Calcium and vitamin D metabolism were evaluated in 5 adult epileptic patients before and during treatment with phenytoin. Significant decreases occurred in serum concentrations of calcium, albumin, and 25-hydroxy-cholecalciferol. The decreases in serum calcium paralleled those in serum albumin. Significant increases occurred in serum alkaline phosphatase and 1 alpha, 25-dihydroxycholecalciferol, in urinary hydroxyproline, and in the fractional gastrointestinal absorption of calcium. Urinary cyclic adenosine monophosphate and serum parathyroid hormone did not change. The results suggest that the bone disease resulting from phenytoin therapy may be associated with a deficiency of 25-hydroxycholecalciferol and not of 1 alpha, 25-dihydroxycholecalciferol, and that reduced gastrointestinal absorption of calcium or changes in parathyroid function may not be necessary for the development of bone disease.

The immobilization of purified influenza virus, rubella virus, crude nuclear cytomegalovirus antigen and of mycoplasma on polystyrene tubes was studied using radio-iodinated preparations. The antigen activities on tube surfaces were determined using sequentially specific human antibodies and alkaline phosphatase-conjugated anti-human IgG in an enzyme-immunoassay (EIA) reaction. In addition, immobilization of radio-iodinated human IgG, IgM, and IgA, serving as model proteins, was studied using the respective anti-immunoglobulin conjugates in EIA directly. Pretreatment of the surface with albumin and glutaraldehyde inhibited the adsorption and antigenicity of IgG. Increase of temperature and thus of speed of adsorption did not affect the fraction of antigen eluted during the test procedure. Only with IgG and IgA was it necessary to saturate the polystyrene surface in order to achieve maximal reactivity in EIA. With other antigens, maximal reactivity in EIA was obtained with amounts of protein much lower than the maximal amount that could be adsorbed per tube. IgM was found to have an exceptionally high affinity to polystyrene.

Serum gamma glutamyl transpeptidase (GGTP), isocitrate dehydrogenase (ICD), ornithine carbamoyl transferase (OCT), alanine aminotransferase (AlT), aspartate aminotransferase (AsT), and alkaline phosphatase (ALP) activities were assayed in 67 alcoholics and 40 drug dependent patients. Bilirubin, total protein, albumin, and globulin were also measured. GGTP elevation was observed in 48% of alcoholics and in 50% of drug dependents. The incidences of elevated levels of other enzymes were: ICD 39 and 38-7%; OCT 23-7 and 36-1%; AlT 30 and 33%; AsT 24-2 and 21-7%; ALP 10-4 and 5% respectively. Measurement of GGTP is thus more useful as a screening test for involvement of the liver in alcoholics and drug dependent patients than that of the other enzymes.

Changes in plasma sodium, potassium, chloride, total carbon dioxide, urea, creatinine, glucose, total bilirubin, iron, total protein, albumin, alkaline phosphatase (AP), aspartate amino transferase (AST), calcium, inorganic phosphorus, cholesterol and triglycerides were studied in 45 Thoroughbred foals 15 min to 28 days after birth. The results were analysed in 3 groups; Group 1 (0--12 h), Group 2 (12--36 h), Group 3 (1--4 weeks). When Group 2 was compared to Group 1, there were significant reductions of sodium, creatinine, iron and calcium and elevations of total protein and bilirubin. When Group 3 was compared to Group 1 there were significant reductions of sodium, chloride, urea, creatinine, bilirubin, iron and AP. Significant elevations occurred in glucose, total protein, AST, inorganic phosphorus and triglycerides.

A safety evaluation of Aspergillus fumigatus I21, grown in a cassava carbohydrate and salts medium, was undertaken. Male weanling rats were fed the fungus at 20, 30 and 40% of the diet for 90 days. A control group was given soybean oil meal as the sole source of protein. Weekly determinations of the body weights and feed consumptions were made. A few days prior to termination of the feeding study, a kidney function test was undertaken on the rats. At the end of the feeding period hematology, blood biochemistry, urine analyses and histopathology studies of various tissues were carried out, and organs were weighed. Rats fed A. fumigatus I21 gained less weight than the controls, but kidney weights were increased. Increases in serum alkaline phosphatase and glutamic-oxaloacetic transaminase were not related to dose level. The blood urea nitrogen was increased for the rats fed 40% of the fungus. Rats fed 30 and 40% of the fungus I21 showed a significant drop in albumin. Deficiency in methionine or other essential amino acids through a limited feed consumption may have caused a decrease in albumin synthesis. Rats fed the highest level of the fungus showed increases in neutrophils and monocytes concomitant with decreases in lymphocytes and eosinophils which may be a response to stress. The urine analyses did not reveal any significant differences. The test rats were capable of concentrating urine adequately when deprived of water for 24 hours. No significant differences between the control and experimental groups were found by histopathological examinations.

