.1% Nonidet P40, chromatography on a hyaluronate-conjugated Affi-Gel 15, and fast protein liquid chromatography. The hyaluronate-binding alkaline phosphatase has an apparent molecular weight of 110,000, contained sialic acid and was more stable to heat treatment than were two other species (Mr = 120,000 and 130,000) extracted from PDL. The heat stability and influence of inhibitors on its activity show that the hyaluronate-binding alkaline phosphatase of periodontal ligament was similar to the bone alkaline phosphatase.

Dehydroepiandrosterone (DHEA) treatment is effective in preventing or delaying the onset of various genetic and induced disorders of mice and rats. Associated with the beneficial therapeutic effects exerted by action of this steroid is the development of hepatomegaly. To determine whether the changes associated with hepatomegaly also involve alterations in activities of tissue enzymes, we evaluated the effects of DHEA (0.45% in food, w/w) on hepatic protein kinases, phosphatases, and lipogenic enzymes in mice of various strains. The rates of fatty acid and cholesterol syntheses also were evaluated. DHEA administration resulted in profound changes in the sodium dodecylsulfate-polyacrylamide gel electrophoresis patterns of endogenous radiophosphorylated proteins obtained by incubation of liver homogenates with (gamma-32P]ATP. These changes were dependent upon the medium used for homogenization. Thus, when homogenates of liver tissue of DHEA-treated mice were prepared in Tris buffer containing sucrose (0.25 M) there was a marked decrease in phosphorylation of the proteins of relative molecular weight approximately 116,000 (Mr approximately 116,000), approximately 82,000, approximately 80,000, approximately 58,000, approximately 56,000, approximately 48,000, approximately 34,000, and approximately 31,000 compared with controls. With liver homogenates of DHEA-treated mice prepared in Tris buffer alone, there was a marked increase in phosphorylation of the proteins of Mr approximately 70,000, approximately 49,000, approximately 34,000, approximately 31,000, and 28,000 compared with controls. Moreover, the specific activity of kinases for endogenous protein acceptors in liver of control mice was higher than that in liver of DHEA-treated animals. The specific activities of casein kinase, cAMP-dependent protein kinase, and cGMP-dependent protein kinase remained unchanged with DHEA treatment, but the specific activity of histone kinase was increased approximately 30%. Long-term administration of DHEA also was associated with increases in the specific activities of liver AMPase and GTPase (approximately two times), but not of other nucleotidases, alkaline phosphatase, acid phosphatase, glucose-6-phosphatase, or phosphotyrosine phosphatase. The activity of hepatic NADP-linked malic enzyme was increased significantly (two to three times) by DHEA treatment of female mice of three different strains, but was unchanged in male C57BL/6 mice. The specific activities of hepatic glucose-6-phosphate dehydrogenase, NADP-linked isocitrate dehydrogenase, and ATP-citrate lyase were not affected significantly by DHEA treatment of mice. The rate of hepatic lipogenesis, determined by incorporation of tritium from 3H2O into fatty acids, was decreased approximately 70% in DHEA-treated mice, while the rate of cholesterol synthesis was increased approximately 44% compared with controls.

We have conducted studies to obtain practical knowledge regarding the stability, digestion, and analytical determination of the content of 8-hydroxy-2-deoxy-guanosine (8-OHdG) in oxidatively damaged DNA. Utilizing H2O2 plus uv light to form oxidatively damaged DNA, we found that storage of the DNA at -20 degrees C at alkaline pH caused a significant loss of 8-OHdG, whereas storage at -20 degrees C at neutral or acidic pH prevented loss of 8-OHdG. The 8-OHdG within DNA is stable at 100 degrees C for at least 15 min. Formation of 8-OHdG within DNA using uv light and H2O2 as a hydroxyl free radical-generating system yields the highest amounts when low levels of phosphate buffer are used; but the use of Tris or citrate buffers causes a lower yield of 8-OHdG because these buffers act as scavengers for the hydroxyl free radicals. Independent assessment of hydroxyl free radical flux by the use of salicylate trapping allows assessment of competitive radical reactions. Ethanol washing of plastic microfuge tubes prior to DNA enzymatic digestion improved the yield of 8-OHdG and reduced the variability between samples. Digestion of the oxidatively damaged DNA by the use of a method involving DNase I, endonuclease, phosphodiesterase, and alkaline phosphatase produced the highest yield of 8-OHdG.

