In 28 patients suffering from alcoholic well compensated hepatopathy, different laboratory parameters have been controlled before and after treatment with arginine thiazolidinecarboxylate, i.v. a the daily dose of 800 mg for 7-10 days. The compartmental analysis showed that the improvement in the BSF clearance is mainly due to a reduction in the "regurgitation fraction" and therefore to an improvement or restoration of the processes of conjugation, binding and transferring of the dye in the liver cells. A significant increase in platelets and an equally significant decrease in bilirubinemia, gamma GT, OCT, GOT and alkaline phosphatase were reported. Such variations might be at any rate due to the sole alcohol abstinence. The observed increase in transferrin, haptoglobin and albumin cannot be reported to the sole abstinence, but must be considered as consequence of the pharmacological influence, of arginine thiazolidinecarboxylate on the protidosynthetic activities of the liver.

24 hour hydroxyprolinuria was measured in 50 chronic alcoholics divided up into those with simple alcoholism and those complicated by cirrhosis. All the patients had a significant increase in hydroxyprolinuria. Without there being any difference between cirrhotics and alcoholics without cirrhosis. Comparison between hydroxyprolinuria and the tests usually used to follow the course of hepatic involvement in chronic alcoholism: IgA, transferrin, electrophoresis of serum proteins, alkaline phosphatase, show that there is no correlation between hydroxyprolinuria and the diagnostic or prognostic tests of an alcoholic liver among which the variable IgA is the most significant. On the other hand, hydroxyprolinuria has a linear correlation with the calciuria, which suggests that the increase in hydroxyprolinuria in chronic alcoholics is more related to changes in the collagen of bone tissue than with those in liver tissue.

Hepatocytes from adult rats were maintained in primary culture for up to 10-13 days on nylon meshes coated with a thin layer of rat tail collagen gel. Their ultrastructure closely resembled that of the liver parenchymal cell in vivo, but hepatocytes in late culture exhibited a pronounced buildup of microfilaments beneath their apical cell surface. Hepatocytes in early and late cultures secreted albumin, transferrin, and alpha1-acid glycoprotein into the medium; they exhibited a 7- to 10-fold induction of tyrosine aminotransferase activity by dexamethasone; and they expressed an alkaline phosphatase that was similar to that of normal rat liver with respect to its inhibition by the liver enzyme inhibitor L-homoarginine. In addition, the hepatocytes in culture demonstrated phenotypic changes characteristic of fetal liver parenchymal cells. These changes, which paralleled an increase in DNA synthesis, included the expression of and linear increase in the activity of the fetal liver cell enzyme gamma-glutamyl transpeptidase, an increased production of alpha1-fetoprotein, and a change in the substrate specificity of fructose-bisphosphate aldolase to that of the fetal liver isozyme.

Electron spin resonance spectroscopy has proved a useful and simple technique for the measurement of levels of caeruloplasmin and iron transferrin in whole blood from 50 patients with Hodgkin's disease. Those patients with clinically active disease show higher caeruloplasmin levels and lower iron transferrin levels than those with inactive disease. The results indicate that these tests are good indicators of the state of the disease and that serial measurement of these parameters may help in early prediction of clinical reactivity and in monitoring response to treatment. The combined information from iron transferrin and caeruloplasmin levels appears to be more predictive than that from the erythrocyte sedimentation rate and neutrophil alkaline phosphatase score.

Syncytiotrophoblast microvillous plasma membrane (StMPM) preparations were obtained from human full-term placentae by previously published methods of cold saline extraction and phase centrifugation. Purity of these preparations was assessed by electron microscopy, enzyme analysis and hydroxyproline content. IgG, albumin, alkaline phosphatase, transferrin, ferritin and alpha 2-macroglobulin were consistently detected in the aqueous soluble fraction from sodium deoxycholate-solubilised StMPM preparations by antigenic or electrophoretic analysis, beta 2-Microglobulin was not detected in these preparations. Up to 21 discrete protein bands could be demonstrated by SDS--PAGE, and their molecular weights determined. Many of these components need to be further identified, including a glycoprotein of molecular weight 36 500 which was particularly prominent. The soluble fraction from StMPM preparations gave a single strong precipitin reaction in immunodiffusion against wheat germ agglutinin, but not against other lectins studied.