The healing of excisional wounds in rats, which were treated with zinc tape, gauze sponge or porcine skin, was studied. Wound closure was completed earlier in zinc-tape-treated wounds than in wounds treated with a gauze sponge or porcine skin. Wound contraction was more pronounced in gauze- and porcine skin-treated wounds than in zinc-tape-treated wounds. More foreign body giant cells were seen 14 days after operation in gauze-treated contra zinc tape-treated wounds, whereas hydroxyproline concentration was higher in tape-treated wounds. Histochemically the acid phosphatase activity was pronounced in macrophages, foreign body giant cells and fibroblasts. The alkaline phosphatase activity was pronounced in granulocytes and fibroblasts. Quantitatively a reduction in alkaline phosphatase activity was seen in gauze-treated wounds from 7 to 14 days. A decrease was seen in albumin concentration in gauze- as well as zinc-tape-treated animals which was most pronounced after 7 days in the gauze-treated animals.

Sperm transport in the female reproductive tract involves 3 stages: rapid short sperm transport; colonization of reservoirs; and slow prolonged release. Sperm transport through the cervix is affected by contractile activity of vagina and cervix, properties of cervical mucus, directional motility of sperm and possibly female orgasm, in addition to other endocrine, hereditary, immunological and psychological factors. Aided by myometrial contraction, spermatozoa may reach the site of fertilization within 2 to 10 minutes. During transport in the female tract, the sperm are separated from the seminal plasma and resuspended in endometrial and oviductal fluids, reducing the survival time of sperm. In artificial insemination, the physicochemical and immunological factors in the vagina and cervix affect sperm survival and transport into the uterus and oviduct. Spermatozoa are immobilized by vaginal secretions within 1 to 2 hours of insemination, suggesting the importance of rapid transport of sperm in the fertilization process. During ovulation, the cervical mucus provides an environment favorable to sperm survival; the biochemical changes which occur include decrease in albumin, alkaline phosphatase, peptidase, antitrypsin, esterase and sialic acid, as well as an increase in mucins and NaCl. The main cervical mucus components which are favorable to sperm survival have not been identified, although hexosamines and carbohydrate forms are suspected of contributing to sperm survival in the cervix.

In 4 out of 202 consecutive electrophoretic patterns for serum alkaline phosphatase isoenzymes, an extra band was observed in the albumin zone. Subsequent heat inactivation tests and comparison studies with an albumin-based bilirubin standard indicated that the band was due to staining of an albumin-bilirubin complex. The band should not be mistaken for an isoenzyme of alkaline phosphatase.

Normal values for 13 chemical constituents of plasma were estimated from results for 837 presumably healthy children. Ninety microliters of specimen was analyzed for lactate dehydrogenase, aspartate aminotransferase, alkaline phosphatase, inorganic phosphorus, total calcium, total cholesterol, total proteins, albumin, uric acid, urea nitrogen, alanine aminotransferase, total bilirubin, and glucose. We used two Abbott ABA-100 Bichromatic Analyzers interfaced directly to the ABA Data Management System. For each test age- and sex-related variations were assessed and normal values were estimated for six different age groups.

In order to verify the influence of sampling time on blood constituents, populations of supposedly healthy subjects were grouped according to age, sex, deviation from their ideal weight, state of fasting or nonfasting, and time of sampling. Each fasting subject in one group underwent two samplings during the course of a morning: the first at 08.00 and the second between 09.00 and 12.00. In the second group, the first was taken at 13.00, and the second between 14.00 and 16.00. Subjects in the second group had eaten a standard meal of 700 calories at 12.00. Differences between the paired samples from a given individual are discussed with respect to the time of sampling for plasma urea, creatinine, proteins, albumin, calcium, sodium, potassium, cholesterol, uric acid, chloride ions, phosphate, bilirubin, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, creatine phosphokinase, alkaline phosphatase, hemoglobin and erythrocyte and leukocyte counts. Variations due to the time of sampling were large for phosphorus, bilirubin, and leukocyte count.
