1. This experiment was designed to study the effects of fasting and enforced exercise on the physiological, biochemical and physical characteristics of duck muscle. 2. Sixty 75-d-old male ducks weighing 3.0 +/- 0.2 kg were assigned to three treatments: a control, and an 8 and 24 h fast plus enforced exercise for 10 min. The ducks were then sacrificed and the carcass stored at 4 degrees C for 24 h. 3. Although the pH and serum lactate contents gradually increased with fasting time the responses were not significant. The ultimate pH was elevated and the lactate of breast and thigh muscles was lower in stressed birds. 4. The activity of lactic dehydrogenase was significantly increased by the stress, and the activities of
creatine phosphokinase
and
alkaline phosphatase
were also increased slightly. However, no effect was found on the ATPase activity of the myofibrillar protein of either breast or thigh muscle as a result of the stress. The ATPase activity of myofibrillar protein of breast muscle significantly increased with storage time. 5. The extractability of myofibrillar protein increased with storage time for all treatments. The SDS-PAGE patterns of myofibrillar proteins were also studied. 6. Consequently DFD-like muscle was observed in the breast and thigh muscles of ducks which had been stressed.

1. The effects of
alkaline phosphatase
on platelet aggregation, secretion and thromboxane B2 (TxB2) generation induced by the full dose-range of common platelet agonists were studied in human platelet-rich plasma and washed platelets. 2. Platelet aggregation and adenosine 5'-triphosphate (ATP) secretion induced by threshold and supramaximal concentrations of arachidonate and stable TxA2 and prostaglandin endoperoxide-mimetics (compounds U46619 and EP171) were abolished in the presence of
alkaline phosphatase
(0.5-1 u ml-1), even though the synthesis of TxB2 persisted. In contrast, platelet aggregation by PAF-acether and by supramaximal concentrations of thrombin as well as the primary wave of aggregation by adenosine diphosphate (ADP) and adrenaline were unaffected by
alkaline phosphatase
under conditions where the secondary wave of aggregation by ADP was blocked. 3. Alkaline phosphatase, unlike prostacyclin, failed to raise the adenosine 3':5'-cyclic monophosphate (cyclic AMP) content of the platelets. Also, the pretreatment of platelets by inorganic phosphate or by ATP plus creatine phosphate/
creatine phosphokinase
reversed the inhibitory effect of
alkaline phosphatase
. 4. Experiments performed in the guinea-pig in vivo showed that
alkaline phosphatase
was effective on thrombocytopenia induced by arachidonate. 5. Our results provide the first direct evidence for a specific inhibitory effect of
alkaline phosphatase
at a site sensitive to TxA2 and prostaglandin endoperoxides and suggest that its phosphorylation/dephosphorylation state may play an important role in modulating platelet activation. These results also suggest the presence of ecto-protein kinases on membrane platelets.

Seventy-four patients' sera containing macroenzymes or other uncommon enzyme forms were analyzed for total
creatine kinase
, lactate dehydrogenase,
alkaline phosphatase
, or amylase activity on an Ektachem 700XR analyzer. The Ektachem test results correlated well with activities obtained by means of liquid reagents on RA-1000. For each enzyme tested, linear-regression analysis yielded data comparable with those for sera containing exclusively the more common isoenzymes. Increased concentrations of intestinal ALP, however, resulted in higher activities of total ALP being reported for the Ektachem.

1. Analysis of biochemical parameters was carried out on material pooled from 10 baboons, adult male and female, of the Papio c. cinocephalus strain. 2. The values determined were for the common metabolites and enzymes utilized for specific studies in general metabolism (urea, glucose, cholesterol, phospholipids, bilirubin, uric acid, creatinine,
alkaline phosphatase
, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactic dehydrogenase, leucine amino peptidase, gamma glutamyl transpeptidase, total
creatine kinase
and amylase). Total protein and its fractions (ratio albumin/globulin, albumin, alpha 1-globulin and alpha 2-globulin) were also determined. 3. Results of these studies were compared with values for normal human adults. 4. The differences obtained between the respective ranges can be explained by the different physiology of the two species and, possibly, may also be caused by the use of human substrates in the enzyme assays. 5. Through the determination of normal values for the baboon, this study provides values for this species as an experimental animal in biomedical research.

