50 guinea pigs were allergized three times in 3 days intervals by subcutaneous injection of 25% solution of egg-white in physiological saline in a dose of 0-1 ml/100 g of body weight. On the 21 day after the last injection the animals were exposed to aerosol of antigen of egg-white. 11 animals died in acute anaphylactic shock. The control group consisted of 12 guinea pigs which received subcutaneously a solution of physiological sodium chloride of the same dosis--0-1 ml/100 g of body weight and were also exposed to the allergen, together with the experimental group. The removed parathyroids together with the thyroid gland were studied with histologic and cytoenzymatic methods. The activity of alkaline and acid phosphomonoesterase, nonspecific AS-naphtol acetate esterase, succinic (SDH), lactic (LDH) and D-L-alfaglicerophosphate dehydrogenase (alpha-GPD) were tested. No morphological changes in the parathyroids of guinea pigs in anaphylactic shock were found. Instead a decrease of the enzymatic activity of dehydrogenases was found, what might be connected with the decrease of metabolic activity of the cells. The decrease of alkaline phosphatase activity in the endothelial cells of the capillaries was another finding. It is likely that in an acute anaphylactic shock in guinea pigs only functional changes develop which have not counterparts in histology visible under the light microscope.

In this study we investigated the function of the obstructed small bowel of the rat, the behaviour of the mucosal enzymes, the metabolic changes of the small bowel wall and the morphology of the mucosa. We found a decrease of passive transport of 3H-Antipyrine which was equal after 24 and 48 hrs. The active transport of 14C-Glucose was found to be progressively inhibited after occlusion. The metabolic enzymes SDH, G-6-PDH, and GOT remained unchanged, LDH was increased after 48 hrs, which can be explained by enzyme induction. The lactate-pyruvate ratio in the tissue of the obstructed bowel was 3 times as high as in the controls. The brush-border enzymes maltase and especially the alkaline phosphatase are decreased with progressive obstruction, which probably is caused by diffusion into the lumen. By electron-microscopy there are no changes in the brush-border membrane but a swelling of mitochondria which is caused by hypoxia.

In the group of 107 patients poisoned by carbon monoxide (18 patients), ethanol (10), barbiturates (18), glutehimide (10), tranquilizers (19), organic solvents (10),salicylates (3), organochlorines (8), and sulfonamides (5)--the activities of 8 serum enzymes were determined for 6 consecutive days of treatment, the enzymes being as follows: aminotransferases, cholinesterase, alkaline phosphatase, lactate, alpha-hydroxybutyrate, glutamate, and sorbitol dehydrogenase. The antipyrine half-life was also assayed. It has been shown that the poisonings by particular groups of poisons do not bring about characteristic changes in the activity of enzymes that might be of any diagnostic value. The intensity of changes was connected withe depth and duration of toxic coma. Most frequently an increase ensued in the activity of AspAt and AlAt in the third 24-hrs period, and an increase in the activity of SDH in the first 24-hrs period. In the group under examination there were 26 drug abusers in whom a shortening of the antipyrine half-life was discovered. They were less responsive to toxic doses of drugs, and the enzymatic changes in them were less distinct. No changes in the activity of tested enzymes, which are characteristic of toxicomania, were found.

Female rats were given intragastrically, by means of a probe, Vibramycine in suspension, in a dose 4 mg/kg body weight, in different stages of pregnancy. On kidney sections there were performed: H+E staining, PAS reaction, histochemical determination of enzymes activity: alkaline phosphatase, ATP-ase, SDH and LDH. In non-pregnant females receiving the drug there was observed an intensification of alkaline phosphatase activity in brush border of main tubules of the kidney and a slight broadening of the light of the tubules. In pregnant females receiving the drug, most clearly in the second half of pregnancy a slight intensification of the activity of the examined enzymes was observed.

Leukocytic dehydrogenases (succinate and alpha-glucose phosphate dehydrogenases) and acid and alkaline phosphatase hydrolases activities were cytochemically assayed in 20 healthy women with normal menstrual cycles (controls) and 34 ones with habitual abortions. Ovulation in health was associated with a significant elevation of SDH, alpha-GPDH, and alkaline phosphatase activities. The second peak of enzymic activation, less marked, was recorded on days 20-24 of the cycle. Acid phosphatase activity was reduced by the second phase of the cycle. In women with habitual abortions the activities of redox enzymes (SDH and alpha-GPDH) reduced as against the controls (p less than 0.05, less than 0.01, less than 0.001), and no ovulatory peak of enzymic activation was detectable. The curves of enzymic activities were in good correlation with rectal temperature curves of the examinees. Elevated enzymic activities during ovulation and in the second phase of a normal cycle evidenced a sufficiently high energy supply of these processes. The detected reduction of leukocytic enzymic activities in women with habitual abortions may be used for the early diagnosis of an unfavorable pregnancy course and, possibly, for the prediction of placental insufficiency.