A method has been developed to measure the concentration of total phosphorylated zidovudine (ZDV) inside peripheral blood leucocytes (PBLs) using a modified commercial radioimmunoassay (RIA) specific for ZDV. ZDV 5'-monophosphate was readily synthesized and used as a procedural control for RIA modification. PBLs were isolated from healthy volunteers and incubated with ZDV for 24 h to allow metabolic phosphorylation. Viable cells were counted, washed, and extracted overnight with 60% methanol. After evaporation, the extract was reconstituted in Tris buffer, pH 9.5. Because of minimal RIA antibody cross reactivity with phosphorylated ZDV, samples were split into two fractions, one of which was treated with alkaline phosphatase (AP) to liberate phosphate groups. Each fraction was then assayed for ZDV. Concentrations of phosphorylated ZDV were determined by subtracting the concentration of the non-AP-treated fraction from that of the treated fraction. Recovery of phosphorylated ZDV from cell extracts was approximately 90%, and reproducibility was acceptable (coefficients of variation less than 15% for concentrations greater than or equal to 0.25 ng/ml). Intracellular concentrations (0.1-1.4 pmoles/10(6) cells) followed a nonlinear dose-response relationship over the range 0-50 microM extracellular ZDV, with concentration-dependent saturation. These results demonstrate the feasibility of determining concentrations of phosphorylated ZDV in HIV-infected patients, a potentially key step in establishing a therapeutic range and optimal dosing regimen for these patients.

The indoxyl-tetranitro BT method for the demonstration of alkaline phosphatase activity has been optimized and its validity for quantitative histochemistry tested. The study has been performed with model films of polyacrylamide gel incorporating homogenate of rat liver and with cryostat sections from the same livers. Addition of polyvinyl alcohol to the incubation medium greatly improved the localization of the final reaction product in cryostat sections. In polyacrylamide films, the formazan production specifically due to alkaline phosphatase was highest when using a medium containing 100 mM Tris-HCl buffer, pH 9.0, 0.2-1.0 mM substrate, 0.32 mM 1-methoxyphenazine methosulphate, 10 mM MgCl2, 5 mM sodium azide and 1 mM tetranitro BT. For the incubation of cryostat sections in the presence of polyvinyl alcohol, the same medium could be used but the optimum concentrations of substrate and tetranitro BT appeared to be 1-2 mM and 5 mM respectively. The test minus control reaction was specific for alkaline phosphatase activity and could be inhibited completely with tetramisole. The test minus control reaction was linear with time up to 30 min with model films and up to 15 min with cryostat sections. The formazan production was also linear with the amount of homogenate incorporated in model films and with section thickness up to 18 micron and therefore, the reaction obeyed the Beer-Lambert law. Variation of the substrate concentration yielded a KM of 0.05 mM for aqueous media and a KM of 0.55 mM for polyvinyl alcohol-containing media.(ABSTRACT TRUNCATED AT 250 WORDS)

The conditions for staining are 0.23 mM 5-bromo-4-chloro-indoxyl phosphate, 0.55 mM tetranitro blue tetrazolium, 2.0 mM levamisole, 5.0 mM sodium azide, 10.0 mM magnesium chloride, and 0.15 mM 1-methoxyphenazine methosulfate dissolved in 100 mM Tris-HCl buffer, pH 9.5.

Small cultures of human amniotic cells were preincubated for 24 h. Human prolactin was then added to the medium. After a further short period of incubation the tubes were chilled, the medium removed and the cells rinsed with saline. The tubes then received cold Tris-sucrose and were frozen, to disrupt the cells. After thawing, adenosine triphosphatase (ATPase) and p-nitrophenyl phosphatase (PNPase) were measured. Buffer was added containing either ATP or PNP and the tubes were incubated for 30 min. Inorganic phosphate released from ATP and p-nitrophenol was measured spectrophotometrically. Prolactin stimulated both enzyme activities. The ATPase log dose-response curve was linear between approximately 12.5 and 200 mIU/l. It was inhibited by ouabain. Isobutyl-1-methylxanthine inhibited the ATPase but not the alkaline phosphatase activity. One of these human amniotic cell enzymes may provide the basis for a sensitive bioassay for human prolactin.

