The presence of proliferating cells has been sought in the synovium of rheumatoid arthritic (RA) and osteoarthritic (OA) joints using the monoclonal antibody Ki67, which marks a nuclear antigen present in all stages of the cell cycle apart from Go. Synovia were studied from 21 RA and 14 OA cases using the indirect immunoperoxidase technique. Double-staining was performed on 18 RA and 17 OA synovia with the simultaneous labelling of
lysozyme
(muramidase) by the immuno-
alkaline phosphatase
method and with Ki67 by the indirect immunoperoxidase method. Most of the RA and OA synovia showed an absence of Ki67-positive cells in the intimal cell layer. Three RA and four OA synovia showed no more than ten proliferating cells in the whole of the intimal layer examined. Similar results were obtained when double-labelling was performed. Eight RA and six OA synovia showed the presence of occasional Ki67-positive cells in the intimal layer. The total number of intimal cells was measured for each histological section, and the proliferation index calculated as the percentage of total cells in the intimal layer showing Ki67-positive staining. This varied between 0.03% and 0.0033% (between 1:2800 and 1:30,000 cells). In contrast, there were plentiful Ki67-positive cells present in the lymphocytic infiltrate and around blood vessels in the RA synovia and in the synovial infiltrate, where present, in the OA cases.(ABSTRACT TRUNCATED AT 250 WORDS)

The present study demonstrates that normal human fibroblasts (WI-38) exert a profound influence on the growth and differentiation of HSGc-C5, a clonal neoplastic epithelial cell line of human salivary gland origin. Coculture of HSGc-C5 with WI-38 resulted in a slowing of growth and an increase in glycosaminoglycan synthesis by an indirect effect involving a diffusible factor(s). Conditioned medium (CM) from WI-38 grown in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum affected HSGc-C5 as follows. The CM suppressed growth of monolayer cells; inhibited DNA synthesis; suppressed growth (decrease in size of colonies) in semisolid agar; stimulated glycosaminoglycan synthesis, induced expression of functional markers of the salivary gland, such as the secretory component, lactoferrin, and
lysozyme
; inhibited expression of
alkaline phosphatase
; and induced morphological alteration into elongated cells. These findings strongly suggest that WI-38 CM contains a factor(s) which inhibits growth and induces differentiation of HSGc-C5. The CM was also active on other human cancer cells as a growth inhibitor, but not on normal human fibroblasts. Partial purification and characterization of the factor(s) suggests that it may be a novel protein carrying both tumor inhibiting and differentiation inducing activities.

, NAG, acid protease and tubular cell counts in a dose-dependent manner, but did not increase gamma-glutamyl transpeptidase (gamma-GTP) and

excretions. The urinary isoenzyme profiles of LDH in rats treated with GM were different from those with HgCl2. The increase in acid protease excretion outlasted those in LDH and GOT in the high dose group. It was concluded that the severity of renal damage can be readily detected by periodic determinations of the following urinary parameters: tubular cell counts, LDH isoenzyme, acid protease,

and NAG, in addition to either LDH or GOT and one of the enzymes

A 15-year-old boy with lymphadenopathy and marked leukocytosis was evaluated. At 2 years old, he was diagnosed to have malignant testicular carcinoma for which radiotherapy and cytotoxic drugs were administered. On admission, the peripheral blood showed an increase of all stages of the granulocytic cell line and atypical monocytoid cells. Cervical lymph node biopsy showed infiltration by immature granulocytic cells. Eleven percent of the peripheral leukocytes was positive for combined staining by alpha-naphthyl butyrate esterase and naphthol-ASD-chloroacetate esterase activity. The leukocyte
alkaline phosphatase
score was low. Serum and urinary
lysozyme
levels were markedly elevated. Philadelphia chromosome was not detected. Although chronic myelomonocytic leukemia (CMML) has been described in elderly patients, the findings in this pediatric patient were most compatible with a diagnosis of CMML.

It was established in an experiment with 13 sheep of the local improved breed that the single s/c injection of levamisole at the rate of 7.5 mg per kg of body mass led to a rise of the phagocytic activity of neutrophil leukocytes. These cells also had higher cytochemical activity of the
alkaline phosphatase
, glucose-6-phosphatase, and lipids (sudanophilia). No changes in the total count of leukocytes were found. The cytochemical activity of lactate-dehydrogenase, succinate-dehydrogenase, and alpha-glycerophosphate-dehydrogenase was also found to rise. On the base of the marker capacity of the acid naphthylacetate esterase with regard to the T-lymphocyte system there set in on the 3rd day following levamisole injection a 4 to 5 rise in the T-lymphocyte count, particularly in the mature T mu-lymphocytes, to the detriment of the B-cells which dropped in number. On the 7th day all these changes receded, however, the percent of lymphocytes with azure granules in the cytoplasm and the average number of these granules per lymphocyte cell rose. In the entire 7-day period of investigation a lowering trend was shown by the amount of serum

which dropped several times as against the initial level.

A retrospective analysis of 30 patients with chronic myelomonocytic leukemia (CrMML) was performed to define the natural history of the disease and the risk of acute transformation. Our patients fulfilled the following criteria of diagnosis: blood monocytosis over 1 X 10(9)/l, blast cell percentage in bone marrow up to 30, and in peripheral blood less than 5. The most common presenting feature was anemia; seven patients had fever; three patients complained of purpura and bleeding. Anysopoikilocytosis and macrocytosis were frequent. Abnormal granulocyte morphology, defective granulation and abnormal leukocyte

were often observed. Blast cells in peripheral blood smears were found in 14 patients. Serum and urine
lysozyme
levels were increased in 82 per cent and 93 per cent, respectively. Dysplastic changes involving erythroid, granulocytic and megakaryocytic lineages were constant features in all cases. Agranulated blasts above 5 per cent of marrow nucleated cells were seen in 13 patients (43 per cent). Seven of the 20 patients showed non-specific chromosomal abnormalities at diagnosis. Median survival from diagnosis was 18 months (range, 3-112). Evolution into acute myeloid leukemia occurred in 11 patients. No difference in survival was found between patients who developed acute leukemia and patients who did not. A shorter survival has correlated to the following parameters: leukocytes greater than 10 X 10(9)/l, the presence of blasts in peripheral blood and agranulated blasts in the marrow above 5 per cent.

Intragastric administration to mice of solutions of crystalline T-2 toxin in a dose aqual to the LD50 or in doses from the 1/5 to 1/50 of the LD50 brought about a stable decrease in

In addition, acute and subacute T-2 mycotoxicoses were characterized by a marked lowering of hemoglobin concentration, the total red cell and leukocyte counts (doses 1/5-1/20 of the LD50) and lymphosytes (doses 1/5-1/50 of the LD50) in the peripheral blood toward the end of experiments. Variation of the hematological and biochemical characteristics after administering the toxin in low doses was detected despite the absence of the clinical symptoms of intoxication. These characteristics are the most sensitive to the toxic action of T-2 toxin.
