The thymol turbidity test (Macglan's test) and the determination of serum cholesterol are still performed routinely as liver function tests in many laboratories. In this report, we have employed quantitative methods in order to evaluate the real usefulness of these two parameters. In a first step, the value of the thymol test and of cholesterol determination for the discrimination of the 13 most frequent hepato-biliary diseases was studied by analysis of variance, and compared to that of the following tests : serum bilirubin, serum glutamic pyruvic transaminase, serum alkaline phosphatase serum protein electrophoresis and prothrombin time. It was found that of all these parameters, the thymol test and cholesterol measurement had the lowest discriminatory powers. In a second step, the consequences of the suppression of the two tests were examined by linear discrimination analysis, which was done for all the possible pairs of diseases. It appeared that in each case, the loss of information due to the elimination of the thymol test or of cholesterol determination was nil or negligible. We conclude therefore that the thymol turbidity test and determination of serum cholesterol should be abandoned as routine tests of liver function.

Commercial feed mixture was buffered with a 2% and 3% admixture of bentonite buffer in two beef cattle herds in the course of one year. The mixtures were fed on a continuous basis. The two-per-cent buffer concentration was tested in 110 test animals with 104 control animals and the three-per-cent concentration in 50 test animals with 50 controls. Throughout the trial the over-all health condition remained unchanged, the hematocrit and hemoglobin values were balanced in both groups. The biochemical indices were better in the test groups: hypocalcemia improved (in the controls it grew worse), magnesiemia was slightly increased, the inorganic serum factor did not go beyond physiological limits, and acidosis did not occur (as distinct from the control animals). The levels of transaminases (GOT, GPT), glutamic acid dehydrogenase, total serum protein, alkaline phosphatase as well as ammonia and urea in blood serum were at physiological values with po-differences within groups. In the case of the three-per-cent buffer concentration the daily gains were higher by 0.073 kg, and in the two-per-cent concentration by 0.058 kg, in the test animals. The average annual gain was higher by 25.5 kg, and by 18.3 kg, respectively. With respect to the price of buffer and to the efficiency of the animals tested, the economic indices of feed mixture buffering are highly effective.

In six animals hunted and four immobilized animals of red deer (Cervus elaphus L.) of both sexes and of different age, kept at three game preserves in Bohemia, the psychological values were ascertained: the number of erythrocytes and leucocytes, the hemoglobin content, the hematocrit, MCV, MCH, MCHC, the white blood count, the total serum protein, the fractions: albumin, alpha-, beta- and gamma-globulin, the activity of SGOT, and SGPT, the alkaline phosphatase and the metabolism of calcium, phosphorus, and magnesium in the blood serum. The blood from the heart shortly after killing of the animal was examined, and in immobilized animals the blood was taken from the vena jugularis. In the red deer, a variable intensity of the polyvalent infection of parasites of the species Dictyocaulus viviparus, Bicalulus sagittatus, Fascioloides magna, Paramphistomum spec., Haemonchus contortus, Ostertagia ostertagi, O. circumcincta, Trichostrongylus axei, Nematodirus filicollis, Chabertia ovina, and Trichocephalus globulosa was found. In the immobilized deer no marked deviations were found in the examined values of blood that had been taken within 10 minutes after the calming of the animals.

Forty-seven male Macaca mulatta, 3 to 4 kg weight, were inoculated intravenously or subcutaneously with various doses of yolk sac-grown Rickettsia rickettsii. Thirty-four macaques became febrile and exhibited signs of infection ranging from transient illness with a few days of fever to severe illness with subsequent death. The rash appeared more frequently in the macaques inoculated subcutaneously. Febrile macaques that survived had leukocytosis, with concomitant neutrophilia. Febrile macaques that died had, in addition, marked terminal leukopenia and thrombocytopenia. Packed cell volume of all febrile macaques decreased. In almost all of the febrile macaques, there were increased serum urea nitrogen, glutamic-oxaloacetic transaminase, and lactate dehydrogenase and decreased total serum protein and amylase concentrations. A few febrile macaques had increased bilirubin values and decreased sodium, chloride, phosphorus, and alkaline phosphatase concentrations. Changes did not occur in serum glucose, potassium, calcium, and glutamic-pyruvic transaminase values. The experimental form of Rocky Mountain spotted fever in the macaque provides a subhuman primate model for studying the pathophysiology of this disease.

