An acidic polysaccharide fraction composed of glucose and N-acetylmannosaminuronic acid with a small portion of peptidoglycan was isolated by enzymic digestion and subsequent ECTEOLA-cellulose chromatography from the cell walls of Micrococcus lysodeikticus. On mild acid treatment, the fraction became Morgan-Elson positive and formed the Morgan-Elson chromogen on heating with phosphate buffer (pH 7). The product of mild acid treatment released inorganic phosphate on treatment with phosphomonoesterase. After gel-chromatography on Sephadex G-25 and DE-32, the acidic polysaccharide fraction contained less glucosamine than muramic acid. By reduction of this fraction with borohydride, a part of the glucosamine was converted into glucosaminitol. Based on these results, it is suggested that the acidic polysaccharide is linked to glucosamine by a (1-3) linkage, which is linked to the 6 position of a muramic acid residue by a phosphodiester linkage.

Cerebrospinal fluid (CSF) samples were collected from the atlanto-occipital (AO) and lumbosacral (LS) subarachnoid spaces of 24 horses and 21 ponies that had no clinical evidence of neurologic disease. Depth of needle insertion, pressures, refractive index, rapid reagent strip test (protein, glucose, blood, pH) results, cell counts, content of protein, glucose, sodium, potassium, chloride, calcium, phosphorus, urea nitrogen, and cholesterol, and activities of creatine phosphokinase, aspartate transaminase, lactic dehydrogenase, and alkaline phosphatase were determined. The resulting clinical reference values obtained were discussed in light of the published normal values for CSF from horses, other animals, and man. White cell counts in CSF were found to be from 0 to 6/microliters. Values for protein content were distributed between wider limits than previously reported values. The LS-AO difference is proposed as a criterion for clinical evaluation of CSF protein content. Ponies were found to have more protein in their CSF than did the horses, and CSF from the LS site contained more glucose than that from the AO site. The CSF electrolyte composition was similar to that of previous reports. Enzyme activities in equine CSF are reported for the 1st time.

The normal values of serum glutamic oxaloacetic transminase, serum glutamic pyruvic transaminase, serum lactic dehydrogenase and serum alkaline phosphatase, total protein, urea, creatine, cholesterol, glucose, magnesium, calcium and inorganic phosphorus were measured monthly over a 12-month period from 10 "pure" Native and 10 Native X British Crossbred pigs. Except for cholesterol, no significant difference was found between the two groups. Similar estimations were made for 5-month and 11-month Village pigs in which the serum alkaline phosphatase, inorganic phosphorus, total protein, urea, creatinine and calcium were significantly lower when compared with the corresponding age group of the pure Native pigs and Crossbred pigs. These lower values are thought to be due to the effects of the malnutrition-parasite complex of Village pigs.

This report summarizes a one year evaluation of Abbott's ABA 100, with respect to mechanical parts (syringe plates, precision and linearity of photometry, band width of several filters, multicuvet precision, temperature control) and the reliability of several methods (endpoint procedures: determination of the glucose concentration with hexokinase- and the glucose dehydrogenase method, and of the protein concentration; enzyme activities: alanine and aspartate aminotransferase, creatine kinase, alkaline phosphatase). The critical batch size was estimated as an indicator of economy (about 40 samples per day for the glucose concentration). Various aspects of the instrument are discussed with respect to its use in clinical chemistry.

Changes in serum biochemical values as a function of serum-clot contact time were investigated. Time extending to 48 hours were studied by drawing sets of six specimens from ten subjects. Glucose, lactic dehydrogenase, the transaminases, potassium, alkaline phosphatase, and iron showed significant changes. Other common tests, including electrophoretic separations, were not affected.

Serum chemistry test results were correlated with discharge diagnoses of 4,295 persons admitted to an acute care general hospital in a seven-month period. The diagnostic value of the test results, separately and in combinations of up to 12 tests, were calculated at each of ten levels as measured from the mean of normal in standard deviations. All patients had a serum profile consisting of total calcium, inorganic phosphorus, glucose, urea nitrogen, uric acid, cholesterol, total protein, albumin, total bilirubin, alkaline phosphatase, lactic dehydrogenase, and glutamic oxaloacetic transaminase levels. Analysis by manual and computer technics is illustrated. Computerized analysis of a chemistry profile is reported as a list of diagnostic possibilities in ranking order, with the calculated probability for each. In this preliminary report individual diseases are replaced by categories of disease taken from The International Classification of Diseases, Adapted (ICDA).

The author carried out experiments on white rats and discussed the role of the placental insufficiency in the perinatal pathology under the action of fungicide basfugine. After administration of the preparation singly at the critical 13th day of embriogenesis and repeatedly during the course of the gestation the author examined biochemically the activity of the following enzymes: glucose-6-phosphatdehydrogenase, lactatdehydrogenase and thermostable alkaline phosphatase. Basfungine, administered in effective teratogenic doses, inhibited the activity of the indicated enzymes in the placenta, manifesting in this way its functional insufficiency, which was most probably the substantial moment in the pathogenesis of the induced anamaly in the fetal development.

Blood samples from rats, rabbits and beagles were subjected to simultaneous analyses for ten parameters using the RaBA-System and the results were compared with those obtained by the routine manual methods. Although the mean values measured by the two methods differ in several items, a significant correlation between the two methods in the majority of the determinations makes the use of the RaBA-System acceptable. The RaBA-System is thus considered to be applicable to evaluation of hemoglobin, total protein, glucose, BUN, total cholesterol, GOT, GPT, and alkaline phosphatase in blood samples from rats, rabbits and beagles. Plasma LDH and albumin, however, cannot be determined with accuracy by the system.

In order to establish the normal baseline of blood values in rats using the RaBA-System, the following eight parameters were analyzed in 8-22-week-old Wistar-Imamichi rats of both sexes: hemoglobin, total protein, glucose, BUN, total cholesterol, GOT, GPT, and alkaline phosphatase. With the exception of glucose, all these parameters were significantly different depending upon ages. Significant sex differences were observed in the values of hemoglobin, total cholesterol, GOT, and alkaline phosphatase.

Cells of Escherichia coli strain B develop large intracellular vacuoles and exhibit an abnormally long lag phase when inoculated into a defined medium to glucose and salts containing 3 times 10-6 M Cd2+. Early in this lag, about 95% of the cells fail to form colonies when plated on nutrient broth-NaCl-agar. Prior to the initiation of proliferation, the morphology of these cells becomes normal. They regain viability in the absence of deoxyribonucleic acid replication. The rate and extent of growth are normal once proliferation begins. This reversible phenomenon of accommodation to a growth-inhibiting concentration of Cd2+ does not appear to result from a selection of mutant cells. Cells which are proliferating in the presence of Cd2+ accumulate the ion to a very high concentration. In membranes and 31% in the cytoplasm. In unaccommodated cells, the figures are 2%, 75%, and 23%, respectively. The activity of alkaline phosphatase, a zinc-metalloenzyme which is inhibited by cadmum and is located between cell wall and membrane, is not abnormally low in accommodated cells, suggesting that the cadmim is compartmentalized in these cells. Molecular sieve chromatography of cell extracts shows that the Cd2+ is associated with two classes of macromolecules. It appears that accommodation of E. coli to the presence of Cd2+ involves exclusion of the ion from the cell and reversal of damage caused by prior exposure to the ion.
