Bone marrow involvement was seen in 11 percent of patients with Hodgkin's Disease which was determined from pre-treatment biopsy specimens using established histopathologic criteria. Analysis of 32 evaluable patients with marrow involvement showed male preponderance with a peak in fourth decade of life. Twenty four cases (75%) had B-symptoms and 15 (46%) presented within six months of onset of symptoms. On categorizing for clinical staging, 21 (65%) belonged to stage III and IV. Hepatomegaly (greater than or equal to 2 cms) was present in seven cases (21%) and splenomegaly in 13 cases (40%). Mixed cellularity and lymphocytic depletion histopathologic subtypes showed the highest frequency of involvement (21 cases; 65%). Out of 28 cases ESR was raised in 27 cases (96%). Eighteen cases (56%) showed elevated serum
alkaline phosphatase
levels. Serum
copper
levels were determined in 14 cases, out of which 12 (85%) showed elevated levels. These parameters along with anemia (hemoglobin of 12 g/dl or less) in 26 cases (81%), correlated well with the disease activity. Only four cases had leukopenia at presentation pointing to no hindrance for aggressive chemotherapy. All cases received minimum of six courses of standard combination chemotherapy with or without local radiotherapy. Sixteen cases (50%) relapsed subsequently and were managed accordingly. A five year follow-up revealed a minimal 31 percent overall survival, and 18 percent of patients were disease free and well since the time of treatment.(ABSTRACT TRUNCATED AT 250 WORDS)

This experiment was carried out to investigate the effects of intravenous injection of an essential trace-element preparation (TE-5) on iron, zinc,
copper
and manganese concentrations, and blood biochemical and hematological parameters in rats. The rats were treated by intravenous injection of TE-5 for 7 days and the following results were obtained: 1) Neither a 0.04 nor a 0.4 ml/kg/day injection of TE-5 affected the iron, zinc,
copper
and manganese concentrations in tissues. 2) At a higher dose (1.2 ml/kg/day), iron concentrations in liver and spleen, zinc concentrations in liver, kidney, tibia and plasma,
copper
concentrations in heart, kidney and whole blood, and manganese concentrations in brain, heart, spleen, kidney, femoral muscle, tibia and whole blood increased. 3) At the highest does (4 ml/kg/day), all rats died and iron, zinc,
copper
and manganese concentrations in tissues increased remarkably. 4) With injections of TE-5 (0.04-1.2 ml/kg/day), hemoglobin, hematocrit,
alkaline phosphatase
activity and blood urea nitrogen decreased slightly. These results suggest that iron, zinc,
copper
and manganese concentrations, and blood biochemical and hematological parameters are maintained at doses up to 0.4 ml/kg/day of TE-5, but that doses higher than that destroy homeostasis.

In order to determine the specific action of cadmium on bone metabolism, the effect of cadmium on
alkaline phosphatase
activity, a marker enzyme of osteoblasts, was compared with that of other divalent heavy metal ions, i.e., zinc, manganese, lead,
copper
, nickel and mercury (10 microM each), using cloned osteoblast-like cells, MC3T3-E1. Cadmium had the strongest inhibitory effect on
alkaline phosphatase
activity of the cells among the metals tested. At the same dose, however, cadmium failed to inhibit cellular glucose-6-phosphatase and lactate dehydrogenase activities, suggesting that the inhibitory effect of cadmium on
alkaline phosphatase
was specific and was not dependent on cell injury. Cadmium treatment caused a significant decrease in cellular zinc level, but mercury treatment had no such effect at the dose inhibiting
alkaline phosphatase
activity. There was a good correlation between decrease of cellular zinc level and inhibition of
alkaline phosphatase
activity in cadmium-treated cells. Concomitant treatment of the cells with zinc prevented the cadmium-induced inhibition of
alkaline phosphatase
activity. However, this was not the case in the mercury-induced inhibition. Cadmium also inhibited the mineralization of osteoblasts. When 10 or 20 microM zinc was concomitantly added to the cultures, the inhibition of mineralization was prevented. These data suggest that the inhibitory effect of cadmium in osteoblasts may be closely related to its influence on the cellular zinc metabolism.