Twenty calves were infected with 1000 metacercariae of Fasciola hepatica, the activities of 10 enzymes in plasma or serum were assayed and concentrations in serum of proteins, urea and bilirubin were determined. These values were compared with control data obtained from 14 uninfected calves. Aspartate aminotransferase, lactate dehydrogenase, sorbitol dehydrogenase, glutamate dehydrogenase, ornithine carbamoyl transferase and gamma-glutamyl transpeptidase activities increased in infected calves. Total serum protein increased, albumin decreased, globulin increased and the albumin/globulin ratio was decreased in infected calves. Plasma alanine aminotransferase, leucine aminopeptidase, alkaline phosphatase and cholinesterase activities and serum concentration of urea and bilirubin were unaffected. It was concluded that glutamate dehydrogenase and gamma-glutamyl transpeptidase were the most sensitive indicators of liver cell damage in fascioliasis.

A 48-year-old woman with severe carcinoid syndrome caused by multiple liver metastases was treated by hepatic dearterialization. After the operation, urinary 5-HIAA excretion fell transiently to normal levels and the patient was symptom-free for six months. After one year the symptoms of carcinoid syndrome had recurred and a re-dearterialization was performed. A favourable clinical effect was again achieved, but the relief of symptoms lasted only six months. The changes in urinary 5-HIAA excretion and SGOT, SGPT, bilirubin, alkaline phosphatase, cholesterol, PP activity as well as serum protein levels were uniform after both operations to an astonishing degree. Histological specimens taken at reoperation showed that the carcinoid metastases in the anterior parts of the liver were largely necrotic. The necrotic areas has peculiar features, e.g. the necrosis was partly incomplete and ghost like carcinoid structures could be seen.

Studied were the changes in the values of some hematologic indices (hemoglobin, erythrocytes, leukocites) as well as the values of some biochemical (total protein and protein fractions, urea, total lipids, bilirubin, inorganic, phosphorus calcium) and enzyme factors (lactatedehydrogenase, alkaline and acid phosphatase, aldolase, amino transferrases, creatinephosphokinase) in geese prior to and after experimental infection of Borrelia anserina. It was found that in the peak stage of spirochetemia the content of hemoglobin, total lipids, and calcium, the percent of albumins and alfa-globulins, and the activity of the alkaline phosphatase in the blood decreased. The prealbumin fraction of the serum protein was completely reduced. The activity of the lactatedehydrogenase, acid phosphatase, amino transferrases, and aldolase showed higher values that were statistically significant, while the activity of the creatinephosphokinase rose to a slighter extent. The urea, bilirubin, inorganic phosphorus, and the alfa-, beta-, and gamma-globulins correlated positively with the course of the infection.

LDH isoenzymes and heat-stable alkaline phosphatase were studied in the serum and placental extract of 20 cases of pre-eclampsia and 10 normal pregnancies as a control. The starch-gel electrophoretic serum and placental isoenzymogram showed that LDH4 and LDH5 were the main isoenzymes in the placenta while LDH1 and LDH2 were the main isoenzymes in the serum in pre-eclampsia. The electrophoretic serum protein pattern in pre-eclamlobulins with decreased albumin fraction, while in the placenta, the albumin fraction was increased together with a decrease in the alpha-globulins. The electrophoretic pattern of serum alkaline phosphatase showed a main band of activity at the B-globulin zone in all cases of normal pregnancy and pre-eclampsia. In the placenta, two additional bands were detected.

Clinical and laboratory findings from 15 patients with icteric viral hepatitis during pregnancy (VHP) and from 22 patients with intrahepatic cholestasis during pregnancy (CJP) were evaluated statistically in order to find out which parameters might help in order to find out which parameters might help in differentiating the two diseases. Diagnosis was established by needle liver biopsy in all cases. The following data were considered: history, physical examination, erythrocyte sedimentation rate (ESR) serum cholesterol, prothrombin time, total serum bilirubin, SGOT, SGPT, serum alkaline phosphatase, serum protein, serum flocculation tests, BSP blood clearance and serum HB Ag. Vomiting, high GOT and GPT serum levels, and serum HB Ag positivity suggest VHP diagnosis. Otherwise a severe itching with scratching lesions, high ESR, elevated total cholesterol and serum alkaline phosphatase values mainly if occurring in the later stage of pregnancy are consistent with CJP diagnosis. When clinical and laboratory data from a jaundiced pregnant female do not allow diagnosis, this can be established only on the basis of needle liver biopsy.

In order to assess the functional state of the liver in 45 repair service workers of a chemical plant producing pesticides the serum concentration and electrophoretic pattern of proteins, the concentration of bilirubin and the activity of alkaline phosphatase, gamma-glutamyl- transpeptidase, alanine and aspartate aminotransferase, and lactic and malic dehydrogenase were determined. As compared to 35 healthy controls, not exposed to noxious chemicals, a significantly lower serum protein concentration with higher percentage of gamma-globulins and lower albumins and alpha 2-globulins were observed, the serum alanine and aspartate aminotransferase activities were significantly elevated. Ultrasound examination of the hepatic structure revealed liver steatosis in 11 (24.4%) workers. The results of our study point to a discrete lesion of the liver.