The effect of matrix components extracted Bovine periodontal ligament (PDL) on cell proliferation and alkaline phosphatase activity of cultured human periodontal ligament fibroblast (HPLF) were examined in order to understand the cell-tissue interaction of periodontal ligament. Bovine PDL tissue was sequentially extracted with 0.05 M Tris HCl buffer, pH 7.4, containing 1M NaCl or 4M GdmCl. After seeding 24 hours, the cultured HPLF were exposed to the extracts for two through eight days. Nine days after seeding, HPLF indicated four times high activity on ALP and 1.6 times the amount of total protein than those of control (without extract), while DNA synthesis increased only 1.2 times. On the contrary, the NaCl extract depressed the ALP activity of HPLF. The GdmCl extract enhanced both the total protein and ALP activity in dose-dependently. The ALP increasing activity of GdmCl extract on HPLF is stable to heat (78 degrees C, 20 min) and collagenase treatment but partially inactivated by trypsin digestion. Since the GdmCl extract also induced colony formation of NRK-49F cell in soft agarose, it was suggested that the extract contain EGF and TGF-beta like factor. Molecular size of this factor was estimated as 20-50Kd using Sepharose CL-6B gel chromatography. Furthermore, this factor from Sepharose CL-6B were separated into two forms by ion exchange CM-Sepharose column chromatography. Purified preparation from reversed phase column chromatography contained 14Kd, 15Kd, 17Kd, 20Kd, 28Kd40Kd, and 46Kd components on SDS-PAGE. This factor may accumulate in extracellular matrix, and may play a role of cell-tissue interaction and homeostasis in periodontal ligament.

Saito et al recently reported that the alkaline phosphatase (ALPase) of human periodontal ligament fibroblasts (HPLF) showed remarkably high activity which was similar, but not identical phenotype, to that present in osteoblasts, and suggested that HPLF could be termed as "osteoblastic fibroblast." The present study attempts to explore the ALPase synthesized on HPLF in relation to 1,25(OH)2D3. These HPLF were obtained by the explantation method and then subcultured in D-MEM containing 2 mg FCSP/ml, 50 micrograms ascorbic acid/ml and penicillin/streptomycin after trypsinization. The HPLF were inoculated at a cell density of 1.25 x 10(4) cells/cm2 in culture wells. After 24hr, the HPLF were treated every two days for 7 days with 0.5-10nM 1,25 (OH)2D3. Then, ALPase activity, DNA and protein contents were assayed by the methods using p-nitrophenylphosphate, diaminobenzoic acid and Coomassie Brilliant Blue, respectively. Also, ALPase was prepared from the confluent HPLF incubated with 5 nM 1,25 (OH)2D3 for 12 days, and digested with and without trypsin. The crude ALPase which was solubilized with 10mM Tris-HCl, pH 7.4 containing 0.2 mM MgCl2 and 0.1% NP-40 was applied to 5-15% gradient SDS-PAGE and stained with beta-naphththylacid phosphate and First Blue BB salt in 60 mM borate buffer pH 9.7. The cell growth which was assayed by DNA contents and the incorporation of 3H-thymidine was decreased by 1,25(OH)2D3. On the other hand, ALPase activity was increased approximately 3.6 fold at 6 day by the addition of 5 nM 1,25(OH)2D3. From the separation of ALPase activity on SDS-PAGE, 110 K and 120-130 K ALPase were identified. The 110 K ALPase, which was not changed by 1,25(OH)2D3, was converted to 100K, releasing 10K peptide after trypsin treatment. This 110K ALPase might be tightly associated with cell membrane structure. The 120-130K ALPase was remarkably increased by 1,25(OH)2D3 on SDS-PAGE and completely digested with trypsin. The ALPase in the cultured HPLF might be located not only on the plasma membrane but also in the extracellular matrix. Therefore, 1,25(OH)2D3 may regulate the cell cycle and also the gene expression of ALPase of HPLF.

Phosphorylated rabbit cardiac alpha alpha-tropomyosin has been prepared either enzymatically (Montgomery, K., and Mak, A.S. (1984) J. Biol. Chem. 259, 5555-5560) or by fractionation of the phosphorylated and nonphosphorylated forms on a Mono Q column in 9 M urea, 50 mM Tris, pH 8.0. Although the phosphorylated and nonphosphorylated forms showed no difference in their F-actin binding properties, the phosphorylated protein had substantially higher viscosities at low ionic strengths, indicating a greater propensity for head-to-tail interaction. Similar measurements showed the strengthening of this interaction by whole troponin to be substantially reduced by phosphorylation even though the binding of whole troponin and troponin T to tropomyosin was demonstrated by affinity chromatography to be, if anything, strengthened by phosphorylation. In a reconstituted actin (4 microM) plus myosin subfragment 1 ATPase assay (50 mM ionic strength), significantly higher activities over a range (1 to 8 microM) of subfragment 1 concentrations were observed with phosphorylated tropomyosin compared with the nonphosphorylated protein. In the fully reconstituted system with troponin, there was no significant difference in the inhibition of ATPase in the absence of Ca2+. However, in its presence, the activities were appreciably increased with the phosphorylated tropomyosin compared to those with the nonphosphorylated form. These differences were eliminated by treatment of the phosphorylated tropomyosin with alkaline phosphatase. This is the first demonstration of an effect of phosphorylation on the functional properties of tropomyosin.