The Authors studied the changes of 17 blood parameters of calves before, during and after a traditional weaning. Concentrations of total protein, albumin, urea nitrogen, creatinine, glucose, cholesterol, triglycerides,
alkaline phosphatase
, total bilirubin and phosphorus were determined by an automated assay system; sodium, potassium and chloride by a flame photometer; calcium, magnesium,
copper
and zinc by an atomic absorption spectrophotometer. Observed changes can be referred to: 1) the change of the digestion (glucose); 2) the age of the subjects (
alkaline phosphatase
); 3) the increase of weight (creatinine); 4) the type of feeding (phosphorus,
copper
and zinc). Sodium, potassium, chloride, calcium and magnesium did not change during the weaning.

Myo-Inositol hexaphosphate (phytic acid), a naturally occurring plant constituent capable of forming coordinate complexes with polyvalent cations, reversibly inhibits the hydrolysis of p-nitrophenyl phosphate by
alkaline phosphatase
. Kinetics of the inhibition is of the strictly competitive type with Ki equal to 260 microM at pH 8.0 and 25 degrees. A similar result is also obtained in the presence of an inositol hexaphosphate-cupric ion coordinate complex. This inhibition of substrate hydrolysis by either inositol hexaphosphate or its
Cu(II)
complex is to be differentiated from the effect of the latter on
alkaline phosphatase
in the absence of substrate. In this case total inhibition of the enzyme is obtained in a time-dependent process which apparently involves a metal ion exchange reaction, i.e., substitution of the enzyme's zinc atoms for
copper
atoms. The effect of inositol hexaphosphate on
alkaline phosphatase
contrasts with that of inorganic phosphate which gives inhibition kinetics of the partially competitive type with Ki (for its effect on KM) 17.1 microM; a value close to that of KM for the substrate (15.2 microM). At a low phosphate concentration relative to that of inositol hexaphosphate in the assay solution the observed inhibition was partially additive. At a higher phosphate to inositol hexaphosphate ratio, the inhibition was essentially a phosphate effect. This provides additional support for the conclusion that Inositol hexaphosphate and, by extension, an inositol hexaphosphate-
Cu(II)
complex inhibits
alkaline phosphatase
by interaction at the active sites in competition with the substrate.

Metabolic and hormonal responses of eight adult male collared peccaries (Tayassu tajacu) to an ad libitum diet intake, or 25% of an ad libitum intake, were examined. Blood samples for hematological, serum-biochemical and hormonal profiles were collected at three week intervals during the nine week experiment starting 4 August 1983. Males fed on the restricted diet lost an average of 26% of their body weight during the trial, compared to a slight weight gain for those fed ad libitum. Characteristics of the red and white blood cell populations were not influenced by diet intake, with the exception of mean corpuscular volume, which was consistently lower amongst males fed on the restricted diet. Restricted food intake resulted in significantly elevated serum values for urea nitrogen, urea nitrogen:creatinine, urea index, alpha globulin:beta globulin, gamma globulin:albumin, nonesterified fatty acids,
alkaline phosphatase
and lactate dehydrogenase isozymes (LD1 and LD2). Restricted food intake resulted in significantly lowered serum values for total alpha globulin, alpha-1 globulin, total beta globulin, beta-1 globulin, beta-2 globulin, glucose, triglycerides, calcium, magnesium, sodium, chloride,
copper
and triiodothyronine. Serum levels of creatinine, total protein, albumin, alpha-2 globulin, uric acid, total bilirubin, cholesterol, aspartate aminotransferase, alanine aminotransferase, gamma glutamyltransferase, lactate dehydrogenase, phosphorus, calcium:phosphorus, potassium, iron, zinc and thyroxine were unaffected by diet intake level. Semen evaluation indicated spermatogenesis was not affected by dietary restriction despite reductions in scrotal circumference and ejaculate gel volume. Serum testosterone levels were significantly lower among males fed on the restricted diet after nine weeks. These data suggest male libido might be depressed during poor range conditions, while maintenance of spermatogenesis might permit them to take immediate advantage of improved range conditions. Blood analysis of metabolic and hormonal function can provide useful information for predicting the adult male's nutritional and reproductive condition.