Polymorphism for the number of the blood proteins (transferrin, alkaline phosphatase, haptoglobin, plasma esterase and erythrocyte esterase) have been exposed by means of starch gel electrophoresis of northern fur seal Callorhinus ursinus from three regions: Pelagic of Commander Islands (n = 164), Rockery of Robben Island (Sakhalin population, n = 308), winter todder region to the east of Honshu Island (n = 110). The reliable differences have been exposed between fur seals of the Commander and Sakhalin populations by the gene frequencies Tf and Es pl. Fur seals from winter todder region turned out to be closer to Commander population, that to the Robben one. However, originality of gene frequencies of locus Ap (alkaline phosphatase), revealed from the todder region animals, is difficult to explain by the mixture of the Commander and Robben population only. Perhaps, this originality is caused by the influence of Pribylov population. It is established that hemoglobin of northern fur seal consists of two components (Hb I + Hb II). Individual variability of hemoglobin results in the variation of the colour intensity of the "slow" component (Hb II).

In a previous paper we reported that serum, urine and fecal zinc levels fell markedly in women taking a combination oral contraceptive agent (+OCA) and in women with normal menstrual cycles (-OCA) when they consumed a low-zinc diet (less than 0.2 mg/day) for 35 days. We evaluated other biochemical and clinical data in order to determine if depletion of accessible body zinc and/or physiologic adjustment to conserve body zinc stores had occurred. Neither low zinc intake nor oral contraceptive use appeared to influence nitrogen balance or body weight. Use of contraceptive drugs appeared to influence the response of blood parameters to zinc depletion. Serum transferrin and cholesterol declined significantly in the -OCA group, whereas alkaline phosphatase and gamma-globulin changed significantly in both groups. Clinical problems developed in all the subjects with serum zinc levels below 50 microgram/dl during the study; three of the six with serum zinc levels above 50 microgram/dl also complained of clinical symptoms. The results suggest that zinc deficiency through depletion of accessible body zinc stores developed during the 35-day study.

The effect of 10 days of total fasting (energy deprivation) on blood polymorphonuclear granulocyte functions, leukocyte numbers, iron and transferrin levels was evaluated in 14 healthy, normal-weight males. Granulocytes from 7 of the subjects were tested in vitro. A statistically significant depression was noted in their bactericidal capacity against Staph. aureus. The 14 subjects showed a marked decrease in the stainable activity of granulocyte alkaline phosphatase and decreases were noted in plasma iron and serum transferrin levels. The iron saturation of serum transferrin was unchanged. Thus, impairment of granulocyte bactericidal functions may occur secondarily to short-term total energy deprivation, in the absence of iron deficiency.

The polymorphism observed among the enzymes involved in the respiratory metabolism (lactate dehydrogenase, malate dehydrogenase, phosphoglucomutase, phosphohexoseisomerase, glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase fructose 1-6 diphosphate dehydrogenase) is less important than that of the enzymes physiologically less essential, such as the various esterases, the alkaline phosphatase, the alcohol dehydrogenase, and of the non-enzymatic proteins (ovalbumin, ovoglobulins, ovomucoid, conalbumin, transferrin, etc.).

The alkaline phosphatase in homogenates of human liver was separated into two components by the addition of Triton X-100 to an agarose gel electrophoretic system. One of these components migrated at a rate identical to that of the original one and similar to alpha2-macroglobulin. The other component migrated more slowly, at a rate that resembled that of beta1-transferrin. Human serum samples regularly contained an identical fast-migrating fraction, whereas an identical slowly migrating fraction only appeared in serum obtained from patients with various diseases, especially from patients with malignant tumours, even though the liver did not contain tumour metastases. Slow isoenzyme was found in a few sera that had alkaline phosphatase activity within the normal range. Histochemical examinations of liver tissue from patients whose serum contained the slowly migrating isoenzyme showed a pronounced reaction of alkaline phosphatase in the bile canaliculi, and this isoenzyme seems to arise from the canaliculi. The fast-migrating isoenzyme might arise from the endothelial cells of the liver, to which the activity is usually confined in histochemical stainings.