The effects of ZK 36374, a prostacyclin analogue and UK 38485, a thromboxane synthetase inhibitor were studied in guinea pigs after performing mesenteric arterial occlusion. In this study, while ZK 36374 significantly lowered the
alkaline phosphatase
and
creatine phosphokinase
values two hours after mesenteric arterial occlusion when compared with the control group (p less than 0.005), UK 38485 did not induce any change. In guinea pigs, when given together, ZK 36374 and UK 38485 lowered the enzyme levels to preligation values and the difference was nonsignificant (p greater than 0.1). The histopathologic investigation of the small intestine after giving ZK 36374 and UK 38345 together revealed minimal changes. These findings stress the importance of preserving the PGI2 levels in the PGI2/TXA2 ratio in preventing the increase of lysosomal enzyme levels and histopathologic changes after mesenteric arterial occlusion in guinea pigs.

The time course of changes in serum proteins and other blood constituents after eccentric exercise of the forearm flexors by six nonweight-trained female subjects (age, 19.7 +/- 1.9 years) was investigated. Eccentric muscle actions are those in which the muscle lengthens as it exerts force, as when a person lowers a weight. Serum levels of
creatine kinase
, lactate dehydrogenase, aspartate aminotransferase, alanine aminotransferase,
alkaline phosphatase
, gamma-glutamyl transpeptidase, myoglobin, as well as urea nitrogen, uric acid, creatinine, calcium, and phosphorus were examined before and for 6 days after exercise. Creatine kinase increased dramatically (peak value ranged from 6740 to 24,200 U/L) and aspartate aminotransferase, lactate dehydrogenase, alanine aminotransferase, and myoglobin followed the same time course as
creatine kinase
, but their peak values were lower. These proteins did not increase significantly until 48 hours after exercise and reached peak values 3 to 5 days after exercise. Alkaline phosphatase, gamma-glutamyl transpeptidase, uric acid, urea nitrogen, creatinine, calcium, and phosphorus showed no change. There is either a delay in muscle protein release by damaged muscle fibers, or the proteins are unable to leave the interstitial area for the 24 to 48 hour period after exercise. Because of the long delay, care should be taken when blood protein levels are interpreted in persons who have exercised strenuously (even if only for a short period of intense effort) several days before any diagnostic tests are performed.

To estimate the information content of different methods of diagnosing biliary diseases, 103 children aged 5 to 15 years, including 25 practically healthy children as control, were examined. Abdominal organs, particularly the liver and gallbladder were examined by echography. The duodenal contents was explored by microscopy of the sediment and biochemistry which included analysis in the vesicular and liver portions of the concentrations of phospholipids, activity of phospholipases A and C as well as measurements of
creatine kinase
, lactate dehydrogenase,
alkaline phosphatase
, C-reactive protein and of the diphenylamine index. Ultrasonography was found to be highly informative. The characteristic echographic alterations seen in dyskinesia of the biliary tract, dyscholias and chronic cholecystitis were defined. For differential diagnosis of dyscholia and chronic cholecystitis it is necessary to carry out biochemistry of the duodenal contents including measurements of phospholipids, activity of phospholipases A and C,
alkaline phosphatase
,
creatine kinase
, lactate dehydrogenase, C-reactive protein and the diphenylamine index. Based on the similarity of the alterations seen in children with different cholepathies, a concept is advanced of the mechanisms by which functional and organic diseases of the gallbladder are formed.