Time dependent changes in urinary biochemical indicators for renal tubular injury and dysfunction were determined in female Wistar rats after an intraperitoneal injection of cadmium-metallothionein (Cd-MT) (50, 150 or 300 micrograms Cd/kg body wt) to further characterize the tubular damage caused by Cd. The Cd-MT injection caused dose-dependent increases in urinary activities of the enzymes (
alkaline phosphatase
; gamma-glutamyl transpeptidase; lactate dehydrogenase, LDH; N-acetyl-beta-D-glucosaminidase) on day 1, which appeared to reflect the tubular injury. The rate of increase in LDH, a cytosolic enzyme, was the largest among those of the enzymes. This result coincided with the data reported for repeated administration of ionic Cd to rats, suggesting that the feature of tubular injury caused by an injection of Cd-MT is similar to that by chronic exposure to ionic Cd. Changes in urinary glucose and total protein, indicators of tubular dysfunction, and metals (Cd, zinc and
copper
) were accompanied with those in urinary enzymes. Hydrocarbons in breath of rats injected with Cd-MT at a dose of 300 micrograms Cd/kg body weight were also determined as an indicator of in vivo lipid peroxidation. The levels of ethane and propane were significantly increased at 12 h after injection, which suggests that lipid peroxidation is partly involved in the tubular damage reflected by the increases in urinary enzymes.

The prognostic value of the aminoterminal propeptide of type III procollagen (P3NP) was investigated in 63 patients with primary biliary cirrhosis (PBC) followed for up to 87 months. No patient with an initially normal serum P3NP level died during the study; survival was significantly worse with increasing serum P3NP levels. Cox multivariate analysis confirmed that serum P3NP was an independent prognostic variable. Positive correlations were found between serum P3NP and histological stage, pericellular fibrosis, piecemeal necrosis, and serum concentrations of alanine aminotransferase and aspartate aminotransferase. Raised P3NP levels also correlated with the degree of cholestasis as evaluated by serum bilirubin, serum
alkaline phosphatase
, and
copper
binding protein deposition in the liver. Serum P3NP is of prognostic value because it reflects the major pathophysiological features of PBC.

A double-blind study was done giving 10 mg of
copper
/day as
copper
gluconate or placebo capsules for 12 wk. The seven subjects receiving
copper
gluconate had no change in the level of
copper
in the serum, urine, or hair. There was also no change in the levels of zinc or magnesium. There was also no significant change in levels of hematocrit, triglyceride, SGOT, GGT, LDH, cholesterol, or
alkaline phosphatase
. The side effects of nausea, diarrhea, and heartburn were the same in the subjects receiving
copper
gluconate and subjects receiving placebo capsules.

Activities of the enzymes lactase, sucrase, maltase,
alkaline phosphatase
, and superoxide dismutase (SOD) were measured in mucosa of duodenum and ileum of the rat after 70% resection of mid-small intestine or sham operation (transection). We also measured the concentrations of zinc,
copper
, and manganese in several tissues to assess trace metal homeostasis postresection. Resection resulted in decreased specific activities of disaccharidases and
alkaline phosphatase
in duodenum, while specific activities remained unchanged in ileum. Specific activity of total SOD (the sum of Cu-Zn and Mn SOD) and Mn SOD was the same in duodenum after resection but was markedly increased in ileum. Tissue trace metal concentrations changed minimally. Because of postresection mucosal growth, total segmental activity of disaccharidases and
alkaline phosphatase
was the same in duodenum and increased in ileum of resected compared to transected rats. Segmental activity of total SOD and Mn SOD doubled in duodenum and trebled in ileum of resected as compared to transected rats. Thus, total segmental enzyme activity is maintained or increased postresection by increased enterocyte proliferation rate and mucosal growth.
