Cultured osteoblastic cells were studied to determine the effects of laser irradiation on their rates of proliferation, differentiation, and calcification. A continuous wave
Helium
-Neon laser with a wavelength of 632.8 nm was used for this study. Clonal osteoblastic cells (MC3T3-E1) were exposed to laser beam at various energy densities. The cell growth rate and DNA synthesis were increased by laser irradiation only in the growing phase of culture. During long-term culture, 45Ca accumulation was enhanced by laser irradiation at 1.0 J/cm2, with four sessions of irradiation resulting in a 46% increase over controls. In contrast, no significant increase in
alkaline phosphatase
activity was produced by laser irradiation. Electron microscopy revealed a tendency of enlargement of the Golgi apparatus in the laser-treated cells. These results suggest that laser irradiation photoactivates osteoblastic cells, accelerates osteoblastic cell growth and calcification in vitro, and therefore, may promote bone regeneration.

The examination of 68 patients with biliary dysfunction determined hypokinesia of the gallbladder in 40, sphincter of Oddi spasm in 15 and combination of the two conditions in 13 patients. Blood biochemical indices showed no differences in patients with biliary dyskinesia compared to normal subjects except for
alkaline phosphatase
levels elevated in 35.3% of patients. This suggests the development of biliary hypertension and cholestasis. All the patients demonstrated disturbed colloid stability of the bile, in those with combined dyskinesia it became lithogenic.
Helium
-neon and semiconductor laser radiation of biologically active points and the hepatic region, respectively, improved the patients' performance status. The pain and dyspepsia discontinued. The function of the gallbladder and sphincter of Oddi recovered. Positive changes occurred in the blood and bile biochemistry. Laser therapy promoted bilirubin and bile cholesterol decrease. Cholic acid concentration grew, lithogenic characteristics of the bile returned to normal. It is inferred that laser therapy of biliary dyskinesia proved effective.

This paper reports on the use of
alkaline phosphatase
cytochemistry and combined conventional and confocal reflection and fluorescence scanning light microscopic modes in the study of human marrow stroma. It was found that the end product of the enzyme reaction using Napthol AS phosphate as substrate and Fast Blue BB as coupler reflected the 633 nm (red) light from a
Helium
-Neon laser. Serial optical sections suitable for 3-D reconstruction and selectively depicting the marrow reticulum cells could be obtained from thick glycol methacrylate sections reacted for Alkaline phosphatase. Furthermore, the yellow background of uncoupled diazonium salt over cytochemically unreactive structures in the same specimens and fields was used for imaging haemopoietic cell mass by operating the microscope at 488 nm (argon ion laser, blue-green). These methods may offer advantages in the investigation of the bone marrow stroma and its interplay with haemopoiesis and osteogenesis in normal and disease conditions.