Ten intact male Holstein calves averaging 75 kg of BW and 59 d of age were used to study the effects of daily injections of 0 (control) or 10 mg of sometribove (recombinant methionyl bST) for 6 wk on performance, health, carcass composition, N metabolism, chemical blood characteristics, and hormone profiles. Average daily gain, feed intake, feed:gain ratio, and height at withers, hip, and hock were not influenced by bST. Carcasses from bST-treated calves contained 5% more protein and 36% less lipid than controls. Circulating concentrations of Ca, P, glucose, urea N,
alkaline phosphatase
,
creatine phosphokinase
, insulin-like growth factor-I, and insulin were not affected by bST. Packed cell volume was decreased about 7% (29.9 vs. 32.4%) in the bST calves. Hormone injection did not adversely affect health of the calves as measured by body temperature and by pulse and respiration rates. The most profound effects of sometribove were a reduction in carcass lipid and an increase in body proteins. These effects may be of some practical importance when leanness of carcass is desirable.

Two trials were conducted to determine the effect of monensin in broiler litter on sheep receiving the broiler litter in their diets. Broiler litter from chickens fed monensin as a coccidiostat, and from chickens receiving no coccidiostat, was included at a level of 30% in 2 sheep diets. In a further 2 treatments, monensin (15 mg kg-1) was added to each of the 2 diets to give a 2x2 factorial experimental design. In the first trial, copper (20 mg kg-1 feed) was added to the diets. These lambs were fed individually at a slightly restricted level of intake. No differences between treatments were observed in feed intake, average daily gain or efficiency of feed utilisation or in the concentrations of zinc, iron and manganese in the liver, glutathione peroxidase in erythrocytes and
creatine kinase
concentrations in the plasma. Hepatic copper content and copper retention in the livers of the sheep receiving the added monensin were significantly higher (P less than 0.05 and less than 0.01 respectively) than in those not receiving added monensin. The aspartate transaminase and
alkaline phosphatase
concentrations in the plasma of these sheep were also higher (P less than 0.05) than in those not consuming added monensin. In the second trial, the lambs were group-fed according to treatment and received the diets on an ad lib basis. The mean intakes of the groups receiving the diets with the added monensin, were lower than the intakes by the other groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Plasma alanine aminotransferase, aspartate aminotransferase,
creatine kinase
and
alkaline phosphatase
activities were studied in clinically healthy Danish landrace and dwarf kids in seven herds from birth to 12 months of age. The purpose was to evaluate the influence of age, breed and herd on reference values. The mean enzyme levels +/- standard deviation (s) in neonatal dwarf kids were 0.09 +/- 0.04, 1.23 +/- 0.24, 2.79 +/- 1.50 and 18.3 +/- 11.0 mu kat/l respectively. The respective values in landrace kids were 0.13 +/- 0.06, 1.06 +/- 0.22, 2.44 +/- 1.60 and 37.6 +/- 23.6 mu kat/l. In 8-12 months old dwarf kids they were 0.30 +/- 0.11, 1.49 +/- 0.13, 3.28 +/- 0.44 and 11.1 +/- 2.4 mu kat/l respectively and 0.23 +/- 0.05, 1.12 +/- 0.34, 3.68 +/- 1.63 and 14.1 +/- 8.40 mu kat/l respectively in landrace kids of the same age. The 5th to 95th percentile intervals of the enzyme activities were within mean +/- 2s for most age groups in both breeds except
alkaline phosphatase
. The means and medians were close to each other for the values of alanine aminotransferase, aspartate aminotransferase and
creatine kinase
but not for
alkaline phosphatase
. Alanine aminotransferase, aspartate aminotransferase and
creatine kinase
levels were low at birth and increased with age, whereas for
alkaline phosphatase
it was vice versa. Significant differences were observed in mean enzyme activities between kids of different ages (within breeds), breeds (in same age kids) and herds (within same breed and age kids). Sex variations (within the breeds) were not observed. It was concluded that plasma enzyme activities are dependent on age, breed and environment.
