Sequential therapy to regulate menstruation was administered to 30 women to determine if liver functions are disturbed by contraceptive use. 15 women aged 16-22, used a sequential preparation consisting of ethinyl estradiol and norethisterone acetate. These patients showed significant decreases in aminotransferase GPT and
alkaline phosphatase
in the last (gestagen) stage of treatment, and slight increases in
alpha-amylase
and beta-globulin throughout the therapy. In the 15 women aged 17-34 who used ethinyl estradiol and chlormadinone acetate, the ZST showed a significant increase during the therapy. No significant changes in the liver function could be shown by these tests. These preparations are recommended for treatment of disturbances of menstrual cycles; tests of liver functions during treatment are unnecessary. The length of the estrogen and estrogen-gestagen phases of the treatment should be equal to prevent hormonal imbalance.

During the first cycle of treatment the influence of four forms of sequence therapy with mestranol and ethinyl estradiol as estrogen and the two gestagens chlormadinone acetate and norethisteron acetate in women at fertile age was examined for the aminotransferases (GOT and GPT), the activity of
alkaline phosphatase
and
alpha-amylase
, for the cholesterol, total bilirubin and total protein content of the serum and for the half-life period of indocyanine green. Under the application of mestranol/chlormadinon acetate an estrogen-induced increase of the activity of the alanine aminotransferase (GPT) from 6.38 units/l to 12.14 units/l may be established, after addition of the gestagen chlormadinon acetate a decrease to 5.34 units/l was to be established. Under the sequence therapy with mestranol/norethisteron acetate only an increase of the alpha1-globulin proportion was to be ascertained. All the other changes of the tested parameters including the half-life period of indocyanine green were not essential.

The effects of two sequential therapies using mestranol as an estrogen and chlormadinone acetate and norethisterone acetate, respectively, as a gestagen on the aminotransferase (GOT and GPT),
alkaline phosphatase
and
alpha-amylase
activities, the cholesterol, total bilirubin and total protein contents and the indocyanine green (Ujoviridin, VEB Chemiekombinat Bitterfeld) half-life were studied in two groups, each comprising 12 women of fertile age. The serum protein was separated by paper electrophoresis and both the thymol turbidity and zinc sulphate tests were performed during the first treatment cycle. An estrogen-induced rise in alanine aminotransferase (GPT) activity from 6.31 U/liter to 12.14 U/liter was observed during application of the mestranol/chlormadinone acetate therapy. The value dropped to 5.34 U/liter when chlormadinone acetate was administered as a gestagen in addition to the estrogen. No significant changes were noted in the other biochemical parameters. Sequential therapy involving application of mestranol and norethisterone acetate produced only statistically secured rises in the alpha 1-globulin content and the GPT activity. All other changes in the different parameters remained within the normal range and were statistically insignificant.

The hitherto existing results of determinations of enzymatic activities in stool are presented in this review. The chymotrypsin activity is diminished in advanced exocrine pancreatic insufficiency. The faecal
alpha-amylase
activity has up to now no significance in the diagnosis of pancreatic diseases. Up to five amylolytic enzyme activities are detectable. The
alkaline phosphatase
is mostly of intestinal origin. Up to 4 enzyme bands can be exhibited with the disc electrophoresis in polyacrylamide gel. Lysozyme and N-Acetyl-beta-D-glycosaminidase can also be detected in stool.

A quantitative study has been made on the enzymic, chemical and ultrastructural changes that occur in the parotid glands of rabbits as a result of Isoprenaline-induced secretion. Emphasis has been placed on correlating changes in organelle and membrane content which are evident 2 hr after Isoprenaline administration and which have been measured stereologically with the levels of appropriate enzymic or chemical markers, taking into account the contribution made by both the acinar and duct tissue. Lower protein,
alpha-amylase
and beta-glycerophosphatase levels correlated with reductions in zymogen granule and lysosome volume with whilst plasmalemmal and Golgi membrane areas and their marker enzyme concentrations remained unchanged. However, declines in
alkaline phosphatase
and succinate dehydrogenase activity (illustrated histochemically), and p-nitrophenyl phosphatase activity at pH 4-5 in the presence of tartrate occurred without any detectable decrease in membrane area. Conversely, an increase in rough endoplasmic reticulum area was measured stereologically but no increases in chemical markers were detected. The extent of correlation of the data is discussed in the context of the mechanism of secretion and the action of Isoprenaline.

The content of proteins and nucleic acids and activity of acid and alkaline proteases, RNases, phosphatases and

were studied in the gum and alveolar bone or rats at the age of 1, 3, 6, 12, 18 and 24 months. It is found that a degree of periodontal atrophy strongly and directly correlates with the age of rats. The concentration of DNA and RNA in alveolar bone and of RNA in gum decreases with the age. The hydroxyproline content of periodontal tissues continuously increases till the age of 18th month and then considerably decreases. The activity of trypsin-like proteases, cathepsins and alkaline RNase in periodontal tissues increases reaching the maximum at the age of 6-18 months, and the activity of
alkaline phosphatase
decreases in the process of aging.

Investigations for the proof of isoenzymes of the
alkaline phosphatase
and the
alpha-amylase
in stool were carried out. The measurements of the faecal enzyme activities were performed in 96 patients with intestinal diseases. The disc electrophoresis in the polyacrylamide gel served for the separation of isoenzymes. The separated enzyme bands of the extracts of stool were compared with those ones of serum duodenal juice and extracts of the mucous membrane of the duodenum, the jujunum, the ileum as well as of the colon. Hereby up to 4 very much anodically wandering bands were present, which might be subunits or fission products of the
alkaline phosphatase
. In the
alpha-amylase
up to 5 bands could be proved, which were compared with those ones of the serum, the duodenal juice and the urine. A coordination of these isoenzymes concerning their origin is not possible. The faecal activities of enzymes correlate with the weights of stool. In exocrine pancreatic insufficiency and ulcerous colitis increased faecal activities of enzymes are found, the genesis of which is discussed.

The intermethod analytical variability of 59 commercially available control sera was compared with that of patient sera. For this purpose, patient and control sera were assayed with respect to ten constituents (albumin,

, cholesterol, glucose, iron, lactate dehydrogenase, creatinine, protein, and urea), each with two analytical methods. Only 6 of the 59 control materials showed an intermethod analytical variability comparable to that of the patient sera for all of the determinations. The use of patient sera for the calibration of routine analytical methods is recommended.

The method for the determination of enzymic activity in turbid, lipaemic sera, which involves clearing by polyanion precipitation with heparin and magnesium chloride, was critically reviewed. In the diagnosis of diseases of the liver and pancreas, which are frequently associated with hyperlipoproteinaemia, only residual enzyme activities are measured in the cleared serum after polyanion treatment. In the measurement of glutamate dehydrogenase and in the Phadebas test for
alpha-amylase
, the enzymes are inactivated by treatment with heparin and magnesium chloride. On the other hand, as a result of polyanion precipitation gamma-glutamyl transferase is transferred, together with lipoproteins and chylomicrons, to the lipid-rich supernatant. , cholesterol, total bilirubin and proteins of the serum, TTT, and indocyanine green were measured. A little significant decrease of the activity of alaninamino transferase (GPT) was to be stated. Whereas at the end of the 6th cycle the TTT as well as the contents of total proteins and albumin, showed a little significant decrease and the contents of alpha-2-globulin, beta-globulin as well as cholesterol were statistically shown to grow. The indocyanine green elimation was longer at the end of the 6th cycle without any pathological worth from the clinical point of view being proved.