Activities of the following enzymes were assessed in cryostat sections of human embryonic and fetal placentae aged 7 to 22 weeks of the intrauterine life using the standard methods recommended by Lojda et al. (1978): alkaline phosphatase (AIP), and acid phosphatase (AcP), non-specific esterase (ANE), ATP-cleaving enzymes (ATP-ase), beta-glucuronidase, thiamine pyrophosphatase, dipeptidylaminopeptidase IV (DPP IV), aminopeptidase A and M (APA, APM), gamma-glutamyltransferase (GGT), glycero-3-phosphate dehydrogenase and succinate dehydrogenase (alpha-GPDH, SDH). Since week 7 high activity of AIP has been proved in the apical zone of the plasmodiotrophoblast. At the same time the DPP IV activity appeared in the plasmodiotrophoblast, in the stroma of villi, and, latter on, in vascular endothelium. In the fetal placenta the APA activity was pronounced both in the cytotrophoblast and the stroma of villi. The activities of AcP and ANE were relatively weak. In the course of development the activities of most enzymes were gradually increasing.

Intraperitoneal injection of 5 mg or 10 mg per kg body weight of technical grade lindane twice weekly in Charles-Foster female rats induced marked disturbance of the estrous cycle, prolonging the proestrus phase considerably (5-7 times more compared to the normal) and thereby delaying ovulation. Exfoliated cells at proestrus are marked by high concentrations of glycogen, SDH and alkaline phosphatase while estrus is characterized by increased activity of acid phosphatase and mucopolysaccharides. Corresponding changes in the polysaccharides concentrations and enzyme activities of the exfoliated cells seen in the lindane treated rats were well correlated with disturbances of the cycle. The data suggest that lindane exerts an estrogen-like effect by inducing a temporary negative feedback on pituitary gonadotropin secretion.

An inguinal sweat gland carcinoma is described including the unusual occurrence of alpha hCG. The alpha hCG was demonstrated in the tumor presenting choriocarcinomatous differentiation and undifferentiated polygonal cells in ordinary tumor nests. beta hCG and SP-1 were weakly and sporadically demonstrated in a small area of the tumor presenting choriocarcinomatous differentiation. On the other hand, hPL and histochemical reaction of placental type alkaline phosphatase were not observed in the choriocarcinomatous and undifferentiated tumor cells. CEA was observed in both ordinary sweat gland carcinoma nests and choriocarcinomatous differentiated regions. In the ordinary tumor nests, enzyme histochemical reactions of phosphorylase and SDH were positive. And S 100 protein was sporadically demonstrated. In spite of chemotherapy and radiotherapy, the patient died after a short duration of the disease.

1. The incidence of microscopically detectable degenerative characteristics in 5 skeletal muscles (m. pectoralis thoracicus, m. supracoracoideus, m. biceps femoris, m. semitendinosus, m. femorotibialis medius) of turkeys was investigated. 2. Samples were obtained from 30 Large White turkey males 14, 16 and 18 weeks old. Hyaline degeneration, infiltration of mononuclear cells and necrotic fibres were observed. 3. Individual fibres varied greatly in size and muscle fibre nuclei were often shrunken and pyknotic. 4. Weak and/or uniform reaction for Ca++-ATPase and SDH in all types of muscle fibres and loss of alkaline phosphatase activity in cell membranes were noted. A positive reaction for acid phosphatase occurred in regions of perivascular infiltration and in necrotic muscle fibres. The majority of muscle fibres possessed high activity for phosphorylase a and b. 5. Based on the use of fluorescein alpha-bungarotoxin conjugate, motor end-plates appeared to be morphologically intact. Direct immunofluorescence with anti-chicken IgG showed positive reaction in muscle fibres undergoing necrosis and in the involved connective tissue. 6. Degenerative changes varied with age and were most marked in the oldest birds. 7. Because gross degenerative symptoms were absent from both the birds and the meat from them, the condition appears to be either different from or a precursor to the degenerative myopathy characterised by other authors.

We studied the testis of Wistar rats weighing 280-300 gms. following the administration of a single, acute intracardiac dose of methionine-enkephalin (100 microliters of 50% met-enkephalin solution), or a chronic intramuscular dose (50 microliters of 40% met-enkephalin solution). Rats were sacrificed at 15, 30 and 60 minutes following acute injection. Those on chronic treatment were injected once daily for 10 or 20 days. For the study, we utilized 105 male Wistar rats; 30 comprised the control group, and 75 comprised the study group. The following staining methods were used: 1) succinate dehydrogenase, 2) lactate dehydrogenase, 3) ATPase, 4) acid phosphatase, 5) alkaline phosphatase. We observed marked histoenzymological changes in the rat testis. Particularly noteworthy was a marked change in the energy pathways consisting of a decreased activity of aerobic pathways (decreased SDH activity), increased anaerobic activity (increased LDH activity), and consequently, decreased cellular energy stores (decreased ATPase activity). Similarly, changes were observed in other nonspecific enzymes that led to a fall in acid phosphatase activity and a rise in alkaline phosphatase activity.
